Neglected diseases, primarily found in tropical regions of the world, present a significant challenge for impoverished populations. Currently, there are 20 diseases considered neglected, which greatly impact the health of affected populations and result in difficult-to-control social and economic consequences. Unfortunately, for the majority of these diseases, there are few or no drugs available for patient treatment, and the few drugs that do exist often lack adequate safety and efficacy. As a result, there is a pressing need to discover and design new drugs to address these neglected diseases. This requires the identification of different targets and interactions to be studied. In recent years, there has been a growing focus on studying enzyme covalent inhibitors as a potential treatment for neglected diseases. In this review, we will explore examples of how these inhibitors have been used to target Human African Trypanosomiasis, Chagas disease, and Malaria, highlighting some of the most promising results so far. Ultimately, this review aims to inspire medicinal chemists to pursue the development of new drug candidates for these neglected diseases, and to encourage greater investment in research in this area.
Objetivo: Este trabalho de revisão tem como objetivo descrever brevemente a evolução da Biologia Sintética em bactérias e a criação de circuitos genéticos induzidos pela luz, conhecida como optogenética. Método: Foram utilizados bases de dados online como PubMed-NCBI e OptoBase, para realizar a busca e análise dos artigos científicos escolhidos usando como palavras-chave: “optogenetics”, “optogenetics and applications”, e “optogenetics and synthetic biology”. Resultados: Um dos marcos da história da biologia sintética é o uso da luz visível para o controle da regulação genética em bactérias, principalmente no modelo Escherichia coli. Proteínas fotorreceptoras ocorrem naturalmente em microrganismos e eucariotos (como fitocromos e opsinas), mas também podem ser construídas através da fusão de diferentes domínios sensíveis à luz combinados a diversos outros domínios efetores. Estes novos sistemas responsivos, representados geralmente pelo complexo Cph1–Rcp1, CcaS/R e YF1–FixJ, são induzidos por comprimentos de onda de luz entre 650-705 nm (luz vermelha/vermelho longo), 563 nm (luz verde) e 470 nm (luz azul), respectivamente. Além da descrição do funcionamento desses receptores de luz, alguns exemplos de suas aplicações são citados. Conclusão: O uso de sistemas optogenéticos permitiu obter um maior controle espacial e temporal da expressão gênica em bactérias, abrindo novas perspectivas para a automatização de processos biológicos e diminuição dos custos de bioprodução no setor da biotecnologia industrial.
