Theoretischer Teil.Wir haben darüber berichtet 1 ), daß Prolyl-glycin und Prolylglycyl-glycin durch Hefeauszüge und auch durch rohe, lediglich vom tryptischen Anteil befreite Erepsinlösungen aus Darm gespalten werden, während mit Trockenpräparaten der Dipeptidase und der Amino-polypeptidase aus Hefe oder auch mit solchen des ereptischen oder defc tryptischen.Enzymgemisches tierischer Herkunft keine Spaltung beobachtet wurde. Dieser Befund, zusammen mit den vorliegenden Erfahrungen über den Wirkungsmechanismus der Aminopeptidasen schien uns die Annahme einer besonderen, auf die Spaltung von Prolylpeptiden eingestellten Peptidase nahezulegen.Inzwischen haben E. Abderhalden und 0. Zumstein 2 ) das Verhalten mehrerer Prolinpeptide geprüft und dabei unsere Ergebnisse nicht bestätigen können. Nach den Befunden der genannten Autoren werden von den Lösungen des Darmerepsins Prolyl-glycin gar nicht, Prolyl-glycyl-glycin und Prolyl-alanin sehr wenig, Prolyl-leucin und Prolyl-leucyl-glycin aber rasch und weitgehend gespalten. Danach würden die einzelnen Prolylpeptide in bezug auf ihre enzymatische Angreifbarkeit ohne ersichtliche Regel voneinander unterschieden sein, ihr Verhalten würde ein überzeugendes Beispiel für die "vollkommene. Launenhaftigkeit" sein, die nach A. Po dor 3 ) für die Spezifität der Peptidasen
In unserer Mitteilung über die enzymatische Spaltbarkeit der Prolinpeptide 1 ) haben wir angegeben, daß reine Prolinpeptide keine Ninhydrinreaktion zeigen. Herr Abderhalden macht uns darauf aufmerksam, daß auch mit völlig reinen Prolinpeptiden orangerote bis weinrote Färbungen erhalten werden, wenn man die Reaktion bei genau neutraler Reaktion und in hinlänglich konzentrierter Lösung ausfuhrt Wir können dies auch für die von uns dargestellten Peptide bestätigen. Die kurze Angabe ,Ninhydrinreaktion negativ* in unserer Arbeit rührt daher, daß die verhältnismäßig schwachen und im Farbton von der typischen blauen Ninhydrinreaktion stark verschiedenen Färbungen unsererseits nicht als positive Reaktion gewertet worden sind. Ein wesentlicher Unterschied in bezug auf den Ausfall der Ninhydrinreaktion scheint uns danach zwischen den von Abderhalden und den von uns dargestellten Peptiden nicht mehr zu bestehen. -Bezüglich der enzymatischen Spaltbarkeit der Prolinpeptide sind Abderhalden und Mitarbeiter inzwischen zu Befunden gelangt, die im wesentlichen mit den unseren übereinstimmen und nach Abschluß der Versuche an anderer Stelle mitgeteilt werden sollen." l )
