Complete separation of the enantiomers ( 2 a ) and ( 3 a ) was achieved by liquid chromatography of their diastereomeric esters (2c) and ( 3 c ) with (-)-camphanic acidr4] [silica gel columns, petroleum ether/ethyl acetate (92 : 8), separation factor: 1.11. Pure (2c) can also be obtained in 30% yield by By Giinter Helinchen and Gerhard Staiged"] Rigid chiral hydrocarbons and ketones are of special interest as test cases for chiroptic theories. Outstanding representatives are the enantiomeric trishomocubanes (Zf) and ( 3 f ) , in that they are apparently the smallest stable organic molecules of the point symmetry group Dj[l1.While racemic trishomocubane is very readily accessible via rearrangement of the alcohols ( 1 a ) and (1 b ) to the stereoisomeric 4,7-diiodotrishomocubanes with hydriodic acid and their subsequent reduction with zinc, only relatively tedious methods exist for the synthesis of derivatives appearing suitable for separation of the enantion~ers[21. We have how found that 4-hydroxy-7-iodotrishomocubanes are formed in good yields as intermediates in the first mentioned reaction and that these can be easily resolved into their enantiomers. [ Jahrg. 89 ~_ _~_ _ _Wahrend nach den Angaben der Literaturs) bisher nur gelbe bis rote Stoffe als primiire Dehydrierungs-bzw. Oxydationsprodukte von Hydroxyindolen beobachtet wurden, haben wir a.us Dihydro-bzw. 5-Hydroxy-indolen blaue bis violette Chinone erhalten2). Fur diese kommen entsprechend ihrer Bildungsweise nur die Strukturen I oder 11 in Betracht. Zwischen diesen beiden Moglichkeiten konnte zu Gunsten der Struktur I entschieden werden, da wir analoge Verbindungen nicht nur aus den p-Hydroxyderivaten des Skatols, 2-Phenyl-indols und Tetrahydrocarbazols2) sondern auch des 7-Methyl-und 7.8-Dimethyl-tetrahydrocarbazols erhalten haben. Auch 2.3-Dimethyl-, 2.3.7-Trimethyl-und 2.3.6.7-Tetramethyl-dihydroindol gehen in entsprechende Substamen (blaue Nadeln) iiber4), die sich durch ihre Reduzierbarkeit mit Dithionit sowie spektroskopisch und polarographisch als o-Chinone zu erkennen geben.Die Ausbeuten, die man, ausgehend von den p-Hydroxyverbindungen, erhalt, betragen je nach pH -Wert bis zu 95 ?$ der Theorie. Die Reaktion wurde gewohnlich bei p , 6-7 durchgefiihrt.Ausgeprlgt ist die pH-9hhlngigkeit heim 5-Hydroxy-skatol imd 6-Hydroxy-tetrahydrocarhrczol, weniger heim 5-Hydroxy-2-phenyl-indol.Oxydiert. man 5-Hydroxy-ska.tol bei ptt 7 odnr schwach alkalisch (in Aceton .;-Wasser als I.osungsmittel), so erhiilt man eine tiefroto5) Ihsung, die nur wenig Chinon abschcidet.Bei schwach saurem h dagegen kriatallisiert nach vorubergehender Rotfarbung (die nicht in Chloroform geht) innerhalb weniger Xinuten das blauc Chinon. Die Dauer jener Rotfarbung der Gsung, die der Abscheidung des dunklen Stoffes vorausgeht, ist um so kiiner, je murer oxpdicrt wird. Auch ist die Ausbeute dann groBer (vergl. im Vemuchsteil des Beispiel des 7.8-Dimethyl-tetrahydrocerbazolchinons-(5.6) ).Oxydiert man 6-Hydrox~~-tetrahydmcarbazol*) bei p,7, so entsteht ebenfalls zunachst einc intensiv gelbrotc Losung, aus der nach wenigcn Minuten gut ausgebildete, violette Nadeln vom Schmp. 248" kristallisieren. Oxydiert man bei etwa h 5 (KQPO,-Puffer), so scheidet sich fast augenblicklich ein feinkristallines, blauviolettes Produkt in hoherer Ausbeute (80-90%) ab, das bis 310" nicht geschmolzen ist. Mit der Untersuchung dieses Stoffes sind wir noch beschiiftigt.Bemerkenswert ist die lokale Reizwirkung der blauen Chinone, insbesondere auf die Schleimhaute. Sie schmecken nachhaltig brennend, pfefferartig, bisweilen auch wie Wasserstoffperoxyd.
fiillt beim Verdunnen mit Wasser ein feinkristalliner, hellgelber Niederschlag aus; nach Waschen mit Waeser und Trocknen im Exsiccator 50 mg (91.6y0 d. Th.), Schmp. 221-2200. Zur Reinigung werden 40 mg in 20 ccm warmem Benzol gelost. Man filtriert durch eine Aluminiumoxyd-Siiule und wasoht rnit 30 ccm Benzol das gesamte Phenacin wieder heraus, wiihrend eine dunkle Verunreinigung zuriickbleibt. Die Benzol-Lijsung wird eingedampft. Ihr Ruckstand wird in 400 ccm siedendem Petroliither (40°) aufgenommen und die filtrierte Losung auf 160 ccm eingeengt. Durch Tieftemperatur-Kristlliaation werden zitronengelbe Nadeln gewonnen, die bei 221 -222O schmelzen (Lit.-Angabe des Schmp. ebenso). Man trocknet bei 80/1 Torr. Cz0H,,N2 (280.3) Ber. C85.69 H4.32 N9.99 Gef. C 85.79 H4.72 N9.89 Die Lijsung in konz. Schwefelsiiure ist preuBischblau gefarbt. Chinolinchinon-(5.8) a u s 8-Oxy-chinolin: 1.16 g 8-Oxy-chinolin (8 mMol) werden in 80 ccm Methanol gelost. Die Lijsung wird auf 5 O abgekuhlt und mit der ebenfalls kalten Lijsung von 4.8 g Fmmyschem Salz in 360 ccm Wasser + 80 ccm m/6 KH,PO, versetzt. Die Farbe der Mischung iindert sich nur lsngmm; nach 2 Stdn. ist sie hellbraun, nach Stehenlassen uber Nacht dunkelrot. Nach 20 Stdn. wird mit Chloroform extrahiert (3 x 250 ccm). Der mit Waaser gewaschene und iiber Natriumsdfat getrocknete Auszug hinterliiBt beim Abdampfen einen dunkelbraunen Sirup, der von gelben Kristallen durchaetzt ist. Man oimmt den Ruckstand mit 100 ccm warmer, etwa 7nHCI a d . Die durch eine Fritte fltrierte Lijsung liiBt beim Erkalten gelbe Nadeln eines H y d r o c h l o r i d s kristellisieren, die sich gegen 1500 sinnobemt, bei etwa 2200 dunkelbraun frirben und eich gegen 330-2350 vollstiindig zeraetzen. Die Ausbeute betriigt 750 mg oder 38% d.Theorie. 150 mg werden aus 55 ccm 3nHCl von 800 umgelost: 100 mg (Absaugen bei OO). Daa Umkristllisieren wird zur Analyse noch zweimal wiederholt. Verlust bei 100°/12 Torr/ 12 Stdn. 7.6y0, bei 55O/1 Torr/24 Stdn. 6.2%; ber. fiir 1 H,O 7.2%. C,H,O,NCI.HCl (232.1) Ber. C46.58 H3.04 Gef. C47.08 H3.22 Die bei 1 0 0 0 getrocknete Substanz ist orangegelb. Durch Umlosen aus 3nHC1 von SOn geht eie wieder in das hellgelbe Hydrat iiber und zersetzt sich dann bei 230-235O.Sie wird lufttrocken analysiert.(',H,02NCl~HCl+H,0 (250.1) Ber.
Starre chirale Kohlenwasserstoffe wie (1) und (2) interessieren als Testfälle für chiroptische Theorien. (‐)‐Trishomocuban (1) und (+)‐Trishomocuban (2), die jetzt enantiomer rein erhalten wurden, sind offenbar die kleinsten stabilen organischen Moleküle der Punktgruppensymmetrie D3.
