Os fungos endofíticos vivem em simbiose com a planta hospedeira, sendo que muitos podem apresentar a capacidade de produzir os mesmos metabólitos ativos que seus hospedeiros, tornando-se um reservatório de uma infinidade de produtos naturais. Esta pesquisa investigou a planta Mirindiba (Buchenavia tomentosa) e a atividade antimicrobiana exercida pela planta e por fungos endofíticos isolados de suas folhas. Extratos aquosos da Planta e dos fungos foram testados contra bactérias e um fungo filamentoso, sendo que os extratos do vegetal e dos fungos apresentaram inibição das seguintes bactérias patogênicas: Salmonella Typhimurium, Staphylococcus epidermidis e Shigella flexneri e do fungo fitopatogênico Rhizopus stolonifer, demonstrando seu potencial biotecnológico como produtores de substâncias antimicrobianas. As concentrações inibitórias mínimas (CIMs) dos fungos que apresentaram propriedades antimicrobianas foram determinadas. Dois fungos (FC12 e FC31) inibiram as 3 bactérias testadas. Todas as bactérias foram inibidas por pelo menos um fungo na concentração de 25 µg/ml. Uma atividade antifúngica também foi observada pela difusão de substâncias produzidas pelo isolado (FC123), que inibiu em 53% o crescimento de Rhizopus stolonifer, causador da podridão mole em frutos. Este é o primeiro relato de isolamento e avaliação das propriedades antimicrobianas de fungos endofíticos da B. tomentosa.
A vida pós-colheita da manga é limitada pela deterioração fisiológica causada pelo excessivo amadurecimento da fruta e pelo desenvolvimento de patógenos que ocasionam podridões. A Mancha angular é provocada pelo microrganismo Xanthomonas campestris PV mangiferaeindicae é uma doença que ataca ramos, folhas, inflorescências e frutos da mangueira, em períodos úmidos prolongado. A crescente preocupação com o conceito de qualidade mercadológica e a preservação do ambiente têm aumentado a procura por frutas saudáveis e sem resíduos de agroquímicos, havendo uma necessidade crescente de estratégias de controle alternativas. A primeira medida de controle é a escolha do local, deve-se evitar locais sob influência de grandes massas de água, como represas das hidroelétricas, pois criam um ambiente favorável ao desenvolvimento da bactéria. A segunda medida é o uso de mudas sadias e de viveiros localizados em regiões onde a doença não ocorra. Várias pesquisas estudam a estrutura química e a atividade antimicrobiana natural de plantas, frutos, hortaliças, grãos e ervas. Os esforços têm-se concentrado na utilização de recursos naturais como extratos e óleos essenciais. Palavras-chave: Xanthomonas campestris, manga, controle biológico.
The conventional methods used for angular stain control are generally chemical methods, however the use of these products can cause high environmental impact and damage to consumer health if it is used in large quantities and undiluted and applied correctly. Based on this problem, this work aimed to evaluate in vitro alternative forms of control using Saccharomyces yeast (with probiotic potential), ethanolic extracts of Mauritia flexuosa (Buriti) and Miconia albicans (Cinnamon-old) plants. To evaluate four GRAS substances in angular leaf spot control caused by Xanthomonas campestris pv. Mangifera indica, during the postharvest period in mangoes. In vitro results using antagonist yeasts showed no inhibitory effect against X. campestris. However, the extracts of the plants Miconia albicans and Mauritia flexuosa showed a significant inhibition. Thus, as two GRAS substances, 1%, 1.5% and 3% sodium carbonate and 3% sodium bicarbonate inhibited X. campestris growth 100%. Given the results obtained, the plant extracts and the GRAS substances tested were effective in controlling phytobacteria and proved to be an alternative in controlling angular leaf spot, thus avoiding economic losses during the mango postharvest phase.
Fungos endofíticos habitam o interior das plantas sem causar-lhe danos, sendo encontrados em órgãos e tecidos vegetais como folhas e ramos. Os pressupostos da interação endófito-planta hospedeira sugerem que as propriedades terapêuticas podem estar no fungo e não na planta ou, provavelmente, na interação entre ambos; por isso é crescente o interesse por endófitos de plantas medicinais. A bactéria grampositiva Streptococcus pneumoniae e a gram-negativa Klebsiella pneumoniae são bactérias patogênicas importantes, já que atuam como agentes de infecções, incluindo doenças respiratórias e infecções hospitalares. Este trabalho teve como objetivo a obtenção de compostos voláteis de fungos endofíticos isolados da planta medicinal Costus spiralis (jacq) Roscoe (Costaceae) e a avaliação antibacteriana dos mesmos contra S. pneumoniae e K. pneumoniae. O teste de verificação de substâncias voláteis foi realizado utilizando placas de Petri com remoção da faixa do ágar da porção central, a fim de evitar o efeito de difusão. Foi inoculado, em lados opostos da placa, o endofítico e o patógeno e as placas foram seladas com parafilme. Os controles negativos foram realizados com a inoculação de cada patógeno no meio de cultura sem o antagonista, sendo que não foi constatada a inibição do crescimento dos patógenos incubados na presença do endofítico teste, comparando-se com o controle. Desse modo, a descoberta de novos antibióticos se faz importante devido o aparecimento de patógenos resistentes, bem como a evolução de novas doenças e a toxicidade de medicamentos que já existem. A Amazônia detém enormes reservas nativas, ricas em frutas que apresentam grande importância social e econômica, dentre as quais destaca-se a Pupunha (Bactris gasipaes). Por meio da biotecnologia pode-se fazer uso de microrganismos com potencial probiótico associados a esses frutos regionais e disponíveis nessa região. Assim, o trabalho teve como objetivo isolar linhagens microbianas (Lactobacillus sp. e Saccharomyces sp.) com potencial probiótico e biotecnológico, obtidas da fermentação espontânea do fruto Pupunha. Os frutos, obtidos em feiras livres de Manaus, foram submetidos a fermentação espontânea durante 15 dias. Após este período isolou-se 185 microrganismos que foram caracterizados quanto a sua morfologia, teste de catalase, coloração de gram, e alguns testes de potencial probiótico como o antagonismo frente a enteropatógenos e teste de temperatura. Os isolados foram agrupados em 4 morfotipos diferentes apresentando coloração de gram +, catalase – e micromorfologia de cocobacilos sendo sugestivos de Lactobacillus sp.. Todos apresentaram resistência a altas temperaturas e os morfotipos I e II apresentaram maior potencial antagonista frente as cepas patogênicas. Durante todo o processo de fermentação espontânea da Pupunha não foram observados isolados das leveduras do gênero Saccharomyces sp..
