O leite é um dos alimentos do ponto de vista nutricional mais completo, sendo um importante substrato para microrganismos, tornando-o altamente perecível. Dessa forma, a pasteurização é imprescindível para eliminar patógenos que possam contaminar o leite além de comprometer a segurança alimentar. O controle deste tratamento térmico é baseado na mensuração de duas enzimas naturalmente encontradas no leite cru: fosfatase alcalina e peroxidase. Este trabalho teve como objetivo a comparação dos métodos de detecção da fosfatase alcalina no leite pasteurizado frente a aplicabilidade e sensibilidade dos procedimentos. Foram realizadas quatro etapas de desenvolvimento metodológico, dentre elas a comparação dos métodos de detecção, processos de pasteurização, período de reativação enzimática e limiar de detecção. Utilizou-se amostras de leite cru, pasteurizado e ultrapasteurizado (UHT). Nestas amostras realizaram-se seis diferentes testes para cada uma delas. Para avaliar o comportamento da enzima fosfatase alcalina utilizaram análises colorimétricas qualitativas e quantitativas, além de ressaltar o desempenho dos métodos em variações do binômio tempo e temperatura. As análises colorimétricas qualitativas, apresentam de leituras de interpretações subjetivas com propensão ao erro. Sendo assim, os resultados são indicativos da necessidade da validação de uma metodologia oficial brasileira (MAPA) aplicável na rotina que seja rápida, segura, eficaz e com baixo limiar de detecção quantitativa referente a atividade da fosfatase alcalina residual em leites.
