Short-chain fatty acids and their corresponding acyl-CoAs sit at the crossroads of metabolic pathways and play important roles in diverse cellular processes. They are also precursors for protein post-translational lysine acylation modifications. A noteworthy example is the newly identified lysine 2-hydroxyisobutyrylation (K) that is derived from 2-hydroxyisobutyrate and 2-hydroxyisobutyryl-CoA. Histone K has been shown to be associated with active gene expression in spermatogenic cells. However, the key elements that regulate this post-translational lysine acylation pathway remain unknown. This has hindered characterization of the mechanisms by which this modification exerts its biological functions. Here we show that Esa1p in budding yeast and its homologue Tip60 in human could add K to substrate proteins both in vitro and in vivo. In addition, we have identified HDAC2 and HDAC3 as the major enzymes to remove K. Moreover, we report the first global profiling of K proteome in mammalian cells, identifying 6 548 K sites on 1 725 substrate proteins. Our study has thus discovered both the "writers" and "erasers" for histone K marks, and major K protein substrates. These results not only illustrate the landscape of this new lysine acylation pathway, but also open new avenues for studying diverse functions of cellular metabolites associated with this pathway.
Abstract:A wide area quantum key distribution (QKD) network deployed on communication infrastructures provided by China Mobile Ltd. is demonstrated. Three cities and two metropolitan area QKD networks were linked up to form the Hefei-Chaohu-Wuhu wide area QKD network with over 150 kilometers coverage area, in which Hefei metropolitan area QKD network was a typical full-mesh core network to offer all-to-all interconnections, and Wuhu metropolitan area QKD network was a representative quantum access network with point-to-multipoint configuration. The whole wide area QKD network ran for more than 5000 hours, from 21 December 2011 to 19 July 2012, and part of the network stopped until last December. To adapt to the complex and volatile field environment, the Faraday-Michelson QKD system with several stability measures was adopted when we designed QKD devices. It is well known that the unconditional security of quantum-key distribution (QKD) can be guaranteed by quantum mechanics. However, practical QKD systems have some imperfections, which can be controlled by the eavesdropper to attack the secret key. With current experimental technology, a realistic beam splitter, made by fused biconical technology, has a wavelength-dependent property. Based on this fatal security loophole, we propose a wavelength-dependent attacking protocol, which can be applied to all practical QKD systems with passive state modulation. Moreover, we experimentally attack a practical polarization encoding QKD system to obtain all the secret key information at the cost of only increasing the quantum bit error rate from 1.3 to 1.4%.
ZnS coated on N,S co-doped carbon (ZnS/NSC) composite has been prepared utilizing zinc pyrithione (CHNOSZn) as raw material via calcination. Through activation using NaCO salt, ZnS nanoparticles encapsulated in NSC (denoted as A-ZnS/NSC) with mixed-crystal structure has also been obtained, which reveals much larger specific surface area and more bridges between ZnS and NSC. Based on the existence of bridges (C-S-Zn and S-O-Zn bonds) and the modification of carbon from N,S co-doping, the A-ZnS/NSC composite as an anode for sodium-ion batteries (SIBs) displays significantly enhanced electrochemical performances with a high reversible specific capacity of 516.6 mA h g (at 100 mA g), outstanding cycling stability (96.9% capacity retention after 100 cycles at 100 mA g), and high rate behavior (364.9 mA h g even at 800 mA g).
We propose a wavelength-saving topology of a quantum key distribution (QKD) network based on passive optical elements, and we report on the field test of this network on commercial telecom optical fiber at the frequency of 20 MHz. In this network, five nodes are supported with two wavelengths, and every two nodes can share secure keys directly at the same time. We also characterized the insertion loss and cross talk effects on the point-to-point QKD system after introducing this QKD network.
