Bovine astrovirus (BoAstV) is associated with gastroenterical disorders such as diarrhea, particularly in neonates and immunocompromised animals. Its prevalence is >60 % in the first five weeks of the animal's life. The aim of this study was to detect and perform a phylogenetic analysis of BoAstV in Brazilian cattle. A prevalence of 14.3 % of BoAstV in fecal samples from 272 head of cattle from different Brazilian states was detected, and 11 samples were analyzed by nucleotide sequencing. The majority of positive samples were obtained from diarrheic animals (p < 0.01). Phylogenetic analysis revealed that Brazilian samples were grouped in clades along with other BoAstV isolates. There was 74.3 %-96.5 % amino acid sequence similarity between the samples in this study and >74.8 % when compared with reference samples for enteric BoAstV. Our results indicate, for the first time, the occurrence of BoAstV circulation in cattle from different regions of Brazil, prevalently in diarrheic calves.
Nebovirus is a new genus of viruses belonging to the Caliciviridae family recently characterized in cattle, and is associated with gastrointestinal disorders, such as diarrhoea, anorexia and intestinal lesions particularly in calves. The aim of this study was to investigate the prevalence of neboviruses in Brazilian cattle and analyse phylogenetically the virus strains detected. A prevalence of 4·8% of neboviruses in faecal samples from 62 head of cattle from different Brazilian states was detected. All positive animals were aged 96·0% nt (100% aa) sequence identity between the virus sequences in this study and >88·8% nt (>94·4% aa) identity with Newbury1/UK. Our results indicate, for the first time, the occurrence of neboviruses in Brazil as well as in South America, and the first Newbury1-like nebovirus found outside the UK.
A VDAC é a proteína mais abundante na membrana mitocondrial externa. Exerce o controle da atividade desta organela através da regulação da troca de metabólitos e tem função crucial no mecanismo de apoptose. Em nosso caso, os estudos dos complexos protéicos, das interações entre a VDAC e outras proteínas presentes no interior do neurônio que auxiliam na manutenção das funções das organelas e da célula, fazem parte da chamada interactômica. O presente estudo determinou o interactoma do complexo protéico Hexoquinase-VDAC-ANT presente em cérebros murino, bovino e aviar. Nosso objetivo foi identificar se as expressões diferenciadas da VDAC1 e VDAC2 verificadas nos cérebros murino, aviar e bovino, estão associadas a diferenças nos interactomas dessas proteínas. Este estudo revelou que as espécies aviar e bovina apresentaram o maior número de complexos protéicos contendo VDACs (5) quando comparadas com os neurônios de rato (1), o que é indicativo de uma cinética diferencial de montagem ou desmontagem do complexo. Além disso, a VDAC mitocondrial neuronal aviar também interage com mais proteínas em relação à VDAC mitocondrial neuronal bovina, o que é resultado de uma composição de subunidades diferenciada. Tais resultados indicam diferenças significativas quanto ao metabolismo energético e apoptótico no cérebro aviar, bovino e murino, existindo interações diferenciais da VDAC no cérebro aviar.
Objetivo: Verificar a atividade e padrão eletroforético da LDH em resposta ao exercício em esteira em ratos submetidos à fotoestimulação por laser de baixa intensidade (780 ηm). Método: Foram utilizados nesse estudo 54 ratos machos jovens (30dias), Winstar, com peso inicial médio de 112 ± 4,7g, os quais foram divididos em quatro grupos: dois permaneceram em repouso, GRC (grupo repouso controle, n= 9) e GRL (grupo repouso laser, n= 10), sendo o segundo irradiado por laser, e outros dois foram submetidos ao treinamento aeróbio e testes de esforço crescente em esteira (em degraus descontínuos) visando à determinação do limiar de anaerobiose (LA) durante 5 semanas, GEC (grupo exercício controle, n= 16) e GEL (grupo exercício laser, n= 19), sendo o último também irradiado por laser. O laser foi aplicado no quadríceps, glúteo máximo, sóleo e tibial anterior (TA), bilateralmente, imediatamente após cada sessão de treinamento, usando: 3,8J/cm 2 , 15mW, 10s, modo contínuo, durante 5 semanas. Amostras de sóleo, TA e coração foram removidas 48 horas após a última sessão de exercício para análise eletroforética e espectrofotométrica. A estatística foi realizada através do teste ANOVA e post-hoc de TUKEY. O nível de significância foi considerado (p≤ 0,05). Resultados: Foram observadas reduções na atividade da LDH causadas pelo laser (de 19% a 30%), exercício (de 41% a 66%) e as ações combinadas (de 47% a 66%) (p< 0,01). A eletroforese mostrou uma predominância da subunidade B para o sóleo e coração, e uma predominância da subunidade A no TA. Conclusões: Estes dados sugerem uma melhor resposta adaptativa dos animais ao treinamento aeróbio associado à terapia laser, no que se refere à otimização da via oxidativa.
Electrokinetic, thermic, and kinetic properties of products of NADP-dependent isocitrate dehydrogenase (IDHP; EC 1.1.1.42) loci of Astyanax scabripinnis (Pisces, Characidae) collected at three different altitudes (700 m, 1,800 m, and 1,920 m) of Grande Stream at Campos do Jordão, State of São Paulo, Brazil, were analyzed. Two IDHP bidirectionally divergent loci, a single skeletal muscle, the IDHP-A*, and a single liver IDHP-B*, both polymorphic, were detected in the three different altitude populations. The variant allele *128 at the IDHP-A* locus, had its highest frequency detected in the 1,920 m population (0.494). Among the nine variant alleles detected at the IDHP-B* locus (*37, *57, *69, *79, *85, *114, *119, *124, and *140), the *37 and 79 were detected only in 1,800 m population. Chi-square values showed that only the 700 m population is not in Hardy-Weinberg equilibrium for the IDHP-A* locus, while for the IDHP-B* locus, no population is. Homogeneity Chi2 test indicated that the populations are significantly different in their A and B phenotype frequencies. Wright's FST mean value (0.036 and 0.32, IDHP-A* and IDHP-B*, respectively) was 0.178 for the three altitude populations which means that 82% of total genetic diversity was found among individuals of each one of the populations. Stability at environmental temperatures (16 degrees to 21 degrees C), and apparent Km and Vmax values of each A-phenotype skeletal muscle crude extract suggest different roles of A-isoforms during the increased lipogenesis that occurs in fish at low temperatures.
ResumoEm fêmeas bovinas, a liberação de prostaglandina F 2a (PGF 2a ) é induzida in vivo pelo estradiol (E 2 ). Acredita-se que o E 2 estimule a síntese de proteínas essenciais na produção de PGF 2a . Objetivou-se avaliar o efeito do E 2 no incremento da concentração de proteínas totais e na modificação da composição proteica em explantes endometriais de fêmeas bovinas tratadas com E 2 no 17º dia do ciclo estral. Novilhas cruzadas foram tratadas no 17° dia do ciclo estral, via intravenosa, com 0 mg (Grupo Controle; n = 6) ou 3 mg de E 2 (Grupo E 2 ; n = 6) e abatidas duas horas após. Explantes endometriais foram isolados, submetidos à extração de proteínas totais, quantificados e avaliados por Eletroforese Unidimensional em gel de poliacrilamida 10% SDS-PAGE. A concentração de proteínas totais não diferiu entre os grupos, 6296,10 + 439,90 µg/mL para o Grupo Controle e 8426,56 + 1156,00 µg/mL para o Grupo E 2 (p = 0,1158). Não houve diferença significativa (p > 0,05) no perfil proteico dos explantes endometriais em géis corados com Coomasie Blue. Em géis corados com Nitrato de Prata verificou-se no Grupo E 2 maior porcentagem relativa das bandas referentes ao peso molecular de 75 a 76 kDa (8,40% vs. 4,89%; no Grupo E 2 e Controle respectivamente; p < 0,05) e 108 a 110 kDa (6,85% vs. 3,84%; no Grupo E 2 e Controle respectivamente; p < 0,05). Observou-se no Grupo E 2 menor porcentagem relativa da banda referente ao peso molecular de 90 kDa (5,78% vs. 9,83%; no Grupo E 2 e Controle respectivamente; p < 0,05). Conclui-se que o E 2 não incrementa a concentração de proteínas no endométrio, entretanto, altera a composição proteica nos explantes endometriais, indicando que o E 2 altera a expressão de proteínas específicas.Palavras-chave: Bovino. This study aimed to evaluate the effect of E 2 in increasing the concentration of total protein and modifying the protein composition of endometrial explants from bovine females treated with E 2 at the 17 th day of estrous cycle. Crossbred heifers were treated at 17 th day of estrous cycle intravenously with 0 mg (Control Group; n = 6) or 3 mg of E 2 (E 2 Group; n = 6) and killed two hours after. Endometrial explants were isolated, subjected to extraction of total protein, quantified and were analyzed by onedimensional electrophoresis on polyacrylamide gel 10% SDS-PAGE. The concentration of total protein did not differ between groups, 6296.10 + 439.90 µg/mL for the Control Group and 8426.56 + 1156.00 µg/mL for E 2 Group (p = 0.1158).
