The application of a recently described PCR test for the detection of Haemophilus paragallinarum in China is described. The test was used to examine a total of 127 chickens sourced from a challenge trial (38 chickens), a respiratory disease-free experimental chicken farm (50 chickens) and eight farms with suspect infectious coryza (IC) outbreaks (39 chickens). The PCR results were compared with traditional culture. The PCR detected 14/14 infected birds in the challenge trial as compared with 13/14 for culture. The 50 chickens from the disease-free experimental farm were all negative by both PCR and culture. PCR yielded 15/39 birds and 6/8 commercial farms positive as compared with 8/39 birds and 4/8 farms positive by culture. All farms positive by PCR had chickens showing the typical clinical signs of IC, indicating that culture failed to confirm coryza on two farms that had the typical clinical signs of the disease. Although chickens on two commercial farms were thought initially to be suffering from coryza, detailed clinical examination yielded no birds with typical clinical signs. The 12 chickens examined from these two farms were negative by both PCR and culture. The results suggest that the PCR test for H. paragallinarum is a suitable alternative to culture even under the typical field and laboratory conditions that operate in China.
In this paper, we give a lower bound of Bergman kernels for a sequence of almost Kähler-Einstein Fano manifolds, or more general, a sequence of Fano manifolds with almost Kähler-Ricci solitons. This generalizes a result by Donaldson-Sun, Tian for Kähler-Einstein manifolds sequence with positive scalar curvature. As an application of our result, we prove that the Gromov-Hausdorff limit of sequence is homomorphic to a log terminal Q-Fano variety which admits a Kähler-Ricci soliton on its smooth part.2000 Mathematics Subject Classification. The present study aimed at examining the curative effect of modified posterior operation on treatment of Kümmell's disease.About 30 patients of Kümmell's disease with complete image and clinical data treated during June 2004 to July 2013 were conducted with anterior and posterior approaches, respectively. Kyphotic Cobb angle, fractured vertebra wedge angle, and the anterior and posterior heights of fractured vertebra were all measured through x-ray before and after operation, and the pain visual analog scale (VAS) was determined for evaluating the effect of operations. The injury and restoration of neurological function were assessed using Frankel classification.Patients in group A were treated with anterior operation, whereas group B was posterior operation. Postoperatively, VAS score, kyphotic Cobb angle, anterior vertebra height, and pathologic vertebra wedge angle were all significantly improved in patients with Kümmell's disease receiving modified posterior operation (group B). Similar results were also observed in patients with anterior operation. The improvement of VAS and correction rate of kyphotic Cobb angle indicated equivalent effects of posterior and anterior operations. Meanwhile, alleviated neurological function damage was observed in 2 groups. Relevant factor analysis illustrated that there was no significant correlation of the severity and improvement rate of pain symptoms with age, medical history, anterior and posterior vertebra heights, kyphotic Cobb angle, and pathological vertebra wedge angle.Compared with traditional anterior approach, modified posterior operation, adopting transpedicular vertebral body grafting combined with vertebral pedicle screw fixation, could produce equivalent effects on kyphosis correction, pain relief, and improvement of neurological function in patients with Kümmell's disease.
Strategic control of mitochondrial movements and cellular distribution is essential for correct cell function and survival. However, despite being a vital process, mitochondrial movement in plant cells is a poorly documented phenomenon. To investigate the roles of actin filaments and microtubules on mitochondrial movements, Picea wilsonii pollen tubes were treated with two microtubule-disrupting drugs, two actin-disrupting drugs and a myosin inhibitor. Following these treatments, mitochondrial movements were characterized by multiangle evanescent wave microscopy and laser-scanning confocal microscopy. The results showed that individual mitochondria underwent three classes of linear movement: high-speed movement (instantaneous velocities >5.0 microm/s), low-speed movement (instantaneous velocities <5.0 microm/s) and variable-speed movement (instantaneous velocities ranging from 0.16 to 10.35 microm/s). 10 nM latrunculin B induced fragmentation of actin filaments and completely inhibited mitochondrial vectorial movement. Jasplakinolide treatment induced a 28% reduction in chondriome motility, and dramatically inhibition of high-speed and variable-speed movements. Treatment with 2,3-butanedione 2-monoxime caused a 61% reduction of chondriome motility, and the complete inhibition of high-speed and low-speed movements. In contrast to actin-disrupting drugs, microtubule-disrupting drugs caused mild effects on mitochondrial movement. Taxol increased the speed of mitochondrial movement in cortical cytoplasm. Oryzalin induced curved mitochondrial trajectories with similar velocities as in the control pollen tubes. These results suggest that mitochondrial movement at low speeds in pollen tubes is driven by myosin, while high-speed and variable-speed movements are powered both by actin filament dynamics and myosin. In addition, microtubule dynamics has profound effects on mitochondrial velocity, trajectory and positioning via its role in directing the arrangement of actin filaments.
The biochemical and serological properties of 11 isolates of Haemophilus paragallinarum from outbreaks of infectious coryza in the People's Republic of China are described. All 11 isolates had the typical biochemical properties of H. paragallinarum, and all belonged to Page serovar A.
