This work aimed to assess the clonal distribution among 94 strains of Staphylococcus aureus isolated from cow's milk, raw cheese, and a milking machine in 12 dairy farms in northeast Brazil, by analyzing different typing methods and detecting resistance and toxigenic profiles. For the first time, isolates of this region were assessed simultaneously by the polymorphism of the 3'-end coa gene and 16S-23S rDNA, pulsed-field gel electrophoresis, antibiotic resistance phenotyping, and toxigenic arsenal. Although pulsed-field gel electrophoresis patterns showed a wider variation (discriminatory index 0.83) than the PCR-based methods, the internal transcribed spacer-PCR proved to be a useful and inexpensive procedure for conducting epidemiological surveys of S. aureus on a regional scale. Each dairy farm had its own resistance profile, and in two herds, 63% of the strains were multiresistant, probably due to the indiscriminate use of antibiotics in bovine mastitis treatment. No methicillin-resistant S. aureus strains were detected in this study; however, 93.6% of S. aureus strains harbored variable profiles of staphylococcal enterotoxin genes seg, seh, sei, and sej. Transcriptional analysis revealed that 53.3% of staphylococcal enterotoxin genes actually transcribed, pointing out the food poisoning risk of these dairy products to consumers in the region. Based on the detection of the most prevalent clones in a herd or region, appropriate antibiotic therapy and specific immunization can be used for the treatment and control of staphylococcal mastitis.
Objetivou-se com este estudo avaliar a sensibilidade in vitro de Staphylococcus spp.frente a alguns desinfetantes comerciais utilizados no pré e pós-dipping em vacas leiteiras. Foram testados um total de 60 isolados de Staphylococcus spp. identificados como S. aureus (50) e Staphylococcus coagulase positiva (10) recuperados de glândulas mamárias de vacas com mastite subclínica procedentes das regiões Metropolitana do Recife, Agreste e Zona da Mata do Estado de Pernambuco. O estudo da eficácia dos desinfetantes utilizados no pré e pós-dipping foi realizado utilizando-se os seguintes princípios ativos: cloro (2,5%), iodo (0,57%), clorexidine (2,0%), amônia quaternária (4,0%) e ácido lático (2,0%) em quatro tempos distintos (15", 30", 60" e 300"). Observou-se que 100% de S. aureus foram sensíveis ao iodo, 93,3% sensíveis a clorexidine, 80% sensíveis a amônia, 35,6% sensíveis ao ácido lático e 97,8% resistentes ao cloro no tempo de 60". Com relação a Staphylococcus coagulase positiva (SCP), 100% dos isolados foram sensíveis ao iodo, 81,8% sensíveis a amônia quaternária, 99,9% sensíveis ao ácido lático, 72,7% sensíveis a clorexidine e 100% resistentes ao cloro no tempo de 60". Conclui-se que a maior atividade desinfetante in vitro foi verificada para o iodo e clorexidine frente a S. aureus e do iodo e ácido lático frente aos SCP e que há necessidade de avaliação periódica dos desinfetantes utilizados nas propriedades leiteiras nas regiões estudadas, pois, existem variações no perfil de sensibilidade e resistência aos desinfetantes que podem comprometer os programas de controle da mastite bovina causada por Staphylococcus spp.
ABSTRACT.-Mota R.A., Peixoto P.V., Yamasaki E.M., Medeiros E.S., Costa M.M., Peixoto R. (MAP). No record on the occurrence of paratuberculosis in buffaloes in Brazil could be found. Five of 100 buffaloes in a herd in Pernambuco-Brazil showed clinical signs characteristic of PTB. At necropsy, of two animals the lesions were restricted to the small intestine with thickening and corrugation of the mucosa, increase of mesenteric lymph nodes and prominent lymph vessels. Histopathology revealed granulomatous inflammation infiltrated with numerous epithelioid macrophages, Langhans type giant cells, and clusters of Ziehl-Neelsen (ZN) positive organisms within the intestinal mucosa. In the mesenteric lymph nodes there was thickening of the capsule and marked granulomatous inflammation. Smears of feces and scrapping smears were prepared from intestinal mucosa and cut surface of mesenteric lymph nodes and, stained by the Ziehl-Neelsen method for research of acid fast bacilli, with positive results. Lymph nodes and intestinal mucosa revealed at IS900 specific polymerase chain reaction amplification of a fragment of about 110pb, confirmed by the comparison with other sequences of M. avium subsp. paratuberculosis available in GenBank.
Resumo: A mastite é uma doença complexa e considerada uma das principais causas de perdas à indústria leiteira mundial. Objetivou-se com esta revisão compilar informações dos últimos dez anos sobre a mastite em ruminantes no Brasil. A prevalência da mastite subclínica chega a 48,64% na espécie bovina, 30,7% na espécie caprina, 31,45% na espécie ovina e 42,2% na espécie bubalina, destacando-se a etiologia por Staphylococcus spp. Os fatores de risco associados à ocorrência de mastite estão relacionados a problemas no saneamento ambiental e ao manejo dos animais. As bactérias isoladas do leite mastítico apresentam maior percentual de resistência a penicilina, ampicilina, amoxicilina e neomicina e a utilização de técnicas moleculares no diagnóstico dos agentes causadores de mastites no país, ainda é escassa o que dificulta a obtenção de um diagnóstico mais rápido, sensível e específico.
Objetivou-se com este estudo avaliar a sensibilidade antimicrobiana in vitro de 291 isolados de Staphylococcus spp. recuperados de amostras de leite de vacas com mastite subclínica, em 15 propriedades rurais localizadas na Região Metropolitana do Recife (A), Agreste (B) e Zona da Mata (C) do estado de Pernambuco. Dos 291 isolados, 170(58,4%) foram classificados como Staphylococcus coagulase negativa (SCN), 84(28,9%) como Staphylococcus aureus e 37(12,7%) como Staphylococcus coagulase positiva (SCP). Para o estudo do perfil de sensibilidade a antimicrobianos empregou-se a técnica de difusão em discos, foram avaliadas 16 drogas antimicrobianas utilizadas no tratamento das mastites. O antibiótico que apresentou melhor eficácia in vitro foi a associação entre neomicina + bacitracina + tetraciclina com percentuais de 98,4%, 99,3%, 89,7% para as regiões A, B e C, respectivamente. O antibiótico menos eficaz foi a ampicilina que apresentou 56,5% de resistência para as amostras da região A, 72,8% para a região B e 71,8% na região C. Os resultados obtidos mostram a necessidade da realização periódica de testes de sensibilidade in vitro, pois existem variações no perfil de sensibilidade e resistência que podem comprometer o tratamento do animal bem como os programas de controle da mastite bovina causada pelo Staphylococcus spp.
The aim of this study was to identify risk factors associated with Neospora caninum seropositivity in sheep from the State of Alagoas, in the northeast region of Brazil. Twenty-six herds were selected, and blood samples were collected from 343 animals > 6-mo-old. The indirect fluorescent antibody test (IFAT) was used for serological diagnosis of N. caninum infection. Epidemiological questionnaires were applied for each herd to identify the risk factors associated with infection. This study was conducted to investigate the participation of Toxoplasma gondii in reproductive failure in small ruminants raised in the Litoral/ Zona da Mata and Agreste of the State of Pernambuco, Brazil. Twelve ϐlocks were selected from which 262 samples were collected, 167 from goats and 95 from sheep. Indirect Imunoϐluorescent Antibody Reaction (IFA) technique was used to detect antibodies anti-Toxoplasma gondii. A questionnaire was applied in ϐlocks to identify risk factors associated with infection. Positive animals were found in 100% of the farms. Among 167 samples of goat serum analyzed, 31.7% were positive, whereas from 95 samples of sheep, 16.9% were positive. Results of brucellosis tests were negative for 100% of analyzed samples. For sheep, no signiϐicant association was found; however, for goats, there was a signiϐicant association (p<0.05) for intensive handling (OR=2.40), milk exploration (OR=2.10), animals originating from other states (OR=7.89) and natural breeding (OR=5.69). It was concluded that infection for T. gondii is spread in studied goat and sheep ϐlocks and that sanitary measures must be adopted to control risk factors for infection.INDEX TERMS: Indirect imunoϐluorescent antibody reaction, Toxoplasma gondii, risk factors.
A subclinical mastitis study was conducted in nine dairy goat herds in the semi-arid region of Paraíba state, Northeastern Brazil, to determine the occurrence of infection, to evaluate microbiological and cellular profiles of the milk, to test the sensitivity of isolated microorganisms to antimicrobials, and to identify risk factors. One hundred thirty-one dairy goats were used, 261 samples were collected for microbiological culture and 131 samples for somatic cells count (SCC). During collection, the California Mastitis Test (CMT) was conducted and an epidemiological questionnaire was applied for each herd. There was bacterial growth in 30 samples (11.49%), with 25 (83.33%) coagulase-negative Staphylococcus and five (16.66%) Staphylococcus aureus isolated. The SCC mean was 1.39x10 6 cells/ml. CMT presented low sensitivity (46.7%) and low specificity (60.6%) compared with microbiological culture. Gentamicin and the association of neomycin, bacitracin and tetracyclin were the antimicrobials against which the microorganisms isolated showed 100% sensitivity. Penicillin and ampicillin had the greatest resistance rates (66.67% and 63.89%, respectively). Goat breeding is not the main activity on the farms and do not isolate diseased animals were identified as risk factors for caprine subclinical mastitis. Program for the control and prophylaxis of mastitis must be implemented focusing primarily on hygiene cares at milking and correction of the risk factors identified in this study.
