Objetivou-se avaliar diferentes concentrações de Kisspeptina (Kp) e o seu efeito na fertilização in vitro de embriões bovinos quanto à qualidade e taxas de desenvolvimento embrionário inicial. No Experimento 1 foi determinada a concentração mínima de Kp a ser utilizada no meio FIV, a partir dos tratamentos: controle (n=240); Kp 10-5 M (n=223); Kp 10-6 M (n=225) e Kp 10-7 M (n=245). No Experimento 2, os ovócitos, foram distribuídos em quatro tratamentos: controle (n=362); Kp 10-1 M (n=425), Kp 10-7 M (n=304), e antagonista P234 4x10-6 M (n=424). Os espermatozoides foram capacitados por gradiente descontínuo de Percoll ® , e co-incubados com os ovócitos por 16 a 18h em estufa de cultura a 38,5°C com 5% de CO 2. Após esse período, os zigotos foram incubados em Fluido Sintético de Oviduto (SOF) durante sete dias nas mesmas condições. A taxa de clivagem foi avaliada 48h após a fertilização e a taxa de blastocistos e estágio de desenvolvimento/qualidade no sétimo dia. Foi adotado um escore numérico de 1 a 4 para avaliação do estágio de desenvolvimento, onde 1 corresponde a blastocisto inicial, 2 blastocisto, 3 blastocisto expandido e 4 blastocisto eclodido.
