We recently described a maize mutant caused by an insertion of a Helitron type transposable element (Lal, S.K., Giroux, M.J., Brendel, V., Vallejos, E. and Hannah, L.C., 2003, Plant Cell, 15: 381-391). Here we describe another Helitron insertion in the barren stalk1 gene of maize. The termini of a 6525 bp insertion in the proximal promoter region of the mutant reference allele of maize barren stalk1 gene (ba1-ref) shares striking similarity to the Helitron insertion we reported in the Shrunken-2 gene. This insertion is embedded with pseudogenes that differ from the pseudogenes discovered in the mutant Shrunken-2 insertion. Using the common terminal ends of the mutant insertions as a query, we discovered other Helitron insertions in maize BAC clones. Based on the comparison of the insertion site and PCR amplified genomic sequences, these elements inserted between AT dinucleotides. These putative non-autonomous Helitron insertions completely lacked sequences similar to RPA (replication protein A) and DNA Helicases reported in other species. A blastn analysis indicated that both the 5' and 3' termini of Helitrons are repeated in the maize genome. These data provide strong evidence that Helitron type transposable elements are active and may have played an essential role in the evolution and expansion of the maize genome.

In this paper, we describe a novel nonlabeled biosensor with high diagnostic potential for rapid and sensitive detection of antigens in complex biological samples. The biosensor comprises a piezoimmunosensor (PZ) displaying a specially constructed recombinant antibody on its surface. The recombinant single-chain fragment variable (scFv) antibody contained a cysteine within the linker amino acid sequence used to join the scFv variable heavy and light chains. The presence of cysteine induced the scFv construct to self-assemble as a densely packed rigid monolayer on the gold surface of a quartz crystal microbalance. scFv molecules in this self-assembled mono-layer (SAM) exhibited a defined orientation and high areal densities, with scFv-modified microbalance surfaces displaying 35 times as many variable antigen-binding sites per square centimeter as surfaces modified with whole antibody. Experimental data show that the scFv SAM PZ is superior to Fab fragment, Fab fragment containing a free sulfhydryl group (i.e., Fab-SH), and whole antibody PZs regarding sensitivity and specificity. Because of their small uniform size (MW ≈ 27000) and the ease with which they can be modified using genetic engineering, scFv's have significant advantages over whole antibodies in microbalance biosensor systems. We demonstrate here that the use of scFv containing a cysteine within the scFv linker sequence (i.e., scFv-cys) for preparation of biosensor surfaces markedly increases the density of available antigen-binding sites, yielding a system that is highly selective, rapid, and capable of detecting low concentrations of antigens in complex samples.Biosensor systems that detect biological and chemical agents have important medical, environmental, public safety, and defense applications. An ideal biosensor would be sensitive, rapid, reliable, robust, and inexpensive. Piezoimmunosensors (PZs) are a type of biosensor utilizing antibodies and a quartz crystal microbalance (QCM) to detect minute changes in mass as antigens bind to the antibodies on the QCM surface. 1,2 Although their diagnostic potential is theoretically quite high, in practice, the usefulness of PZs has been limited by the fact that typical IgG antibodies can trap or nonspecifically bind irrelevant molecules, thus yielding false positive signals in assays. Additionally, there remains some skepticism concerning their applicability as biosensors due to the complexity of the physical properties of biofilms in a liquid that make it difficult to establish an explicit relationship between the added mass and a change in the resonant frequency. The QCM gives a direct response signal that characterizes a binding event between an antibody layer, immobilized on the surface of the QCM or other transducers, and the antigen to be detected. The major structural proteins present in the paraflagellar rod of Trypanosoma cruzi migrate on SDSpolyacrylamide gels as two distinct electrophoretic bands. The gene encoding a protein present in the faster migrating band, designated PAR 2, has been identified previously. Here we report the isolation and partial characterization of three genes, designated par 1, par 3, and par 4, that encode proteins present in the two paraflagellar rod protein bands. Peptide-specific polyclonal antibodies and monoclonal antibodies against the four proteins encoded by these genes shows that PAR 1 and PAR 3 are present only in the slower migrating paraflagellar rod band, and that PAR 2 and PAR 4 are present only in the faster migrating band. Analysis of the nucleotide sequence of these genes and the amino acid sequence of the conceptual proteins encoded by them indicates that par 2 shares high sequence similarity with par 3 and both are members of a common gene family, of which par 1 may be a distant member. Analysis of gene copy number and steady-state RNA levels suggest that the close stoichiometric ratio of the four PAR proteins is likely maintained by homeostatic regulation of RNA levels rather than gene dosage.

M line Biomphalaria glabrata snails of 4-, 6-, 8-, 10-, 12-, or 20-mm shell diameter were individually exposed to 10 miracidia each of Echinostoma paraensei. Snails 10 mm in size or larger were found to be significantly less likely to harbor intraventricular sporocysts than snails in smaller size categories. The percentage of snails with intraventricular sporocysts that also developed hemocyte encapsulation responses generally increased with snail size, whereas the number of snails that ultimately became heavily parasitized with large numbers of daughter rediae decreased significantly with snail size. However, at least some snails in each size category developed such disseminated infections. Comparative histological study of 6- and 12-mm snails revealed that parasites readily penetrated both groups of snails, but were more likely to be encapsulated and destroyed in larger snails. Encapsulation reactions were noted from 1 to 15 days postexposure (dpe) in 12-mm snails, indicating that unlike other commonly studied models of trematode-gastropod interactions, snail resistance is not always manifested during the first few days following exposure. Upon infection with E. paraensei, both 6- and 12-mm snails showed significant increases in the number of circulating hemocytes/mm3 of hemolymph. In 6-mm snails, such increases occurred concurrently with successful parasite development. Hemocyte counts in 6-mm snails were significantly elevated from 4 to 15 dpe whereas in 12-mm snails they were significantly elevated from 2 to 30 dpe. A significant degree of resistance to E. paraensei develops as B. glabrata grows and attains sexual maturity. A mechanistic understanding of this phenomenon awaits further investigation.
