Hyperglycemia is believed to be the major cause of diabetic vascular complications involving both microvessels and arteries as in the retina, renal glomeruli, and aorta. It is unclear by which mechanism hyperglycemia is altering the metabolism and functions of vascular cells, although changes in nonenzymatic protein glycosylation and increases in cellular sorbitol levels have been postulated to be involved. Previously, we have reported that the elevation of extracellular glucose levels with cultured bovine retinal capillary endothelial cells causes an increase in protein kinase C (PKC) activity of the membranous pool with a parallel decrease in the cytosol without alteration of its total activity. Now we demonstrate that the mechanism for the activation of PKC is due to an enhanced de novo synthesis of diacylglycerol as indicated by a 2-fold increase of ['4Cldiacylglycerol labeling from [14C]glucose. The elevated diacylglycerol de novo synthesis is secondarily due to increased formation of precursors derived from glucose metabolism; this formation is enhanced by hyperglycemia as substantiated by elevated [3H1glucose conversion into water. This effect of hyperglycemia on PKC is also observed in cultured aortic smooth muscle and endothelial cells and the retina and kidney of diabetic rats, but not in the brain. Since PKC in vascular cells has been shown to modulate hormone receptor turnover, neovascularization in vitro, and cell growth, we propose that this mechanism of enhancing the membranous PKC activities by hyperglycemia plays an important role in the development of diabetic vascular complications.Hyperglycemia is probably an important etiologic factor in the development of vascular complications in diabetic patients, such as retinopathy (1), nephropathy (2), and accelerated atherosclerosis (3). Using cultured bovine retinal capillary endothelial cells, a cell type prominently involved in diabetic retinopathy (21), we have previously reported that the membranous pool of PKC activity was increased 100% by elevation of glucose level (22) but not by adding mannitol to the medium. Peroxidase/H2O2-mediated radical coupling of 4-hydroxycinnamaldehydes produces 8-O-4-, 8-5-, and 8-8-coupled dehydrodimers as has been documented earlier, as well as the 5-5-coupled dehydrodimer. The 8-5-dehydrodimer is however produced kinetically in its cyclic phenylcoumaran form at neutral pH. Synthetic polymers produced from mixtures of hydroxycinnamaldehydes and normal monolignols provide the next level of complexity. Spectral data from dimers, oligomers, and synthetic polymers have allowed a more substantive assignment of aldehyde components in lignins isolated from a CAD-deficient pine mutant and an antisense-CAD-downregulated transgenic tobacco. CAD-deficient pine lignin shows enhanced levels of the typical benzaldehyde and cinnamaldehyde end-groups, along with evidence for two types of 8-O-4-coupled coniferaldehyde units. The CAD-downregulated tobacco also has higher levels of hydroxycinnamaldehyde and hydroxybenzaldehyde (mainly syringaldehyde) incorporation, but the analogous two types of 8-O-4-coupled products are the dominant features. 8-8-Coupled units are also clearly evident. There is clear evidence for coupling of hydroxycinnamaldehydes to each other and then incorporation into the lignin, as well as for the incorporation of hydroxycinnamaldehyde monomers into the growing lignin polymer. Coniferaldehyde and sinapaldehyde (as well as vanillin and syringaldehyde) co-polymerize with the traditional monolignols into lignins and do so at enhanced levels when CAD-deficiency has an impact on the normal monolignol production. The implication is that, particularly in angiosperms, the aldehydes behave like the traditional monolignols and should probably be regarded as authentic lignin monomers in normal and CAD-deficient plants.

The charge distribution (CD) method, previously introduced as a development of the bond‐valence (BV) approach, is applied for the first time to mineral structures, and specifically to pyroxenes. CD essentially involves the distribution of the Effective Coordination Number (ECoN) of a cation among all the neighboring anions. This distribution is then interpreted in terms of distribution of `charges', where `charge' represents the formal oxidation state. Differently from BV, the CD description depends upon the geometry of each coordination polyhedron, which is characterized through ECoN (a non‐integer number). The contribution of each cation–oxygen bond to ECoN, labelled `bond weight', corresponds to the bond strength in the BV method, but it is defined in terms of bond‐length ratio in each polyhedron and not as a function of the cation–oxygen pair. The ratio q/Q of the formal oxidation number to the computed charge can be interpreted as a measure of the correctness of the structure (cation ratio) and of the degree of over‐ or under‐bonding (anion ratio). A similar interpretation is not possible for the analogous quantities obtained through the BV approach. The analysis in terms of CD of the pyroxene chains (from 101 structures) shows different trends as a function of composition, temperature and pressure; in particular it shows a different behaviour of the two crystallographically independent chains of orthopyroxenes and of P21/c clinopyroxenes.

The alkali-metal clinopyroxene NaTi(3+)Si2O6, one of the rare compounds with trivalent titanium, was synthesized at high temperature/high pressure and subsequently investigated by single-crystal X-ray diffraction methods between 298 and 100 K. One main difference between the high- and the low-temperature form is the sudden appearance of two different Ti(3+)-Ti3+ interatomic distances within the infinite chain of the TiO6 octahedra just below 197 K. This change can be seen as direct evidence for the formation of Ti-Ti singlet pairs in the low-temperature phase. Mean Ti-O bond lengths smoothly decrease with decreasing temperature and the phase transition is associated with a slight jump in the Ti-O bond length. The break in symmetry, however, causes distinct variations, especially with respect to the two Ti-O(apex) bond lengths, but also with respect to the four Ti-O bonds in the equatorial plane of the octahedron. The TiO6 octahedron appears to be stretched in the chain direction with a slightly larger elongation in the P1; low-temperature phase compared with the C2/c high-temperature phase. Polyhedral distortion parameters such as bond-length distortion and octahedral angle variance suggest the TiO6 octahedron in P1; to be closer to the geometry of an ideal octahedron than in C2/c. Mean Na-O bond lengths decrease with decreasing temperature and the variations in individual Na-O bond lengths are the result of variations in the geometry of the octahedral site. The tetrahedral site acts as a rigid unit, which does not show pronounced changes upon cooling and through the phase transitions. There are neither large changes in bond lengths and angles nor in polyhedral distortion parameters, for the tetrahedral site, when they are plotted. In contrast with the C2/c-->P2(1)/c phase transition, found especially in LiMSi2O6 clinopyroxenes, no very large variations are found for the tetrahedral bridging angle. Thus, it is concluded that the main factor inducing the phase transition and controlling the structural variations is the M1 octahedral site.
