The synthesis and utilisation of acyl azides in a flow apparatus combined with an automated extraction unit is described. This process safely provides multi-100 g quantities of a labile diacyl azide (3) as an intermediate that could not be generated safely by classic batch methods. Its subsequent conversion to the desired amine (4) represents an example for process intensification. The same set-up with an output capacity of >30 g/h was used for the unattended synthesis of benzoyl azide as the final product in solution (tert-butyl methyl ether (TBME), 0.5 M).
A number of N-acryloyl-, N-crotonoyl-, N-(3,3,3-trifluorocrotonoyl)-, N-cinnamoyl-, and N-(3-nitroacryloyl)-4-isopropyl-or -4-phenyl-oxazolidin-2-ones with geminal diphenyl substitution, i.e., 7 -15, have been prepared and used for conjugate additions of organocuprate reagents (Me, i Pr, Ph, 4-MeOPh) in the b-carbonyl ( Table 2) and in the a-carbonyl position (NO 2 -derivative 11 in Scheme 3). The yields and diastereoselectivities are compared with previously tested enoyl-oxazolidinones ( Table 2). Highest diastereoselectivities (> 90%) are always observed with the 4-Ph derivatives (Hruby effect). Nitroacryloyl-oxazolidinones and a corresponding phenylmenthol ester undergo less diastereoselective additions (Scheme 3). A 3-(1-methylethyl)-5,5-diphenyloxazolidin-2-one (DIOZ)-derived Li 2 -enolatenitronate was also tested for a-carbonyl alkylation (Scheme 4). The X-ray crystal structures of three acryloyl-oxazolidinones and of four adducts are described (Tables 1 and 3), and they serve for configurational assignments and description of the stereochemical courses of the additions and alkylation. Possible applications of the nitro compounds for the preparation of b 2 -amino acids are discussed (Scheme 2).
Wir beschrieben in der vorangegangenen Xitteilungl) die Synthese zweier Verbindungen mit dem Gerust des Biotins (IXb), namlieh des DL-epi-Biotins und des DL-epi-allo-Biotins. Bei diesen Synthesen wnrde weitgehend von neuartigen Reaktionen aliphatischer Nitroverbindungctn Gebrauch gemacht. Da zur Zeit, als diese Arbeiten begonnen wurden, relativ wenig uber die Reaktionsnioglichkeiten dieser Stoffklasse bekannt war, wurden die meisten Reaktionen zuerst an einem einfacheren Model1 studiert. Scither ist die Chemie der aliphatischen Nitroverbindungen durch die systematischen Untcrsuchungen in den Laboratorien der ICI in England weitgehend erschlossen worden2).Als Modellsubstanz diente uns das 2-Methyl-3,4-(2'-oxo-tetra8hydro-imidazo1)-thiophan (IXa). Eines der vier Racemate diese-r Striiktur ist von 0. Rchnider et al. 3, beschrieben wordcn.Es wurde zunachst 1-Nitro-2-propanol (Ia), dessen Herstellung aus Nitroniethan und Acetaldehyd gegenuber den beschriebenen Ver-fa8hren4) 5 , verbessert werden konnte, mit Acetylchlorid zum bekannten 1-Nitro-2-acetoxy-propan ( Ib)6) und mit Phosphorpentachlorid zum ebenfalls beschriebenen 1-Nitro-2-chlor-propan ( 1~)~) umgesetzt. Beidc Verbindungen liessen sich durch Reaktion mit Merkaptoaectaldehyd-diathylacetal (11) 7, in Gegenwart von Natriumalkoholat glatt in den Thioather I11 uberfuhren. Dabei liefert die Acetoxyverbindung I b die bessercn Resultate.Die Acetalgruppe in TIT l a s t sich leicht mit vertl. Salzsaure in Dioxan hydrolysieren. Allerdings lasst Rich der dabei in ausgezeichneter Ausbeute entstehende Aldehyd IV nicht durch blosse Destillation analysenrein erhalten. Die intramolekulare Aldolkondensation des Aldehydes IV liefert ein Gemisch isomerer 2-Methyl-3-nitro-4-oxythiophane (Va), welches aus den fruhcr angegebenen Griinden') jel) 111. The results of our measurements are evaluated to support the \-a\lidity of more modern theories. Out of the relation between molecular weight and sedimentation constants the length of the preferential dement A m and the hydrodynamical thickness of the chain tl, are ealculated according t o the work of Kuhn & Edm. The results are found to be 8,s em for Am and 4 a lop8 cm for d,. The viscositymolecular weight relation is graphically evaluated in the usual double logarithmic form, which runs : *Imong the theoretical statements for the calculations of this relationship the one of Debye$ Bueche agrees most closely with our experimental results.An expression for the molar frictional ratio f/fo is tierived for the special case of total immobilisation of the solvent. For this ratio proportionality to the sixth root is found according to the theories mentioned.
