RESUMO -Pesquisas tem sido conduzidas visando determinar métodos de detecção de organismos geneticamente modificados (OGM), devido, principalmente, à importância sobre a atividade comercial. Atualmente, são utilizados bioensaios, que avaliam características fenotípicas das plântulas, testes de ELISA e Kits, que possibilitam identificar proteínas transgênicas específicas de DNA. O objetivo deste trabalho foi comparar a eficiência dos métodos para detectar sementes de soja Roundup Ready (RR). Amostras de sementes de soja geneticamente modificada (GM) resistente ao glifosato e de sementes do parental suscetível foram submetidas a bioensaios (pré-embebidas, umedecidas em substrato, imersas em solução contendo glifosato e pulverização de plântulas), Kit Trait Test e detecção pelo método da PCR. Os bioensaios são métodos mais eficientes comparando a relação custo/benefício na detecção de sementes de soja GM, sendo o método de pré-embebição o mais indicado e a análise das características morfológicas de plântulas é importante nos bioensaios para detecção de sementes de soja GM.Termos para indexação: Glycine max, transgênico, herbicida, OGM. COMPARISON OF METHODS TO DETECT GENETICALLY MODIFIED SOYBEAN SEEDS RESISTANT TO GLYPHOSATEABSTRACT -Due to the great importance of the trade business, there are significant world efforts in establishing GMO detection methods. Currently, bioassays that analyse seedling phenotype, the ELISA test and kits that enable specific transgene protein identification, and PCR to detect specific DNA sequences are frequently used. The aim of this study was to compare the efficiency of different methods to detect RR soybean. Genetically modified seed samples, tolerant to glyphosate and susceptible parental seeds were submitted to bioassays (pre-imbibed, imbibed and immersion in herbicide solutions and seedling spraying), the Kit trait test and PCR detection. Bioassays were shown to be more efficient, comparing the ratio cost/benefit. The seedling visual analysis is a very important parameter to be considered in GM soybean seeds detection.
-The objective of the present study was to evaluate the efficiency of X-Rays in identifying fissures in artificially dried rice seeds and the relationship between damage and seed performance in the germination test. Irrigated rice seeds of the IRGA 417 and IRGA 420 cultivars were harvested with 23.3 and 24.5% water content respectively and submitted to stationary drying treatments at 32, 38, 44 and 50 °C. X-Rays were taken of subsamples of 100 seeds for each treatment, using an MX-20 X-ray equipment. The X-Rayed seeds were classified from 1 to 3, where 1 corresponded to seeds without fissures, 2 to seeds with non-severe fissures and 3 to seeds with severe fissures. The same X-Rayed seeds were planted and on the seventh day the seedlings (normal or abnormal) and dead seeds were photographed and evaluated to verify any relationship between the fissures and physiological potential. Higher drying temperature increased the percentage of fissures in the two cultivars, which can adversely affect their germination. Seeds with fissures can be identified using X-Rays.Index terms: Oryza sativa L., image analysis, seed quality, X-Ray images, germination.Utilização de raios-X na avaliação de fissuras em sementes de arroz submetidas à secagem artificial RESUMO -O presente trabalho avaliou a utilização de raios-X na identificação de fissuras em sementes de arroz, após a secagem e a relação desses danos com a germinação de sementes. Utilizaram-se sementes de arroz irrigado, cultivares IRGA 417 e IRGA 420, colhidas com teores de água de 23,3 e 24,5%, respectivamente, submetidas aos tratamentos de secagem estacionária, sob as temperaturas de 32, 38, 44 e 50 °C. Subamostras de 100 sementes provenientes de cada tratamento foram radiografadas em aparelho de raios-X (MX-20). As sementes radiografadas foram classificadas em: 1 -sementes sem fissuras; 2 -sementes com fissuras não severas e 3 -sementes com fissuras severas. As mesmas sementes radiografadas foram colocadas para germinar e após sete dias foram avaliadas, sendo fotografadas as respectivas plântulas ou as sementes mortas, para o estabelecimento da relação entre fissuras e desempenho fisiológico. O aumento da temperatura de secagem proporcionou aumento da porcentagem de sementes com fissuras das duas cultivares, sendo que as fissuras severas afetaram negativamente a germinação. As imagens de raios-X permitem identificar fissuras em sementes de arroz submetidas à secagem artificial e correlacioná-las com a presença de plântulas normais e anormais no teste de germinação.Termos para indexação: Oryza sativa L., análise de imagens, qualidade de sementes, imagens de raios-X, germinação.1
O cultivo mundial de soja geneticamente modificada (GM) resistente ao glifosato é crescente e a presença dessas sementes em lotes de sementes convencionais tornou-se um problema para o comércio internacional da soja. Reconhecendo a importância dos novos mercados e dos produtos GM, a Tecnologia de Sementes terá que assegurar a pureza genética dos produtos derivados da biotecnologia através de testes confiáveis, práticos e de baixo custo. Nesse contexto, os objetivos do trabalho foram verificar a eficiência do teste de germinação com herbicida no substrato (bioensaios) na detecção e na quantificação de misturas de cultivares GM em amostras de sementes convencionais. Amostras de sementes convencionais foram preparadas com 0, 1, 3 e 5% de sementes de soja GM e submetidas aos métodos de pré-embebição, substrato umedecido e imersão em herbicida, instaladas em bandejas plásticas contendo 25 sementes, com e sem associação ao kit de detecção de OGM. Na seqüência, amostras contaminadas com 0, 1, 3, 5 e 8% de sementes de soja GM foram semeadas em rolos de papel (25 e 50 sementes/rolo) e bandejas plásticas com 25 sementes seguindo o método de pré-embebição em herbicida. Aos seis dias após a instalação, avaliaram-se comprimento de hipocótilo, comprimento de raiz, número de raízes secundárias e comprimento da maior raiz secundária. O método de pré-embebição é o mais eficiente para a detecção e quantificação da presença de sementes de soja GM, permitindo 100% de acertos na detecção, independentemente da porcentagem de contaminantes nas amostras convencionais de sementes de soja e apresenta exatidão na quantificação da presença de até 3% de sementes de soja GM em amostras convencionais, sendo sua precisão condicionada ao porcentual de mistura presente na amostra.
RESUMO -Esta pesquisa teve por objetivos ajustar as metodologias de bioensaios para detecção de sementes de soja tolerante ao herbicida glifosato, estabelecendo o tempo mínimo necessário para uma avaliação segura, bem como a caracterização dos sintomas causados pelo herbicida nas plântulas não geneticamente modificadas. Foram conduzidos três bioensaios, baseados no teste de germinação. No Ensaio 1, as sementes foram pré-embebidas durante 16 horas em substrato umedecido com solução do herbicida. No Ensaio 2, o substrato foi mantido permanentemente umedecido em solução do herbicida. No Ensaio 3, as sementes foram imersas em solução do herbicida. Os parâmetros de avaliação foram: percentagem e velocidade de germinação, comprimentos da plântula, do hipocótilo e da raiz primária, percentagem de plântulas com raízes secundárias e caracterização morfológica. Concluiu-se que os bioensaios permitem detectar sementes de soja geneticamente modificada, tolerante ao glifosato, no prazo de cinco dias. As metodologias da semeadura em substrato umedecido com solução do herbicida, na concentração de 0,03% da formulação 480 g/L de i.a. e a da pré-embebição das sementes durante 16 horas, com solução contendo 0,4% e 0,6% de glifosato da formulação 480 g/L de i.a. são as recomendadas para utilização em rotina de Laboratório de Análise de Sementes. O glifosato causa anormalidade em plântulas de soja não geneticamente modificada. As plântulas apresentam engrossamento, estrias longitudinais e amarelecimento gradativo do hipocótilo, inibição do desenvolvimento da raiz primária e da emissão de raízes secundárias, sendo o hipocótilo proporcionalmente maior do que a raiz primária.Termos para indexacão: Glycine max, metodologia de detecção, OGM, caracterização de sintomas. BIO ASSAYS FOR DETECTION OF SOYBEAN SEEDS TOLERANT TO GLYPHOSATE HERBICIDE1 ABSTRACT -The objective of this study was to adjust methodology to detect soybean seeds tolerant to glyphosate, to characterize the symptoms, to establish the necessary minimum time capable to allow safe and possible evaluation of being reproduced. Three independent assays were done, based on the germination test. In study 1, seeds were pre-imbibed in a substrate containing herbicide solution for 16 hours. In study 2 the substratum was imbibed with the herbicide. In the study 3, seeds were immersed in solution of the herbicide. The standard germination test, speed of germination, length of seedling root and shoot and percentage of seedling secondary roots were used as evaluation parameters. Evaluations were made in the fourth day, until the final count, on the eighth day. All three methods allowed detection of soybean seeds tolerant to the herbicide glyphosate, in the period of five days. -Seed treatment with growth regulators, especially salicylic acid, is a promising alternative to the seed industry because it is an important inducer of resistance to diseases and pests, as well as acting significantly on quality and seed yield. The objective of this study was to evaluate the performance of rice seed treated with different concentrations of salicylic acid, as well as assess the crop yield and seed quality. The treatments consisted of increasing levels of 0, 50, 100, 150 and 200 mg.L -1 salicylic acid. To this was prepared a stock solution of salicylic acid and the highest concentration by successive dilution in distilled water, the other concentrations were obtained. The physiological quality of seeds produced was treated and evaluated by tests of vigor and germination, and after harvest were evaluated seed yield. It follows that treatment of rice seeds with salicylic acid concentrations up to 130 mg.L -1 at a dose of 2 mL.kg -1 seed does not affect the germination and affects the strength, however provides substantial increases in the yield of seeds. -The objective of this study was to assess performance of wheat seeds after application of three components [fungicide, zinc (Zn), and polymer], separately or with their combinations, as well as assessing seed quality and yield. Thus, the treatments consisted in combinations of fungicide + zinc (Zn) + polymer, totaling six treatments, with four replicates per treatment, as follow: T0 -without application of Zn, fungicide, or polymer (control); T1 -Zn; T2 -Zn + polymer; T3 -polymer; T4 -fungicide; T5 -polymer + fungicide; and T6 -Zn + polymer + fungicide. The performance of the seeds originating from plants whose seeds had been pre-treated was assessed by tests of germination and vigor, in addition to analyzes of growth and yield. It was concluded that wheat seeds treated with zinc, fungicide and polymer are positively influenced until 30 days after emergence. The germination of wheat seeds is positively influenced by the treatment with fungicide, as much when separately applied as in combined application with zinc and polymer. The yield per plant increases when seeds are treated with Zn.Index terms: Triticum aestivum L, germination, vigor.Tratamento de sementes de trigo com zinco, fungicida e polímero: qualidade e rendimento de sementes RESUMO -O objetivo deste estudo foi avaliar o desempenho de sementes de trigo após a aplicação de três componentes [fungicida, zinco (Zn) e polímero] isoladamente, ou com a combinação destes produtos, bem como avaliar a qualidade e o rendimento das sementes produzidas. Deste modo, os tratamentos consistiram nas combinações de fungicida + Zn + polímero, totalizando seis tratamentos, com quatro repetições para cada tratamento: T0 -sem aplicação de Zn, fungicida ou polímero (testemunha); T1 -Zn; T2 -Zn + polímero; T3 -polímero; T4 -fungicida; T5-polímero + fungicida; e T6-Zn + polímero + fungicida. O desempenho das sementes produzidas, oriundas das sementes tratadas, foi avaliado por testes de vigor e germinação, além de análises de crescimento e rendimento. Pelos resultados obtidos conclui-se que sementes de trigo tratadas com Zn, fungicida e polímero são positivamente influenciadas até 30 dias após a emergência. A germinação das sementes é positivamente influenciada pelo tratamento com fungicida, tanto quando este é aplicado isoladamente quanto em aplicação combinada com Zn e polímero.
