Die Addition von Sulfenylchloriden 1 an 2-Alkensaureester 2 f i i r t zu Isomerengemischen aus 2(3)-Chlor-3(2)-thioalkansiiureestem 3/4, dagegen ergibt die Addition von Thiolen 7 an 2-Chlorpropensiiure-methylester (6) isomerenreine 2-Chlor-3-thiopropansiiure-methylester 3. Die Isomerisierung von 3 zu 4 in Abhangigkeit von den Reaktionsbedingungen wird untersucht; sie erfolgt besonders leicht bei hoheren Temperaturen. Aus 3 werden mit Natriumazid in Gegenwart von PT-Katalysatoren und Temperaturen unterhalb 55 "C die isomerenreinen 2-Azido-Verbindungen 8 erhalten, die rnit HzS/Pyridin/Wasser oder durch Hydrierung rnit Re2& als Katalysator die entsprechenden S-substituierten Cystein-Derivate 9 bzw. 10 mit Ausbeuten bis zu 70% iiber alle Reaktionsstufen ergeben. DL-CyStein ist als HydrochloridHydrat 16 durch siiurekatalysierte Hydrolyse der 2-Thiazoline 1% . HCI und 15e, die durch HCI-katalysierte Addition von Thioacetamid (11 b) an a-Chloracrylsaure (12) bzw. deren Amid 13 und anschlieknde basische Cyclisierung erhalten werden, auf einfache Weise rnit guten Gesamtausbeuten zuganglich.Cystein wird heute noch hauptsachlich durch reduktive Spaltung aus Cystin gewonnen, das bei der sauren Hydrolyse von Keratin aus menschlichen und tierischen Haaren anfallt; der dariiber hinausgehende Bedarf mu0 jedoch durch Synthesen gedeckt werden4). Besonders einfach ist DL-CyStein durch saure Hydrolyse von 4-Cyan-2,2-dimethylthiazolidin, dem Additionsprodukt von Blausaure an das leicht zugangliche 2,2-Dimethyl-3-thiazolin ' ), zu erhalten. 2-Halogen-3-thiopropans5ure-Derivate, die rnittels nucleophiler Aminierung Cystein-Derivate ergeben sollten, sind durch Addition von Thiolen an a-Halogenacrylslure-Derivate6) oder von Sulfenylchloriden an Acrylsaure-Derivate" zuganglich; bei der Addition von RSCl entstehen aber neben den gewiinschten Produkten auch die isomeren 3-Halogen-Derivate7).Bereits 1952 hat Gunderrnann eine Cystein-Synthese iiber die Addition von Benzylrnercaptan an 2-Chloracrylonitril beschrieben 8a). Das nach Umsetzung rnit Arnmoniak und anschliel3ender saurer Hydrolyse erhaltene Produkt war jedoch nicht S-Benzylcystein, sondern S-BenzylisocysteinRb). Fur eine praktische Anwendung des angefiihrten Syntheseweges, der Arninierung von 2-Halogen-3-thiopropansaure-Derivaten zu Cysteinen, war es deshalb notwendig, deren Jsomerisierungsneigung sowohl bei der Darstellung als auch bei der Umsetzung rnit N-Nucleophilen zu untersuchen, woriiber wir in der vorliegenden Arbeit berichten. ChemInform Abstract The addition of sulfenyl chlorides such as (I) to the acrylate (II) gives mixtures of esters such as (III) and (IV) whereas addition of thiols (V) to the chloropropenate (VI) provides (III) exclusively. The dependence of the isomerization of (III) to (IV) on the reaction conditions is investigated; at higher temp., the formation of (IV) is especially favored. Reaction of (III) with NaN3 (VII) affords the azido esters (VIII) which give the cysteine derivatives (IX) of (X) under appropriate reduction conditions. D,L-Cysteine is obtained in good yield as hydrochloride hydrate (XIV) by HCl-catalyzed hydrolysis of the thiazoline (XIII).
