Isolation and antibiotic susceptibility of microorganisms from dogs with otitis externa Öz Amaç: Bu çalışmada otitis eksternalı köpeklerden alınan kulak sıvap örneklerinde mikroorganizmaların izolasyonu ve izole edilen bakterilerin antibiyotik duyarlılıklarının belirlenmesi amaçlandı. Gereç ve Yöntem: Bu çalışmada, otitis eksterna teşhisi konulmuş 41 köpekten 58 kulak sıvap örneği toplandı. Örneklerden mikroorganizmalar konvansiyonel bakteriyolojik yöntemlerle izole ve identifiye edildi. İzole edilen bakterilerin antibiyotik duyarlılıkları agar disk difüzyon testi ile belirlendi. Bulgular: Örneklerin 52 (%89.66)'sinden 64 mikroorganizma izolasyonu yapılırken, 6 (10.34)'sından patojen etken izole edilemedi. Bu mikroorganizmalar arasında en yüksek oranda Staphylococcus aureus (%31.25) izole edildi. Ayrıca, sırasıyla koagülaz negatif stafilokok (KNS) (%14.06), Streptococcus sp. (%12.5), Corynebacterium sp. (%6.25), Proteus sp. (%4.68), Pseudomonas aeruginosa (%3.13) ve Candida sp. (%28.13) izole edildi. S. aureus, Streptococcus sp., Corynebacterium sp. ve KNS izolatlarının tümü ve Proteus sp. izolatlarının %66.67'si amoksisilin-klavulanik asite duyarlı bulundu. Bu bakterilerin enrofloksasin, amoksisilin, oksitetrasiklin, gentamisin, trimetoprimsulfametoksazol ve eritromisine duyarlılıkları sırasıyla %65.22, %52.17, %47.83, %43.48, %30.43 olarak saptandı. P. aeruginosa izolatlarının tümü enrofloksasin ve gentamisine, %50'si de oksitetrasikline duyarlı bulundu. Öneri: Bakteriyel izolatların çalışmada kullanılan antibiyotiklere duyarlılıklarındaki farklılıklar, otitis eksternalı köpeklerin tedavisi için mutlaka antibiyotik duyarlılık testlerinin yapılması gerektiğini göstermektedir. Ayrıca, çalışmada Candida sp.'nin yüksek oranda izole edilmiş olması, köpeklerin otitis eksterna olgularında mayaların da gözardı edilmemesi gerektiğini ortaya koymaktadır.

Memelilerde katelisidinler ve defensinler olmak üzere iki önemli antimikrobiyal peptid grubu vardır. Bu iki grup arasında da en yaygın peptidler, beta-tabakalı, üç intramoleküler disülfit bağı içeren defensinlerdir. Bu derlemede hayvanlarda sentezlenen defensinlerin yapısal özellikleri, antimikrobiyal etkinlikleri, bağışıklıktaki rolleri ile gelecekte kullanım alanları üzerine bilgiler sunuldu.
Streptococcus canis is an important opportunistic pathogen that infects animals and humans. It causes various infections in dogs and cats such as arthritis, skin infections, otitis externa, septicemia, facial necrosis, and streptococcal toxic shock syndrome. It can also lead to human infections. There are still very few specific studies on the identification and antimicrobial resistance of this pathogen. In this study, the phenotypic and biochemical properties of 12 S. canis isolates obtained from the vaginal discharge of 11 dogs and the open wound of one dog with facial necrosis, and their susceptibility to some antibiotics was investigated. The phenotypic and biochemical properties of 12 isolates, such as esculin hydrolysis, hippurate hydrolysis, synergistic CAMP-like hemolytic activity, growth on Brain Heart-Infusion Agar (BHIA) containing 6.5% NaCl, and carbohydrate fermentation were investigated by conventional bacteriological methods. It was determined that all isolates were in the G group according to Lancefield serogrouping. The presence of the species-specific 16S-23S rRNA gene intergenic spacer region (ISR) and sodA int gene was also detected in all the isolates using the polymerase chain reaction method. All the isolates were detected to be susceptible to penicillin G, cefoperazone, ampicillin, and amoxicillin-clavulanic acid, and resistant to gentamicin and neomycin.
Paratuberculosis (Johne's disease) is a disease of domestic and wild ruminants caused by Mycobacterium avium subspecies paratuberculosis (Map). It is not yet fully understood how the agent, which has a broad host range and has developed various strategies to survive in inappropriate conditions, can survive. In this review; the early host defenses developing in Map infection, the strategies employed by the pathogen to avert these responses and how persistent infection has occured, in macrophages, has been explained.Öz: : Paratüberküloz (Johne hastalığı), Mycobacterium avium subspecies paratuberculosis (Map) tarafından oluşturulan evcil ve yabani ruminantların bir hastalığıdır. Geniş bir konakçı dağılımına sahip olan ve uygun olmayan koşullarda hayatta kalmak için çeşitli stratejiler geliştirmiş olan etkenin nasıl canlı kalabildiği henüz tam olarak anlaşılamamıştır. Bu derlemede; Map enfeksiyonunda meydana gelen erken konakçı yanıtı, bu yanıtı engellemek için patojenin kullandığı stratejiler ve makrofajlarda nasıl kalıcı enfeksiyonun oluştuğu açıklandı.
Laboratuvarına septik artrit şüphesi ile gönderilen 153 buzağıya ait sinoviyal sıvı örnekleri bakteriyolojik olarak incelendi. Sinoviyal sıvı örneklerinden kanlı agar, MacConkey agar ve mikoplazma selektif agara ekimler yapıldı ve standart bakteriyolojik metotlarla izolasyon ve identifikasyonları gerçekleştirildi. Sinoviyal sıvı örneklerinin 100'ünde bakteriyel etken izole edilirken, 53 örnekte izolasyon yapılamadı. Örneklerin 30'undan (% 19,6) Mycoplasma sp., 19'undan (% 12,4) Trueperella pyogenes ve 17'sinden (% 11,1) ise Streptococcus sp. tek başına en fazla izole edilen etken oldu. T. pyogenes, Mycoplasma ve Streptococcus türlerinin eşlik ettiği 16 örnekte de miks enfeksiyon saptandı. Diğer patojenler olarak Staphylococcus aureus, koagülaz negatif stafilokok, Escherichia coli, Pasteurella multocida, Mannheimia haemolytica, Proteus sp., Corynebacterium sp., Salmonella sp., Klebsiella sp.ve Citrobacter sp. izole edildi. Sonuç olarak, Burdur ilinde buzağı artritlerinden sorumlu en yaygın etkenlerin Mycoplasma sp., T. pyogenes ve Streptococcus sp. olduğu ve tedavi öncesi sinoviyal sıvılarının bakteriyolojik incelenmesinin faydalı olacağı kanısına varıldı.
Mycobacterium avium subspecies paratuberculosis, hayvan sağlığını etkileyen ve ciddi ekonomik kayıplara sebep olan önemli bir patojendir. Etkenin mevcut immun sistemden kaçma stratejileri nedeniyle bugüne kadar paratüberküloza karşı etkili bir aşı geliştirilememiş ve uygun bir koruma ve kontrol stratejisi belirlenememiştir. M. avium subsp. paratuberculosis'e karşı doğru kontrol stratejilerinin geliştirilebilmesi için konakçı-patojen etkileşimini düzenleyen hücresel ve moleküler mekanizmalar ile hastalığın patolojik temelinin detaylı olarak anlaşılması gerekmektedir. Bu derlemede; M. avium subsp. paratuberculosis ile konakçı ilişkisi ve konakçıda gelişen immunolojik reaksiyonlar üzerine kapsamlı bilgiler sunuldu.
This study was aimed to search and characterize the AmpC and/or ESBL genes of Escherichia coli, Klebsiella pneumoniae and Pseudomonas aeruginosa isolated from clinical cases of local livestock and companion animals between 2017 and 2019. A total of eight ceftiofur-resistant E. coli (n= 7) and ceftiofur-resistant K. pneumoniae (n= 1) and seven P. aeruginosa were isolated from different cases in local animals. By combination disc method, six E. coli isolates and one K. pneumoniae isolate were found to be ESBL producers. By combination of the disc method and double disc synergy test, no P. aeruginosa isolates were found as ESBL producers. In the agar disc diffusion test (ADDT) performed with cefoxitin and cefoxitin-boronic, only one E. coli was determined as AmpC producer. In ESBL-producing isolates, only the CTX-M class gene was detected by polymerase chain reaction (PCR) and subsequent sequence analysis revealed CTX-M-3 and CTX-M-15 variants. An AmpC positive E. coli isolate was found to carry plasmidic ampC gene in cmy-2 variant from CIT family. It was observed that P. aeruginosa isolates did not carry the plasmidic ampC gene. After the chromosomal ampC gene of one P. aeruginosa was amplified by PCR and sequenced, R79Q and T105A mutations in the chromosomal ampC gene was revealed. This showed that overproduction of the ampC enzyme is involved in the resistance to β-lactams in P. aeruginosa isolates in the study.
