BackgroundThe barnacle Balanus amphitrite is a globally distributed biofouler and a model species in intertidal ecology and larval settlement studies. However, a lack of genomic information has hindered the comprehensive elucidation of the molecular mechanisms coordinating its larval settlement. The pyrosequencing-based transcriptomic approach is thought to be useful to identify key molecular changes during larval settlement.Methodology and Principal FindingsUsing 454 pyrosequencing, we collected totally 630,845 reads including 215,308 from the larval stages and 415,537 from the adults; 23,451 contigs were generated while 77,785 remained as singletons. We annotated 31,720 of the 92,322 predicted open reading frames, which matched hits in the NCBI NR database, and identified 7,954 putative genes that were differentially expressed between the larval and adult stages. Of these, several genes were further characterized with quantitative real-time PCR and in situ hybridization, revealing some key findings: 1) vitellogenin was uniquely expressed in late nauplius stage, suggesting it may be an energy source for the subsequent non-feeding cyprid stage; 2) the locations of mannose receptors suggested they may be involved in the sensory system of cyprids; 3) 20 kDa-cement protein homologues were expressed in the cyprid cement gland and probably function during attachment; and 4) receptor tyrosine kinases were expressed higher in cyprid stage and may be involved in signal perception during larval settlement.ConclusionsOur results provide not only the basis of several new hypotheses about gene functions during larval settlement, but also the availability of this large transcriptome dataset in B. amphitrite for further exploration of larval settlement and developmental pathways in this important marine species.

A generalized model is developed which couples the evaporation at a liquid-air interface with the vapor diffusion processes in air to enable an investigation of the mass transport inside an open microtube.Tube inner diameters ranging from 100 to 1200 microns are considered. Evaporation is strongest at the meniscus junction with the tube wall due to the highest local vapor diffusion flux at this location. A temperature gradient is set up from the axis of the tube to the wall and results in Marangoni convection.The three-dimensional flow structure in the microtube is simulated with the effects of Marangoni convection, buoyancy, and the influx of fluid to the interface being included. For horizontal tubes of diameter 100 microns or larger immersed in a water bath, flow asymmetry due to buoyancy is observed. A large vortex is formed in the lower part of the tube cross-section, while a small vortex forms above.However, the primary cause of asymmetry is found to be the external thermal profile imposed on the microtube, especially when the meniscus is far from the outlet of the tube. The simulated flow patterns are found to be consistent with experimental measurements.

An evaporating meniscus in a microchannel is investigated through an augmented Young-Laplace model and the kinetic theory-based expression for mass transport across a liquid-vapor interface. The complete expression for mass transport is employed without any approximations and boundary conditions for the film profile are developed. The thin-film and the intrinsic-meniscus regions are distinguished based on the disjoining pressure variation along the meniscus. While heat transfer in the thin-film region is found to be relatively insensitive to channels larger than a few micrometers in radius, that in the intrinsic meniscus is quite sensitive to channel size. The role of evaporation suppression due to capillary pressure in both regions is discussed. Compared to the relatively small contribution to overall heat transfer from the thin-film region, the micro region (defined here as extending from the non-evaporating region to a location where the film is 1 m thick) is found to account for more than 50% of the total heat transfer.

Individuals of the basidiomycete fungus Armillaria are well known for their ability to spread from woody substrate to substrate on the forest floor through the growth of rhizomorphs. Here, we made 248 collections of A. gallica in one locality in Michigan's Upper Peninsula. To identify individuals, we genotyped collections with molecular markers and somatic compatibility testing. We found several different individuals in proximity to one another, but one genetic individual stood out as exceptionally large, covering hundreds of tree root systems over approximately 75 hectares of the forest floor. Based on observed growth rates of the fungus, we estimate the minimum age of the large individual as 2500 years. With whole-genome sequencing and variant discovery, we also found that mutation had occurred within the somatic cells of the individual, reflecting its historical pattern of growth from a single point. The overall rate of mutation over the 90 mb genome, however, was extremely low. This same individual was first discovered in the late 1980s, but its full spatial extent and internal mutation dynamic was unknown at that time. The large individual of A. gallica has been remarkably resistant to genomic change as it has persisted in place.
