Aus Clostridium perfringens isolierte und gereinigte Neuraminidase wurde näher charakterisiert. Die Sedimentationskonstante beträgt s20 = 4 ,6 S ; das Mol.-Gew. wurde durch Gelfiltration zu 56000 bestimmt. Die Neuraminidase läßt sich durch 10 -3 M KCN oder 0,03% Rinderserumalbumin stabili sieren. Ihr Temperatur-Optimum liegt bei 50 °C, das pH-Optimum mit Fetuin als Substrat in 0,1 m K-Acetatpuffer bei pH 5,0 bis 5,5.Neuraminidase (V-Acetylneuraminat-glycohydrolase E C 3 . 2 .1 .1 8 ) konnte bei einer Reihe von Bak terien nachgewiesen werden 1_3. Am eingehendsten untersucht wurde das Enzym von Vibrio cholerae, das in kristallisierter Form erhalten werden konnte (1. c. 4~6). Über die Eigenschaften des aus anderen Mikroorganismen gewonnenen Enzyms ist bisher noch wenig bekannt. Die bei Clostridium perfringens gefundene N euram inidase7 konnte von C a s s i d y 8 isoliert und partiell gereinigt werden. In der vorliegenden Arbeit wurde das Enzym aus Clostridium perfringens durch Bestimmung des Mol.-Gew. und einiger enzymkinetischer Eigenschaften näher charakterisiert.
Zusammenfassung: Es wird die Reinigung einer Protease aus Pseudomonas aeruginosa, Stamm 158, beschrieben, die von den Bakterien während der logarithmischen Wachstumsphase in den Kulturüberstand abgegeben wurde. Die Reinigung erfolgte durch 65proz. Ammoniumsulfatfällung, 70proz. Acetonfällung, Gelfiltration an Sephadex G-JOO superfine und Chromatographie an DEAE-Sepha-dex A-50. Das Molekulargewicht wurde mit der Gelfiltrationsmethode zu 22300 bestimmt. Durch isoelektrische Fokussierung konnte die Protease in eine Hauptkomponente mit dem pH-Wert 6.45 und eine Nebenkomponente mit dem pH-Wert 5.7 aufgetrennt werden. Beide Komponenten stimmten in ihrer Substratspezifität und Antigenität überein.
