Efeito da adição do ácido linoleico conjugado no cultivo in vitro de embriões F1 Holandês x Zebu na sobrevivência pós-vitrificação
Este trabalho objetivou realizar uma revisão de literatura sobre bebidas mistas de frutas e vegetais e seus principais componentes no contexto do consumo alimentar. O presente estudo é uma revisão narrativa elaborada a partir da consulta em diferentes bases de dados, como: Web of science e Science Direct. Optou-se pela busca por termos livres. Os termos juice, drinks foram combinados com as palavras fruits “AND” vegetables para localizar estudos sobre os dois temas. Posteriormente, a fim de selecionar estudos com os principais componentes das bebidas mistas, foram utilizados os termos livres “citrus limon”, “Malus domestica”, “Ananas comosus”, “Cucumis savitus”, “Brassica oleracea”, “Spinacia oleracea”, “Zingiber officinale” e “Mentha ssp”. O critério de inclusão dos trabalhos foi pautado na seleção de materiais que tivessem por objetivo estudar os efeitos isolados do consumo dos principais ingredientes das bebidas mistas, como também os artigos que utilizaram a bebida mista com alegação “detox” nos estudos. O direcionamento dos artigos selecionados está relacionado com a prospecção da atividade antioxidante e, apesar da composição das bebidas ser diversificada, destacam-se o uso de maçã, de repolho e de pepino como ingredientes das preparações. As bebidas mistas, sucos ou smoothies apresentaram variável composição que determina os atributos sensoriais, composição físico-química, compostos bioativos e atividade antioxidante. Palavras-chave: Suco. Nutrição. Alimentação. Abstract This study aimed to conduct a literature review on mixed fruit and vegetable drinks and their main components in the context of food consumption. The present study is a narrative review elaborated from the consultation in different databases, such as Web of science and Science Direct databases. It was chosen to search free terms. The terms juice, drinks were combined with the words fruits “AND” vegetables to locate studies on both themes. Subsequently, in order to select studies with the main components of mixed drinks, the free terms "citrus limon", "Malus domestica", "Ananas comosus", "Cucumis savitus", "Brassica oleracea", "Spinacia oleracea" ”,“ Zingiber officinale ”and “Mentha ssp ”. The inclusion criterion of the studies was based on the selection of materials that aimed to study the effects of vegetable consumption alone, which are the main ingredients of mixed drinks and included articles that used the mixed drink or juices with claim “detox” in the studies. The direction of the selected studies is related to the prospecting of antioxidant activity and, although the drinks composition is diverse, the use of apple, cabbage and cucumber as ingredients of the preparations stand out. Plant foods are important sources of nutrients for food. The mixed drinks, juices or smoothies presented variable composition that determines the sensory attributes, physicochemical composition, bioactive compounds and antioxidant activity. The aim of this work was to evaluate, in vitro, the dynamics of nuclear and cytoplasmic maturation of bovine oocytes in traditional IVM medium (CT) and supplemented with fullerol (MF50), for 36 hours. The nuclear maturation of CT (n=300) and MF50 (n=270) every 6 hours, stained with Hoechst33342 and cytoplasmic, the mitochondrial distribution of CT (n=197) and MF50 (n=159) at every 12 hours, stained with Mitotracker Orange. At 6 hours, CT oocytes (19%) were in MI (metaphase I), while in MF50 they were in GV (germ vesicle) or GVB (GV breakeage), repeating at 12 hours. At 18 hours, 46.3% were matured in CT, and 20% in MF50. At 24 hours, 43.9% of maturation was observed in the MF50 group, and 63.8% in the CT. At 30 and 36 hours, the maturation pattern was stable, but with the onset of oocyte degeneration. There was a delay in cytoplasmic maturation with 36 hours (P<0.05) in MF50 (53.9% of mature gametes), compared to CT (69.8%). With immature cytoplasm, they were 10.4% and 31.7% for CT and MF50 (P<0.05), respectively. It was concluded that fullerol possibly interfered in the expansion of cumulus oophorus cells, as well as delayed the meiotic progression and cytoplasmic maturation.
Introdução:Os rótulos fornecem informações para a orientação do consumidor sobre as escolhas alimentares mais adequadas às suas necessidades, sendo um importante canal de comunicação entre o consumidor e o produtor de alimentos. Para cumprir esse papel, é fundamental que essas estejam corretas e atendam aos parâmetros legais estabelecidos. Objetivo:Avaliar a conformidade dos rótulos das bebidas mistas industrializadas à base de frutas e vegetais que apresentavam a denominação suco detox, comercializadas em Belo Horizonte (MG), Brasil, entre os anos de 2017 e 2019. Método:Foram selecionadas inicialmente 23 bebidas mistas e, após a avaliação dos critérios de exclusão, sete amostras de diferentes marcas foram selecionadas para o estudo. A análise da rotulagem foi realizada conforme a regulamentação vigente para alimentos na legislação brasileira, utilizando um checklist com os seguintes itens obrigatórios: denominação do produto; lista de ingredientes; conteúdo líquido; identificação de origem; identificação de lote; prazo de validade; cuidados de conservação; uso de frases e imagens que podem causar erro ou engano ao consumidor; rotulagem nutricional; advertência sobre o glúten e o número do registro das bebidas no Ministério da Agricultura e Pecuária e Abastecimento (MAPA). Resultados:Em 100% (n = 7) das amostras foi observada a irregularidade do uso do termo detoxnos rótulos e ausência do registro junto ao MAPA. Setenta por cento (n = 5) apresentavam erros na lista de ingredientes e informação nutricional e em 57% (n = 4) não estava correta a informação do conteúdo líquido. Conclusões:Os resultados evidenciaram a presença de não conformidades em todos os produtos analisados. Destacaram-se as irregularidades em relação à denominação de venda e a ausência de registro. Resumo Niedenzuella (Tetrapterys) multiglandulosa, uma videira encontrada no Brasil, tem sido correlacionada a surtos de intoxicações em bovinos e búfalos, gerando perdas econômicas relacionadas à morte por insuficiência cardíaca, aborto, natimorto e mortalidade neonatal. O objetivo deste estudo foi examinar o potencial embriotóxico do extrato vegetal aquoso em embriões bovinos in vitro. O estudo in vitro foi realizado em cinco repetições de cultura de embriões bovinos distribuídos em dois grupos: controle, meio de cultura de embriões in vitro sem o extrato aquoso da planta; grupo tratado, com adição de 2,7mg / mL de extrato vegetal aquoso (10%) à cultura do embrião no sexto dia de cultivo. A taxa de clivagem foi avaliada no dia 2 da cultura de células. Viabilidade, eclodibilidade e subdesenvolvimento de blastocistos no sétimo, oitavo e nono dia (D7, D8 e D9, respectivamente) de cultura foram avaliados em microscópio estereoscópico. No dia 7, os blastocistos foram submetidos ao ensaio TUNEL para determinar o índice apoptótico. Observamos redução significativa da produção de blastocisto / número de embriões clivados (60,6% vs 41,5%); redução da produção de blastocistos / número total de oócitos bovinos maturados (35,1% vs 21,3%); e taxas de eclosão embrionária (38,0% vs 10,0%). No entanto, nenhum efeito foi observado na taxa de apoptose. Em conclusão, o extrato aquoso das folhas de N. multiglandulosa reduz a viabilidade do embrião bovino in vitro, sugerindo possíveis efeitos prejudiciais no desenvolvimento embrionário.
Niedenzuella (Tetrapterys) multiglandulosa, a vine plant found in Brazil, has been correlated to outbreaks of poisoning in cattle and buffaloes, generating economic losses related to the death due to heart failure, miscarriage, abortion, stillbirth, and neonatal mortality. The aim of this study was to examine the embryotoxic potential of the aqueous plant extract on in vitro bovine embryos. In vitro study was performed in five replicates of bovine embryo culture assigned in two groups: control, in vitro embryo culture medium without the aqueous plant extract; treated group, with addition of 2.7mg/mL of aqueous plant extract (10%) to the embryo culture on the sixth day of culture. Cleavage rate was evaluated at day 2 of the cell culture. Viability, hatchability and underdevelopment of blastocysts on the seventh, eighth, and ninth days (D7, D8, and D9, respectively) of culture were assessed under stereoscopic microscope. On day 7, blastocysts were submitted to TUNEL assay to determine apoptotic index. In vitro exposure of bovine embryos to of N. multiglandulosa resulted in reduced embryo development and survival, evaluated by dark cytoplasm indicating poor morphology and poor quality with marked reduction of hatchability. We observed a significant reduction of blastocyst production/number of cleaved embryos (60.6% vs 41.5%); reduction of blastocysts production/total number of matured bovine oocytes (35.1% vs 21.3%); and embryonic hatching rates (38.0% vs 10.0%). However, no effects were observed on the apoptotic rate. In conclusion, aqueous extract of N. multiglandulosa leaves reduces bovine embryo viability in vitro, suggesting possible detrimental effects on embryo development.
