Abstract-The aim of the present study was to determine the modeling of muscle fibers in rats submitted to different exercise protocols. Fifty-five Wistar rats were submitted divided into four different groups: Control group (CG; N=16); endurance training group (ETG; N=13), strength training group (STG; N=13) and concurrent training group (CTG; N=13). The intensity of endurance training was determined by the critical workload. Statistical analysis involved the Kruskal-Wallis test for multiple comparisons, followed by Dunn's post test (p<0.05). All animals submitted to training exhibited an increase in the cross-sectional area of the muscle fibers. The largest increase (p>0.05) occurred in the STG and CTG at both four (mean:2952,95 ± 878,39 mean:2988,84 ± 822,58) and eight weeks respectively (mean:3020,26 ± 800.91; mean:3104,91 ± 817,87). The findings demonstrate similar results obtained with strength training and concurrent training, with a greater increase in muscle fiber area in both groups in comparison to the control group and group submitted to endurance training.Keywords: skeletal muscle, soleus muscle, concurrent training, aerobic exercise, anaerobic exercise Resumo-"Efeito do treinamento concorrente nas fibras musculares de ratos Wistar." O objetivo deste estudo foi observar a modelação das fibras musculares de ratos submetidos a diferentes protocolos de treinamento físico. Foram utilizados 55 animais da raça Wistar, dos quais foram submetidos a quatro diferentes tratamentos, sendo estes: o grupo controle (CTLE), treinamento aeróbio (TAE), treinamento de força (TAN) e treinamento concorrente (TCc). INTRODUÇÃO: Os fumantes sofrem redução da massa muscular e da resistência à fadiga e possivelmente a prática de atividade física contribua positivamente neste quadro. OBJETIVO: Este estudo teve por objetivo analisar as adaptações da musculatura esquelética frente à interação da prática de atividade física e exposição à fumaça de cigarro. MÉTODOS: 32 ratos machos foram divididos em grupos expostos à fumaça de cigarro, exercitados (G1) e sedentários (G3), e não expostos à fumaça, exercitados (G2) e sedentários (G4). A exposição à fumaça foi realizada mediante combustão de 10 cigarros em uma câmara de inalação durante 30 minutos, duas vezes ao dia, cinco dias por semana, durante 15 dias; o exercício aeróbio foi composto por sessões diárias de 60 minutos de caminhada em esteira, cinco dias por semana. Após o sacrifício, o músculo sóleo foi coletado e seus cortes foram corados pela técnica de HE e submetidos ao método histoquímico NADH-TR. RESULTADOS: O grupo G1 apresentou maior quantidade de alterações musculares, bem como ausência de atividade enzimática, o mesmo ocorrendo no G3, porém com menor intensidade; no G2 foi observado padrão normal para fibras exercitadas, estando as fibras do G4 preservadas. Quanto à morfometria, houve diferença significante para o fator exercício (p = 0,007), enquanto não foram observadas diferenças significantes para o fator exposição à fumaça (p = 0,668) e para a interação exposição à fumaça e exercício (p = 0,077). CONCLUSÃO: A interação entre inalação de fumaça de cigarro e exercício, realizada durante 15 dias em ratos machos Wistar adultos, acentuou as alterações histológicas do músculo sóleo, levando a uma alteração da atividade enzimática e acarretando em aumento do diâmetro das fibras musculares
ObjectivesTo describe the effects of immobilization, free remobilization and remobilization by physical exercise about mechanical properties of skeletal muscle of rats of two age groups.Methods56 Wistar rats divided into two groups according to age, an adult group (five months) and an older group (15 months). These groups were subdivided in: control, immobilized, free remobilized and remobilized by physical exercise. The pelvic limb of rats was immobilized for seven days. The exercise protocol consisted of five swimming sessions, once per day and 25 minutes per session. The gastrocnemius muscle was subjected to tensile tests, and evaluated the properties: load at the maximum limit, stretching at the maximum limit and stiffness.ResultsThe immobilization reduced the values of load at the maximum limit and the remobilization protocols were not sufficient to restore control levels in adult group and older rats. The stretching at the maximum limit differs only in the older group.ConclusionsThe immobilization reduces the muscle's ability to bear loads and exercise protocol tends to restore the default at control values in adult and older rats. The age factor only interfered in the stretching at the maximum limit, inducing a reduction of this property in the post-immobilization. Level of Evidence II, Investigating the Results of Treatment.
Influência do tabagismo passivo associado ao exercício físico realizado por ratas durante prenhez e lactação, sobre o desenvolvimento dos filhotes Artigo originalResumo OBJETIVOS: o objetivo deste estudo foi avaliar mortalidade, peso e comprimento corporal, e o músculo gastrocnêmio dos filhotes de ratas prenhes submetidas ao programa de natação associado ao tabagismo passivo. MÉTODOS: vinte e quatro ratas foram divididas em quatro grupos: GF (exposto à fumaça de cigarro), GC (Controle), GFN (submetido ao programa de natação e exposto à fumaça de cigarro) e GN (submetido ao programa de natação). A mortalidade e a aferição do peso e comprimento dos filhotes foram realizadas em quatro momentos. O músculo gastrocnêmio dos filhotes foi obtido para avaliação do desenvolvimento muscular. RESULTADOS: a média do nascimento de filhotes foi menor para os grupos GF (10,2) e GFN (10,3) e maior para o GN (12,8). No nascimento, somente o GFN apresentou medidas de peso (p=0,016) e comprimento (p=0,02) significantemente inferiores ao Controle, já na lactação foram os grupos expostos à fumaça de cigarro. O grupo GFN apresentou retardo no desenvolvimento muscular em relação ao GC (p=0,03). CONCLUSÃO: O tabagismo passivo durante a prenhez e lactação exerceu influência negativa sobre o número, peso e comprimento corporal dos filhotes do nascimento ao desmame e sobre o desenvolvimento muscular; o programa de natação influenciou positivamente tais variáveis no momento do nascimento, entretanto, não possibilitou os mesmos benefícios durante a lactação; e a associação destes influenciou negativamente tais medidas. AbstractPURPOSE: the purpose of this study was to evaluate mortality, weight and body length, and the gastrocnemius muscle of the offspring of pregnant rats submitted to a swimming program associated with second-hand smoke. METHODS: twenty-four rats were divided into four groups: GF (exposed to cigarette smoke), GC (control), GFN (submitted to the swimming program and exposed to cigarette smoke), and GN (submitted to the swimming program). The mortality, weight and length of the offspring were measured at four time points. The gastrocnemius muscle of the pups was obtained for evaluation of muscle development. RESULTS: the average number of offspring was lower for GF (10.2) and GFN (10.3) and higher for GN (12.8). At birth, only GFN showed significantly lower weight (p=0.016) and length (p=0.02), whereas during lactation the groups exposed to cigarette smoke showed significantly lower weight. GFN had delayed muscle development compared to GC (p=0.03). CONCLUSIONS: Passive smoking during pregnancy and lactation negatively influenced number, weight and body length of offspring from birth to weaning and muscle development, and the swimming program positively influenced these variables at birth, although it did not provide the same benefits during lactation; and their association negatively affected these measures.
Introduction: Immobilization is a treatment technique often used to reduce pain and prevent worsening of the injury. However, it promotes harmful effects on musculoskeletal tissue, resulting in a marked loss of muscle function, which may be aggravated in the elderly. Physical exercise is an important intervention to mitigate these harmful effects. Objective: To analyze possible morphometric changes in the gastrocnemius muscle of rats after immobilization and remobilization with physical exercise. Methods: Fifty-six rats were divided into adult (A) and aged (E) groups and subdivided into adult and aged control (AC and EC), immobilized (AI and EI), free remobilized (AIF and EIF), and remobilized through physical exercise (AIE and EIE). The hind limbs were immobilized with the gastrocnemius muscle in a shortened position for a period of seven days, except for the control group. The exercise protocol consisted of five swimming sessions, once per day (25 minutes/session). The animals were euthanized by administration of an overdose of ketamine hydrochloride plus xylazine hydrochloride, followed by sample collection and preparation of hematoxylin and eosin slides. Measurements of the smallest diameter of 120 muscle fibers of each animal were taken with software NIS-Elements D3.0 - SP7 - Nikon® instruments Inc., NY, USA. Results: There was a significant reduction in the mean fiber diameter in the AI (38.43 µm ± 4.20; p=0.01) and AIF (36.97 µm ± 3.41; p<0.01) groups compared to AC (45.39 µm ± 3.41) and in the EI (42.26 µm ± 4.39; p<0.01), EIF (36.00 µm ± 4.15; p<0.01), and EIE (41.86 µm ± 4.95; p<0.01) groups compared to the EC (51.37 µm ± 3.86) group. The data showed that exercise was able to restore muscle trophism in the adult groups; however, none of the protocols has succeeded in aged rats. Conclusion: Immobilization in the shortened position induced muscle atrophy and physical exercise was effective in restoring muscular trophism only in adult animals. High caloric diet is regarded as a risk factor for developing cardiovascular diseases, is considered a global public health problem. The aim of this study was to compare the biometric parameters of rats fed with two diet types. Thus, 24 male Wistar rats (90 days) with intake of food and water ad libitum were divided into G1: receiving normocaloric diet, and G2: high caloric diet. Food intake was measured daily. After 18 weeks the rats were euthanized, and previous body mass, naso-anal length and Lee index were measured. Data were analyzed by descriptive statistics, followed by Kolmogorov-Smirnov normality test and ANOVA – One Way, using Tukey post hoc test, assuming 5% of significance. G2 had greater weight gain and therefore higher Lee index compared to G1; however, it was observed a decrease of food intake in G2 compared to G1
RESUMOIntrodução: A imobilização articular é uma técnica de tratamento frequentemente utilizada na ortopedia e, associada ao processo de senescência, promove alterações tanto na estrutura quanto na síntese e biomecânica do tecido cartilaginoso. Objetivo: Descrever os efeitos da imobilização, da remobilização livre e por meio de exercício físico sobre a cartilagem articular de ratos situados em duas faixas etárias. Métodos: Trinta e nove ratos Wistar divididos em dois grupos etários, um grupo adulto (cinco meses de idade) e um idoso (15 meses de idade), subdivididos em: controle, imobilizado, remobilizado livre e remobilizado por meio de exercício físico. Os membros posteriores dos ratos foram imobilizados por sete dias. O protocolo de exercícios foi composto por cinco sessões diárias de natação, de 25 minutos cada. A cartilagem articular do quadril foi submetida à análise termogravimétrica, tendo sido avaliado seu conteúdo de água. Resultados: Nos animais adultos a imobilização reduziu a quantidade de água presente no tecido cartilaginoso, e os protocolos de remobilização foram eficazes para restabelecer a condição inicial do tecido. Nos animais idosos não houve diferença significativa entre os grupos, porém o grupo idoso controle obteve resultado significantemente menor que o grupo adulto controle (X: 13,10 ± 5,24 vs 10,70 ± 1,95). Conclusões: A imobilização e o processo de senescência induzem a diminuição do conteúdo de água da cartilagem articular e os protocolos de remobilização foram eficientes para restabelecer esta propriedade apenas nos animais adultos.
