RESUMO -(Germinabilidade de sementes de espécies endêmicas de Encholirium Mart. ex Schult. & Schult. f. e Dyckia Schult. & Schult. f. (Bromeliaceae) após dessecação, armazenamento e criopreservação). O armazenamento de sementes requer a determinação prévia das condições ótimas de temperatura e luz para a germinação, assim como da tolerância à dessecação e a baixas temperaturas. O objetivo deste trabalho foi o estudo do efeito da dessecação, armazenamento a baixa temperatura e criopreservação de seis espécies de Encholirium e duas de Dyckia, selecionadas de acordo com critérios de vulnerabilidade. A germinabilidade de sementes recém-coletadas variou entre 35 e 95%. As sementes apresentaram comportamento fotoblástico, uma vez que a presença de luz foi necessária para induzir a germinação ou aumentar sua eficiência. A dessecação não afetou significativamente a germinabilidade das sementes testadas, com exceção de E. heloisae e E. scrutor. Sementes dessecadas e armazenadas durante um ano a 4 e -20 °C não apresentaram alterações na germinabilidade, exceto em E. pedicellatum. A germinabilidade após congelamento em nitrogênio líquido foi maior que ou similar à obtida no controle, em todas as espécies estudadas. Entretanto, em E. pedicellatum a tolerância ao congelamento só foi obtida após dessecação das sementes a um conteúdo hídrico de 2,5%. Considerando a tolerância à dessecação e ao armazenamento em baixas temperaturas, as sementes estudadas podem ser classificadas como ortodoxas e conservadas ex situ. Seed storage procedures require previous determination of optimal temperature and light conditions for germination, as well as of tolerance to desiccation and low temperatures. The aim of this paper was to study the effects of desiccation, storage at low temperatures and cryopreservation on the germinability of seeds of six Encholirium and two Dyckia species, which were selected according to vulnerability criteria. Initial germinability of newly harvested seeds varied from 35 to 95%. Seeds presented photoblastic behaviour since light was necessary to induce or increase germination. Except for E. heloisae and E. scrutor, desiccation did not affect significantly the germinability of tested seeds. Storage for one year at 4 and -20 °C did not affect the germinability of desiccated seeds, except for E. pedicellatum. Germinability after freezing in liquid nitrogen was higher than or similar to control seeds for all species. However, freezing tolerance of E. pedicellatum seeds was only achieved after desiccation to 2.5% moisture content. As regards tolerance to desiccation and to storage at low temperatures, the seeds studied here can be classified as orthodox and conserved ex situ.
-(In vitro somatic embryogenesis and adventitious root initiation have a common origin in eggplant (Solanum melongena L.)). Somatic embryogenesis was induced from cotyledon explants of eggplant cultured on MS medium supplemented with 54 µM NAA. Anatomical analysis of somatic embryo initiation and development was performed during the first four weeks. Proembryo formation was observed after the second day of culture, directly from perivascular cells or via pro-embryogenic masses derived from indeterminate meristematic masses (IMMs) originated in the vascular tissue. Those IMMs also gave rise to root primordia after 10 days of culture. The origin of embryos is discussed as well as the similarities between somatic embryogenesis and adventitious root formation.Key words -anatomy, development, Solanum melongena, somatic embryogenesis RESUMO -(Embriogênese somática in vitro e iniciação de raizes adventícias têm uma origem comum em berinjela (Solanum melongena L.)) A embriogênese somática em berinjela foi induzida a partir de explantes cotiledonares cultivados em meio MS suplementado com 54 µM de ANA. A análise anatômica do início de formação e desenvolvimento de embriões somáticos foi realizada durante as primeiras quatro semanas. A formação de pré-embriões foi observada após o segundo dia de cultura, diretamente a partir de células perivasculares ou a partir de massas pré-embriogênicas derivadas de massas meristemáticas indeterminadas (IMMs) formadas a partir do tecido vascular. As IMMs também deram origem a primórdios radiculares após 10 dias de cultura. A origem dos embriões é discutida, assim como as similaridades entre a formação de embriões somáticos e raízes adventícias.
The AtGRP5 gene from Arabidopsis thaliana encodes a glycine-rich protein which has a major activity in protoderm-derived cells and is expressed in cells that undergo the first anatomical modifications leading to somatic embryo development. It has been previously demonstrated that its minimum promoter is 316 bp long including the 5'UTR and presents three putative TATA-boxes sequences and several regions that are homologous to previous characterized cis-acting elements. In order to better characterize the AtGRP5 expression and to identify the promoter regions involved in its preferential epidermal expression, in situ hybridization and 5' promoter deletions were employed. In situ hybridization and GUS expression assays indicate that, besides being present during somatic embryogenesis, AtGRP5 is also expressed during the zygotic embryo development. The sequential 5' deletions indicate that multiple negative and positive regulatory elements are present in the AtGRP5 promoter and operate in order to confer its distinct expression pattern. A 44-bp region was shown to be essential for the epidermal expression of this gene in leaves, stems, flowers and fruits, and is also responsible for high activity of the AtGRP5 promoter in zygotic embryos. An element responsible for the phloem expression was also identified in a 35-bp region.
