Aims: This work aimed to identify the molecular mechanism that allows yeast cells to survive at low pH environments such as those of bioethanol fermentation. Methods and Results: The industrial strain JP1 cells grown at pH 2 was evaluated by microarray analysis showing that most of the genes induced at low pH were part of the general stress response (GSR). Further, an acid‐tolerant yeast mutant was isolated by adaptive selection that was prone to grow at low pH in inorganic but weak organic acid. It showed higher viability under acid‐temperature synergistic treatment. However, it was deficient in some physiological aspects that are associated with defects in protein kinase A (PKA) pathway. Microarray analysis showed the induction of genes involved in inhibition of RNA and protein synthesis. Conclusions: The results point out that low pH activates GSR, mainly heat shock response, that is important for long‐term cell survival and suggest that a fine regulatory PKA‐dependent mechanism that might affect cell cycle in order to acquire tolerance to acid environment. Significance and Impact of the Study: These findings might guide the construction of a high‐fermentative stress‐tolerant industrial yeast strain that can be used in complex industrial fermentation processes.
Fuel ethanol fermentation process is a complex environment with an intensive succession of yeast strains. The population stability depends on the use of a well-adapted strain that can fit to a particular industrial plant. This stability helps to keep high level of ethanol yield and it is absolutely required when intending to use recombinant strains. Yeast strains have been previously isolated from different distilleries in Northeast Brazil and clustered in genetic strains by PCR-fingerprinting. In this report we present the isolation and selection of a novel Saccharomyces cerevisiae strain by its high dominance in the yeast population. The new strain, JP1 strain, presented practically the same fermentative capacity and stress tolerance like the most used commercial strains, with advantages of being highly adapted to different industrial units in Northeast Brazil that used sugar cane juice as substrate. Moreover, it presented higher transformation efficiency that pointed out its potential for genetic manipulations. The importance of this strain selection programme for ethanol production is discussed.
-The aim of this study was to isolate sporogenic bacteria from the honey of stingless bees Melipona sp., in dry forest, and to evaluate their antagonistic potential for medicinal employment purposes and animal production. The honey samples were collected in Serra Talhada-PE, where honey was taken from four different hives (in triplicate), totaling 12 samples. The samples were diluted and subjected to 80 ºC for 20 minutes to eliminate vegetative cells. The dilutions were plated onto nutrient agar and incubated at 30 ºC for 72 hours. Then the colony forming units (CFU) were quantified. The samples were also plated onto malt agar and Sabouraud agar, and incubated at 30 ºC for 14 days for the growth of yeast and molds. Total and fecal coliforms were quantified by the most probable number method (MPN). Seven isolates (I) of sporogenic bacteria (Bacillus) were obtained, however only four showed probiotic potential. Isolate I-5 showed the greatest probiotic potential and inhibited the growth of Escherichia coli, Klebsiella sp., Pseudomonas aeruginosa, Salmonella sp., and Staphylococcus aureus. The growth of the Sarcina sp. was not inhibited by any isolate. No yeast, molds or coliforms were found. The Melipona sp. honey is a source of spore-forming bacteria and is antagonistic to microorganisms that contaminate honey. It has good microbiological quality.Keywords: Stingless bee. Bacillus. Caatinga. INIBIÇÃO DE PATÓGENOS POR BACTEIAS ESPORULANTES ISOLADAS DE MEL DE Melipona sp. (APIDAE: APINAE: MELIPONINI)RESUMO -Este trabalho teve como objetivo isolar bactérias esporulantes a partir do mel de Melipona sp. em ambiente de caatinga e avaliar o seu potencial antagonista com vistas à aplicação na medicina e produção animal. As amostras de méis foram coletadas no município de Serra Talhada-PE, onde o mel foi retirado de quatro colméias diferentes (em triplicata), totalizando 12 amostras que amostras foram diluídas (10 -1 a 10 -5 ) e submetidas a 80 ºC por 20 minutos e resfriadas a 5 ºC para indução de esporos e eliminação de células vegetativas. Em seguida, as diluições foram semeadas em Agar Nutriente e incubadas a 30 º por 72 horas para contagem das unidades formadoras de colônias (UFC). Antes do tratamento térmico foram realizados testes para verificar a presença de fungos filamentosos, leveduras e coliformes totais e termotolerantes. As diluições foram semeadas em Agar Malte e Agar Sabouraud e incubadas a 30 ºC por 14 dias. Coliformes totais e termotolerantes foram quantificados pelo método do número mais provável (NMP). Foram obtidos sete isolados (I) de bactérias esporulantes (Bacillus), porém apenas quatro mostraram algum potencial antagonista. O isolado I-5 apresentou o maior potencial antagonista, inibindo o crescimento de Escherichia coli, Klebsiella sp., Pseudomonas aeruginosa, Salmonella sp. e Staphylococcus aureus. Conhecida no vocabulário popular como abelha-cachorro, irapuã, aripuá ou arapuá, a Trigona spinipes (FABRICIUS, 1793) é uma abelha meliponídea que constrói suas colmeias em árvores e cupinzeiros abandonados. Seu nome vem do Tupi Guarani, yra = mel + puã = redondo, Arapuã: abelha redonda, que significa abelha que faz casa de terra, arredondada. Alguns autores a descrevem como praga agrícola por possuir o costume de cortar os botões florais de certas plantas. Porém, outros a reconhecem e descrevem como polinizadora, podendo até ser utilizada na polinização comercial. A Caatinga apresenta aproximadamente 94 espécies de abelhas nativas, distribuídas em 77 gêneros; um baixo valor de espécies por gênero e uma riqueza relativamente pobre em comparação aos outros ecossistemas brasileiros, mas se por um lado ela possui uma baixa diversidade alfa, por outro possui uma alta diversidade beta, isto é, possui um grande percentual de endemismo e predominância em espécies raras, entre 62 e 67%. O gênero Trigona possui uma distribuição ampla, que vai desde a América Central, passando pelo México, Floresta Amazônica, Floresta Atlântica, Cerrado brasileiro, serras e planaltos do Sudeste brasileiro e marginalmente pelo Sul da América do Sul, em regiões temperadas e subtropicais, incluindo áreas desertas da Argentina e Chaco. No Brasil, a Trigona spinipes ocorre nos estados da Bahia, Ceará, Goiás, Minas Gerais, Mato Grosso do Sul, Paraíba, Pernambuco, Rio de Janeiro, Rio Grande do Sul, Santa Catarina e São Paulo. O trabalho objetivou analisar a difícil convivência da abelha nativa Arapuá e as famílias de agricultores rurais no município de Serra Talhada, do Pajeú Pernambucano.
O comportamento de Apis mellifera em áreas urbanas tem causado acidentes com seres humanos e animais domésticos. Ações que visem minimizar o problema, como a captura, são essenciais. Assim, o presente trabalho buscou compreender a dinâmica de nidificação e a variação anual das ocorrências de enxames e colônias, as suas relações com a precipitação pluviométrica e a floração apícola. O trabalho foi realizado no Sertão Pernambucano, em Serra Talhada, principalmente, e em cidades vizinhas. As ocorrências foram registradas no período de 2014 a 2015. Colônias e enxames foram capturados e levados para o apiário da Unidade Acadêmica de Serra Talhada, ou doados para uma comunidade de assentados. Dados pluviométricos foram coletados, e o levantamento do pasto apícola em Serra Talhada, através de transectos aleatórios de 1.500m a partir do referido apiário. Foram registradas 115 ocorrências, sendo 65% em áreas urbanas e 35% em áreas rurais. Desse total, 76% foram de colônias nidificadas e 24% de enxames provisórios. Em relação ao tamanho, 62% foram considerados grandes, 22% de porte médio e 16% pequeno. O maior número de ocorrências foi verificado entre os meses de março e abril, coincidindo com o período de maior precipitação acumulada e as floradas. Desta forma, as abelhas capturadas possibilitaram uma redução de riscos à sociedade, contribuíram para uma renda extra dos assentados e aulas práticas universitárias. A dinâmica de nidificação de A. mellifera, na área de estudo, mostrou estar relacionada ao período com maior volume de chuvas e também de maior disponibilidade de recursos florais.
