RESUMO O teste imunocromatográfico de fluxo lateral, também chamado de teste rápido, tem sido uma ferramenta cada vez mais utilizada para o diagnóstico de uma vasta gama de alvos biológicos. Devido as suas características, esta plataforma é facilmente adaptada para uso em domicílio ou no campo, não necessitando de grandes recursos tecnológicos ou conhecimento técnico prévio, permitindo o monitoramento qualitativo, e ainda em alguns casos, quantitativo das amostras. Existem diferentes tipos de testes disponíveis comercialmente e a escolha do melhor formato está relacionada aos objetivos pretendidos, ou seja, o tipo de material a ser identificado, tais como antígenos, anticorpos ou qualquer outro material biológico ou químico. Diante disso, este trabalho teve como objetivo uma revisão ampla dos diferentes componentes dos testes imunocromatográficos, vantagens e desvantagens, sua aplicabilidade e avanços para o diagnóstico de doenças de interesse veterinário e humano, além da detecção de resíduos e contaminantes de produtos de origem animal.
The specificity of monoclonal antibodies (mAbs) to desired targets makes these molecules suitable for therapeutic and diagnostic uses against a wide range of pathogens. Phage display antibody libraries offer one method by which mAbs can be selected for, without the use of conventional hybridoma technology. In this work, phage display technology was used to construct, select and characterize a combinatorial single chain fragment variable (scFv) antibody library against bovine herpesvirus type 1 (BoHV-1) from the immune repertoire of chickens immunized with the virus. In silico analysis of the hypervariable domains of the antibody heavy chains revealed a high frequency of scFv fragments with low variability, suggesting that selection had probably been carried out and favored by a few immunogenic viral antigens. The reactivity of the scFv fragments selected against BoHV-1 was demonstrated by Phage-ELISA. A significant increase in antibody reactivity to the target was observed after six rounds of library selection, showing its potential use as a molecule for BoHV-1 diagnosis. The strategy described here opens up a field for the use of phage display as a tool for selection of monoclonal antibodies that could be used for theranostic applications against infectious and parasitic diseases of veterinary interest. Este trabalho teve como objetivo uma revisão dos princípios da técnica de phage display e seu potencial como ferramenta importante para a caracterização de biomoléculas e suas aplicações no diagnóstico e na terapêutica de doenças de interesse veterinário. PHAGE DISPLAY AND THEIR APPLICATION IN VETERINARY MEDICINE ABSTRACTThe phage display technology was first described in 1985. Since that time, different recombinant molecules, such as small peptides or antibody fragments are displayed on the surface of filamentous phage proteins, expanding greatly the applications of this technique. Different prospection methods allow the isolation and characterization, whether in vitro and in vivo, of peptides that are reactive to a wide range of biological targets. This paper aims to review the principles of the phage display technique and its potential as an important tool for the characterization of biomolecules and their applications in the diagnosis and therapy of veterinary diseases.
