The application of phytases for animal feed in developing countries is limited due to the high cost of these enzymes, determined by the importation fees and the expensive substrates used for their production. In this work, we have used agroindustrial byproducts for the production of extracts containing phytases, which were accessed for their stability focusing on the conditions found in the gastrointestinal tract of pigs. The fungus Acremonim zeae presented higher phytase production in medium containing cornmeal, while the yeast Kluyveromyces marxianus produced 10-fold more phytase when cultivated on rice bran. Process optimization increased the difference in productivity to more than 300 fold. The phytase from A. zeae was thermostable, with higher activity at neutral pH and 50 °C, but was inhibited at pH 2.5 and by various ions. The phytase activity in the K. marxianus extract was stable at a wide range of conditions, which indicates the presence of at least two enzymes. As far as we know, this manuscript describes for the first time the phytase production and the characteristics of the extracts produced by both these microbial species. These enzymes could be produced at low cost and have potential to replace enzymes currently imported for this purpose.
Phytic acid is an antinutritional factor in cereal feeds, and the use of phytases increases the bioavailability of nutrients bound to this molecule. However, the application of these enzymes depends on their thermal stability and activity at acidic pH. Therefore, in this study we created a database composed of 59 phytase sequences and analyzed the interactions that stabilize their structures in order to understand whether they contribute to the biochemical properties observed. The sequences were aligned and grouped at 30 % similarity, generating 5 clusters, which highlights the high variability among them. A comparative structural analysis of the cluster 3 phytases revealed conserved catalytic domains, as well as eight cysteine residues along the primary sequence, forming disulfide bonds for stabilizing the three-dimensional structure. However, the number of Van der Waals, ionic, and hydrogen interactions, and disulfide bonds was not determinant for the biochemical characteristics presented by these enzymes. The phytase KM873028, from cluster 3, was selected for characterization studies, but its expression in Pichia pastoris generated a protein with properties distinct from those derived from the same sequence expressed in a prokaryotic system. It is likely that the differences observed are associated with the location of the interactions in the structures, non-conserved amino acid residues found around the catalytic site, and post-translational modifications inherent to the expression systems. These possibilities highlight the relevance of strategic choices related to enzyme expression aiming at its production and industrial feasibility.
A utilização eficiente dos nutrientes pelos animais é fator de grande relevância para indústria de nutrição animal. Esse resultado depende da eficiência da fitase sobre o complexo fitato-minerais, o qual representa, nos alimentos, um fator antinutricional pelo aprisionamento de nutrientes essenciais requeridos para o bom desempenho dos animais. No entanto, a pouca resistência a altas temperaturas e ao ambiente gástrico dos animais podem comprometer a aplicação dessa enzima, reduzindo assim a eficiência de utilização de nutrientes, com consequente problemas econômicos e ambientais. Para tal efeito, torna-se necessário a obtenção de uma fitase que seja capaz de hidrolisar o fitato nestas condições. Assim, entender o mecanismo de ação das fitases permitiria a obtenção de fitases com características adequadas requeridas para sua aplicação. Este trabalho teve como objetivos análise estrutural e estudos de conservação de fitases para otimização de uma fitase para aplicação industrial. Dessa forma, a primeira parte deste trabalho compreende a formação de um banco de dados de fitases e análises comparativas estruturais em relação às propriedades bioquímicas destas enzimas, a fim de compreender fatores que influenciam na estabilidade dessas enzimas. Outra etapa da pesquisa, consistiu no estudo de expressão e caracterização da fitase de Dendroctonus frontalis, selecionada entre as fitases estudadas por apresentar estabilidade térmica satisfatória. Foram também avaliados estudos de secagem dos extratos de fitase obtidos a partir desta, pelos métodos de liofilização e Spray Dryer. Com base nas propriedades bioquímicas e no alinhamento de sequências de aminoácidos, observamos alta variabilidade entre as fitases estudadas, com propriedades bioquímicas bastante distintas, apesar destas apresentarem semelhança estrutural. No entanto, na análise estrutural comparativa, o número de interações de Van der Waals, interações iônicas, hidrogênio e ligações dissulfeto que constituem essas fitases não explicaram asdiferenças nas suas propriedades bioquímicas. A sequência codificadora da fitase selecionada foi sintetizada no plasmídeo pPICZαB e expressa em Pichia pastoris. Após a clonagem e seleção dos transformantes, a expressão da fitase foi avaliada em diferentes composições de meios de cultivo e pHs. O uso dos meios mBMMH e BMMY em pH 5,0 promoveram um aumento de 3 vezes no nível de expressão da fitase em relação à expressão realizada em BMMH. A enzima expressa possuiu maior atividade em pH 4,0 e temperatura de 42 °C. Entretanto, sua estabilidade foi drasticamente reduzida quando submetida a temperaturas de 70 e 80 °C, por apenas 1 min. A secagem dos extratos por liofilização preservou 62 % da atividade enzimática, na presença de maltodextrina a 5 %. Na ausência desse composto, apenas 41 % de atividade foi recuperada. Os extratos submetidos a secagem por Spray Dryer, apresentaram maior atividade residual a 100 °C e 5 % de maltodextrina, acima desta condição a atividade reduziu significativamente. Embora a secagem dos extratos por Spray Dryer tenha causado redução significativa na atividade enzimática, o seu uso permitiu que o pó seja armazenado por pelo menos 240 dias à temperatura ambiente, sem perda de atividade. A partir deste estudo podemos concluir que o mecanismo de ação de uma fitase está intimamente relacionada a sua estrutura e sua eficiência precisa ser analisada individualmente de acordo ao sistema de expressão escolhido. Este trabalho pode auxiliar para uma melhor compreensão do mecanismo de ação das fitases e obtenção de novas fitases para aplicação comercial. Palavras-chave: Ácido fítico. Fitase.
