Chemotherapy, which is one of the common treatments for osteosarcoma (OS), has
many side effects and in some cases has low effectiveness due to
chemoresistance, hence it is vital to study new therapies for OS. In this
regard, we combined melatonin with cisplatin and evaluate their effect on MG63
OS cells. Since melatonin has anti-cancer properties, we hypothesized that its
combination with cisplatin could increase the effectiveness of cisplatin.
Firstly, MTT assay was used to evaluate the cell viability and cytotoxicity of
cisplatin on MG63 cells and the results showed that melatonin in combination
with cisplatin increases the sensitivity of MG63 cells to cisplatin. In
addition, qRT-PCR results showed that the expressions of miR-181 and P53, CYLD,
CBX7 and BCL2 genes change in MG63 cells after treatment with the combination of
cisplatin and melatonin, so that the expression of P53, CYLD and CBX7 increased
and the expression of BCL2 and miR-181b decreases significantly. Furthermore,
analysis of Annexin V/FITC assay data revealed that the rate of
apoptosis in MG63 OS cell line remarkably promoted after treated with cisplatin
and melatonin combination. As a result, our findings show that melatonin in
combination with cisplatin increases the effectiveness of cisplatin in
osteosarcoma cells and this study provides a new therapeutic approach for
OS.
AIM: To investigate effects of anti-TNF biologic drugs on uveitis severity (comparing visual acuity logMAR levels) in Behçet patients.
METHODS: Three databases PubMed, Scopus, and the Web of Science were searched for qualified papers focusing on the anti-TNF-α factors treatment in Behçet’s disease (BD)-associated uveitis. Studies that were designed pre and post anti-TNF drug treatment, were selected. After determining the search strategy for this study, the relevant data were extracted.
RESULTS: The initial search was performed in the target databases and a total of about 1458 articles were found. Fifteen articles were selected for systematic review and only 12 of them had inclusion criteria for Meta-analysis (with visual acuity data). The mean dose of prednisolone before and after biological treatments was reported in 5 studies (28.56 and 7.56 mg/kg, respectively). Also, the preliminary results indicate a significant reduction in visual acuity logMAR levels (MD=-1.5 IU/L, 95%CI: -2.1, -0.01).
CONCLUSION: Biological drugs significantly reduce the dose of prednisolone and affect visual acuity values.
LncRNAs and miRNAs are the two most important non‐coding RNAs, which have been identified to be associated with cancer progression or prevention. The dysregulation of lncRNAs conducts tumorigenesis and metastasis in different ways. One of the mechanisms is that lncRNAs interact with miRNAs to regulate distinct cellular and genomic processes and cancer progression. LncRNA SNHG7 as an oncogene sponges miRNAs and develops lncRNA‐miRNA‐mRNA axes, leading to the regulation of several signaling pathways such as Wnt/β‐Catenin, PI3K/AKT/mTOR, SIRT1, and Snail‐EMT. Therefore, in this article, after a brief overview of lncRNA SNHG7‐miRNA‐mRNA axes' contribution to cancer development, we will discuss the role of lncRNA SNHG7 in the genes expression and signaling pathways related to cancers development via acting as a ceRNA.
