In vitro screening of the crude extracts of some Brazilian coastal seaweeds for cytotoxic activity against a cultured human melanoma cancer cell line using the sulphorhodamine B assay was performed. The crude dichloromethane:chloroform extract of Stypopodium zonale showed good cytotoxic activity against the C32 cell line. The crude acetone extract and aqueous phase of Lobophora variegata did not show any activity, but semi-purified fractions XAD LOB I and II could inhibit the growth of melanoma cells. The crude acetone extract of Caulerpa racemosa showed some cytotoxicity, but caulerpin isolated from this extract did not show any such activity. The crude acetone extract of Spatoglossum schroederi was not able to inhibit the growth of C32 melanoma cells.

RESUMO:Os radicais livres e outros derivados ativos do oxigênio são inevitavelmente coproduzidos em algumas reações biológicas e exercem papel fi siológico importante. No entanto, essas espécies reativas têm sido descritas como fatores que participam diretamente dos mecanismos fi siopatológicos relacionados com a continuidade e as complicações de diversos estados patológicos como a aterosclerose, a diabetes, o câncer, a artrite reumatóide, entre outros. Dessa forma, a terapia antioxidante preveniria o desenvolvimento e a progressão dessas complicações. As algas marinhas representam uma importante fonte de substâncias antioxidantes naturais, uma vez que têm sistemas de defesas antioxidantes bem desenvolvidos. ABSTRACT: "Natural products from marine seaweeds and their antioxidant potential"Free radicals and other reactive species of oxygen are co-produced in some biological reactions and they play important physiological role, and nevertheless they are reported as factors that take straight part in the pathophysiologic mechanism associated with continuity and complications of the several pathological process such as arteriosclerosis, diabetes mellitus, cancer, and arthritis, among others. In this way, the antioxidant therapy should prevent the development and progress of these complications. Seaweeds can be valuable source of natural antioxidant compounds since they have a well-developed antioxidant defense system. The present work is a compilation of the antioxidant activities of marine natural products and benthonic marine seaweeds crude extracts researches.

Highlights Unspecific amplifications were found in 56.4% (495 reactions) of negative samples for SARS-CoV-2. In silico analysis of N2 primers-probe and gel electrophoresis showed dimer formation. Optimization of RT-qPCR conditions reduced the dimerization events. Conditions must be adjusted to avoid extensive test repetition and waste of resources.

BackgroundSeveral species of the genus Aspidosperma (Apocynaceae) are used for the treatment of human malaria in Brazil and other meso- and South American countries.MethodsEthanol extract from Aspidosperma parvifolium trunk bark was submitted to acid–base extractions leading to alkaloid and neutral fractions. The alkaloid fraction was chromatographed over a silica gel column. Ethanol extract, fractions and uleine were analysed by HPLC–DAD, UPLC-ESI–MS/MS and HPLC-ESI-MicroTOF-MS. The anti-malarial activity was assayed against resistant and sensitive chloroquine Plasmodium falciparum strains by microscopic, [3H]-hypoxanthine incorporation and HRPII techniques. Cytotoxicity (CC50) was evaluated against Vero and HepG2 cell lines by the MTT technique; selectivity indexes (SI = CC50/IC50) were calculated.ResultsThe major peak in the HPLC–DAD chromatograms of the ethanol extract, alkaloid and neutral fractions suggested the presence of uleine that was isolated from the alkaloid fraction by column chromatography and was characterized by spectroscopic methods. A total of 15 alkaloids, besides uleine, were identified in the alkaloid fraction by UPLC-DAD-ESI–MS/MS and HPLC-ESI-MicroTOF-MS. The ethanol extract from Aspidosperma parvifolium and the neutral fraction were moderately active against P. falciparum strains. The alkaloid fraction and uleine disclosed high anti-malarial activity against chloroquine-resistant P. falciparum strain (IC50 < 1 µg/mL). The ethanol extract, neutral fraction and uleine showed low cytotoxicity against Vero and HepG2 cell lines (CC50 > 300 µg/mL). The alkaloid fraction showed moderate cytotoxicity to HepG2 cell line (CC50 = 74.4 µg/mL). High SI values (>10) were determined for all samples.ConclusionEthanol extract from Aspidosperma parvifolium trunk bark afforded uleine that is the major constituent of the alkaloid fraction and disclosed a good in vitro anti-malarial activity. Moreover, 15 other indole alkaloids have been identified along with uleine.

Extracts of the Brazilian brown seaweeds Dictyopteris justii and Spatoglossum schroederi showed the presence of a cytotoxic mixture of 24R-and 24S-epimers of 24-hydroperoxy-24-vinylcholesterol (1). The derivation of 1 from fucosterol is proposed. Laboratory experiments assessed the anti-herbivory properties against the crab, Pachygrapsus transversus, of the crude extract and the epimers of 1 found in D. justii. The crude extract did not inhibit feeding, but the mixture of 1 epimers had a significant inhibitory effect. We propose that these unusual sterol compounds cannot be easily converted to assimilable compounds by this crustacean, thus acting as a defensive strategy for the seaweed against herbivory.

RESUMO: "Espécies de Bromeliaceae brasileiras: isolamento de derivados arilpropanoídicos e potencial antiradicalar". Um total de vinte extratos de seis espécies de Bromeliaceae brasileiras foram avaliadas quanto a atividade antioxidante, usando-se o método colorimétrico de redução do radical DPPH. De maneira geral, os extratos polares dos rizomas das espécies de Bromeliaceae testadas revelaram melhor perfil antioxidante do que os extratos das folhas e dos frutos das mesmas espécies. Os melhores resultados foram encontrados para os rizomas de Vriesea procera (Mart. ex Schult.f.) Wittm. e Neoregelia cruenta (Graham) L.B. Sm. O extrato bruto metanólico das folhas de Ananas bracteatus (Lindl.) Schult. & Schult. f. apresentou uma atividade significativa, em relação aos extratos de folhas testados, e o processo de purificação desse extrato resultou na identificação de quatro metabólitos: ferulato de 2-glicerila, p-cumarato de 2-glicerila, 5,7,4'-triidroxi-3,3',5'-trimetoxiflavona e 3-O-β-D-glicopiranosil sitosterol.Unitermos: Bromeliaceae, antioxidante, ferulato de 2-glicerila, p-cumarato de 2-glicerila, 5,7,4'-triidroxi-3,3',5'-trimetoxiflavona. ABSTRACT:A total of twenty different extracts from six Brazilian Bromeliaceae species was screened for antioxidant activity by assessment of their capacity to scavenge the DPPH radical. In a general way, the polar rhizome extracts from Bromeliaceae representatives showed better antioxidant results than the extracts from leaves and fruits of the same species. The best results were found for the rhizome extracts of Vriesea procera (Mart. ex Schult.f.) Wittm. and Neoregelia cruenta (Graham) L.B. Sm. Crude methanol extract of Ananas bracteatus (Lindl.) Schult. & Schult. f. leaf had a significant antiradical activity among the leaves extracts assessed its purification afforded four metabolites: 2-O-feruloyl glyceride, 2-O-p-coumaroyl glyceride, 5,7,4'-trihydroxy-3,3',5'-trimethoxyflavone and 3-O-β-D-glucopyranosyl sitosterol.
