Aims: The objectives of this study were to screen chitinolytic bacteria isolated from soil of Taman Nasional Bukit Duabelas, Jambi, Indonesia. Isolates were selected based on chitinolytic index and antagonism activity of Colletotrichum capsici. Chitinase enzyme from selected isolates was investigated for growth inhibition of C. capsici. Methodology and results: Two chitinolytic bacteria were selected based on their ability to degrade colloidal chitin and inhibit of the growth of C. capsici. Those isolates were KAHN 15.12 and SAHA 12.12, identified as Serratia marcescens and Bacillus thuringiensis respectively based on 16S rRNA gene. The chitinase maximum specific activity of isolate KAHN 15.12 was 52.03 U/mg after 36 h of incubation and SAHA 12.12 was 45.67 U/mg after 24 h of incubation. The enzyme was precipitated by ammonium sulfate 40% and 60% respectively for KAHN 15.12 and SAHA 12.12. The precipitated chitinases were active over a broad range of pH (5 to 10) and temperature (20 to 80 °C). Enzymes were stable in optimum temperature for 180 min. The precipitated of chitinase KAHN 15.12 and SAHA 12.12 had five and two protein bands respectively on SDS-PAGE gel. Chitinases exhibited an antifungal activity against C. capsici at concentration of 60 ppm. Conclusion, significance and impact of study: Isolates KAHN 15.12 and SAHA 12.12 were successfully selected by their ability to degrade colloidal chitin and inhibit the growth of C. capsici. The isolates had a broad range of pH and temperature, moreover relatively stable at the optimum temperature. Chitinase was effective as biological control for anthracnose caused by C. capsici in chilli.
Setiap siswa memiliki motivasi belajar dan prestasi belajar yang berbeda sehingga minat atau dorongan untuk menempuh pendidikan yang lebih tinggi juga berbeda. Tujuan dari penelitian ini adalah (1) untuk mengetahui hubungan antara motivasi belajar dengan minat melanjutkan pendidikan peserta didik kelas XII MIPA SMA Negeri 1 Majene (2) untuk mengetahui hubungan antara prestasi belajar dengan minat melanjutkan pendidikan peserta didik kelas XII MIPA SMA Negeri 1 Majene. (3) untuk mengetahui hubungan antara prestasi belajar dan motivasi belajar dengan minat melanjutkan pendidikan peserta didik kelas XII MIPA SMA Negeri 1 Majene. Populasi penelitian yaitu seluruh siswa kelas XII MIPA SMA Negeri 1 Majene dengan jumlah 156 orang. Adapun teknik pengambilan sampel menggunakan teknik simple random sampling yang terdiri atas 112 siswa dengan perwakilan masing-masing kelas XII MIPA. Pengumpulan data motivasi belajar dan minat siswa untuk melanjutkan pendidikan menggunakan angket yang disebarkan dalam bentuk google form melalui aplikasi WhatsApp, dan pengumpulan data prestasi belajar menggunakan nilai rapor (evaluasi kognitif) peserta didik kelas XI MIPA pada pelajaran biologi semester genap tahun pelajaran 2020/2021. Hasil penelitian ini adalah (1) terdapat hubungan antara motivasi belajar dengan minat melanjutkan pendidikan pada peserta didik kelas XII MIPA SMA Negeri 1 Majene diketahui dari nilai signifikansi (0,000). (2) tidak terdapat hubungan antara prestasi belajar dengan minat melanjutkan pendidikan pada peserta didik kelas XII MIPA SMA Negeri 1 Majene yang ditunjukkan melalui nilai signifikansi (0,141). (3) terdapat hubungan antara motivasi belajar dan prestasi belajar dengan minat melanjutkan pendidikan pada peserta didik kelas XII MIPA SMA Negeri 1 Majene dengan melihat nilai signifikansi (0,000).
Penelitian ini bertujuan untuk mengetahui penerapan aplikasi google classroom sebagai media pembelajaran di masa pandemi covid-19 serta kendala penggunaan google classroom selama pembelajaran daring. Penelitian ini merupakan penelitian kualitatif studi kasus. Data dikumpulkan melalui angket terhadap siswa kelas XI MIPA 2 SMAN 1 Tapalang, wawancara kepada guru, dan observasi kelas. Data dianalisis melalui reduksi data, penyajian data, dan penarikan kesimpulan. Analisis hasil penelitian ini didapat bahwa, penerapan aplikasi google classroom sebagai media pembelajaran cukup efektif dan efisien untuk membantu pembelajaran daring. Penggunaan google classroom mudah diakses akan tetapi dampak yang diperoleh yakni sulitnya siswa dalam memahami pembelajaran sehingga terjadi penurunan hasil belajar yang dipengaruhi oleh faktor internal maupun eksternal siswa. Kurangnya interaksi antara guru dan siswa melalui google classroom menyebabkan siswa lebih memanfaatkan internet untuk menjawab soal evaluasi. Kendala yang dihadapi yakni, keterbatasan paket internet, ketersediaan jaringan dan tidak semua siswa memiliki gawai seperti smartphone dan laptop.
Abstract. In the growth of microorganisms such as fungi, it is necessary to grow a medium that can provide nutrients and as an energy source. Other alternative carbohydrate sources include potatoes, breadfruit, sago, and cassava, which is a distinctive and easy to find food companion. This type of research is descriptive with cross sectional design that aims to determine the difference in alternative media variation from various sources of carbohydrate to Candida albicans. The samples were then made to be the treatment for the creation of alternative media from various carbohydrate sources that further carried out the growth test against Candida albicans. The results of the study are the average growth of colonies on alternative media from Potato carbide sources, namely 655 colonies, breadfruit 1380 colonies, cassava 862 colonies, 372 sago colonies, and semi-synthetic media as a control of 874 colonies. This indicates there is growth of Candida albicans on all alternative media so that it can be used as an alternative medium of fungi growth, as well as the best growth of Candida albicans found in the media Breadfruit Dextrose Agar.
