Three pathogenic fungi of blueberry (Vaccinium spp.) responsible for dieback disease, identified as Pestalotiopsis clavispora, Colletotrichum gloeosporioides and Lasiodiplodia pseudotheobromae, were isolated in the northwestern region of the state of Michoacán, Mexico. The mycelial growth in vitro of these fungi was inhibited by extracts from Lantana hirta, Argemone ochroleuca and Adenophyllum porophyllum, medicinal plants collected in Sahuayo, Michoacán, Mexico. The extracts showed different degrees of inhibition; the most effective were: M5L extract from L. hirta and M6LFr extract from A. ochroleuca, both of which inhibited 100% of the mycelial growth of P. clavispora and C. gloeosporioides; and M4LS extract from A. porophyllum, which inhibited 100% of the mycelial growth of the three pathogens. The extracts were fractionated by thin layer and column chromatography, and the most active fractions were analyzed by gas chromatography-mass spectrometry. The major compounds identified in L. hirta extract were Phytol and α-Sitosterol. The compounds identified in A. ochroleuca were Toluene and Benzene, 1,3-bis(3-phenoxyphenoxy)-. In A. porophyllum, the compound identified was Hexanedioic acid, bis(2-ethylhexyl) ester. These results show the potential of L. hirta, A. ochroleuca and A. porophyllum as a source of antifungal compounds.
<p><strong>Background: </strong><em>Agave potatorum</em> reproduces mainly through seeds is widely collected for the mezcal production, and there is currently a marked decrease in their wild populations. Therefore, to contribute to its sustainable use, it is important to characterize the morphology of its seeds and to know their germinative characteristics.</p><p><strong>Research question:</strong> Do the year and the collection site have an effect on the size, color, viability and germination capacity of <em>Agave potatorum</em> seeds?</p><p><strong>Species studied</strong>: <em>Agave potatorum</em> Zucc.</p><p><strong>Study site and period of research:</strong> Seeds of <em>Agave potatorum</em> were collected in Oaxaca, Mexico: Miahuatlán (MIAH), in 2014; Coixtlahuaca (COIX), in 2015; and Zaachila (ZAA), in 2015 and 2016.</p><p><strong>Methods:</strong> For the seeds, we analyzed four physical descriptors, five color coordinates, and nine germination variables. We used a completely randomized design with four replications (100 seeds). An analysis of variance, a comparison of means, and a linear correlation analysis were performed.</p><p><strong>Results:</strong> We found that the ZAA16 seeds showed the highest values (<em>P</em> < 0.05) for weight and size, and for the luminosity coordinate (L*, 15.2). Color coordinate values revealed a black color of the seeds, which significantly diminished in luminosity with seeds age and varied with their origin. The coordinate L* was correlated significantly with the total dry weight of seedlings and seed volume.</p><p><strong>Conclusions:</strong> <em>Agave potatorum </em>seeds significantly changed their physical dimensions and germination capacity according to their origin. The luminosity of the black color of seeds decreased with seed age and was correlated with seed volume and seedling total dry weight.</p>
Introducción. Actualmente el maíz es el grano más cultivado en el mundo y es factible mejorar la nutrición de sus consumidores mediante el desarrollo del maíz con proteína de calidad (QPM, en inglés), cuya base es la expresión del gen opaco2, la cual incrementa los contenidos de lisina y triptófano del grano. Objetivo. Analizar químicamente la proteína del grano de líneas e híbridos simples experimentales de QPM y conocer las relaciones entre el genotipo y las variables evaluadas de la calidad proteica. Materiales y métodos. Los experimentos se efectuaron durante el 2017 en la Unidad Profesional Interdisciplinaria de Biotecnología del Instituto Politécnico Nacional (Ciudad de México). Se utilizaron granos enteros de líneas parentales (P1 a P6) y de los híbridos simples directos. Se estimaron lisina, triptófano, y cantidad y calidad de proteína. Se utilizó un diseño completamente al azar con tres repeticiones. Se hicieron análisis de varianza, comparación de medias y análisis de componentes principales. Resultados. Se identificaron como QPM significativos (p ≤ 0,05) a las líneas P2, P3 y P6, y a los híbridos P2 x P4 y P2 x P6. El índice de calidad de proteína se correlacionó con el triptófano (r = 0,9*) y la lisina (r = 0,7*), pero no con la proteína total. Conclusiones. El factor fundamental para desarrollar los QPM fueron los aminoácidos esenciales y la limitante para incrementar su valor nutricional fue la calidad de proteína.
Para establecer su huella genómica y estimar su grado de tolerancia al envejecimiento, semillas de cuatro variedades criollas de maíz azul (Zea mays L.), ‘Cuijingo’, ‘Cocotitlán’, ‘Puebla’ y ‘Oaxaca’, se sometieron a dos tipos de envejecimiento artificial: calor húmedo (CH, 41 0C, 100 % HR, 72 h) y calor seco (CS, 60 0C, 0 % HR, 48 h), con sus respectivos testigos no envejecidos. Las variables de respuesta fueron: protrusión radicular entre 0 y 72 h de imbibición y a 7 d de incubación a 25 0C, número de plántulas normales, anormales y semillas muertas, así como peso seco de plúmula, radícula y total. Se hizo la caracterización molecular de los tratamientos mediante marcadores RAPDs y se construyeron dendrogramas. La protrusión radicular fue significativamente susceptible al CH, mientras que el efecto del CS se expresó en reducción de plántulas normales y acumulación de materia seca, así como en mayor proporción de semillas muertas. Las semillas de la variedad ‘Oaxaca’ se distinguieron por su aptitud para la brotación radicular y por generar plántulas normales con alta capacidad para acumular biomasa, tanto en los testigos como en condiciones de calor húmedo. La huella genómica resultó específica para cada variedad, y en los dendrogramas las semillas se ramificaron con un coeficiente de similitud único para cada variedad: ‘Oaxaca’ (31.6 %), ‘Cocotitlán’ (25.8 %), ‘Puebla’ (19.6 %) y ‘Cuijingo’ (18.6 %), valores que podrían estar relacionados con la capacidad genética para amortiguar, tolerar o restaurar los daños causados por el envejecimiento artificial sobre el ADN, la cual se manifestó directamente en un mejor desempeño fisiológico de las semillas de la variedad ‘Oaxaca’.
