-Alterations caused by a genotype III strain of Toxoplasma gondii were assessed with respect to the number and the morphometry of the myenteric neurons in the terminal ileum and the descending colon. Eighteen rats were divided into four groups: Acute Control Group (ACG, n=4); Acute Experimental Group (AEG, n=4); Chronic Control Group (CCG, n=5) and Chronic Experimental Group (CEG, n=5). NaCl solution was administered through gavage to the animals in the ACG and CCG. Toxoplasma gondii tachyzoites (10 4 ) from a genotype III strain were orally administered to the AEG and CEG. Acute Groups were died after 24 hours, and the Chronic Groups after 30 days. Neuronal loss was not observed in both organs. The neurons atrophied in the terminal ileum as the opposite occurred with the neurons at the descending colon during the chronic phase of infection. In the terminal ileum, the neurons atrophied during the chronic phase of the infection as no alteration was found during the acute phase. For the descending colon, the neurons became hypertrophic during the chronic infection in opposition to the atrophy found during the acute phase.KEY WORDS: enteric nervous system, toxoplasmosis, morphology. Alterações do plexo mientérico do íleo e cólon descendente causadas por Toxoplasma gondii (genótipo III)Resumo -Objetivou-se avaliar as alterações causadas por uma cepa genótipo III de Toxoplasma gondii, sobre o número e a morfometria de neurônios mientéricos, do íleo terminal e do cólon descendente. Dividiuse dezoitos ratos em quatro grupos: controle agudo (GCA, n=4), experimental agudo (GEA, n=4), controle crônico (GCC, n=5) e experimental crônico (GEC, n=5). Os animais do GCA e GCC receberam solução de NaCl por gavagem, e os animais do GEA e GEC 10 4 taquizoítos de uma cepa genótipo III de T. gondii por via oral. Os grupos agudos após 24 horas foram mortos e os crônicos após 30 dias. Observou-se que não houve perda neuronal em ambos os órgãos. No íleo terminal, os neurônios atrofiaram-se na fase crônica da infecção, enquanto nenhuma alteração ocorreu na fase aguda. Já no cólon descendente, os neurônios tornaram-se hipertróficos na fase crônica da infecção, em oposição à atrofia observada na fase aguda. O Cymbopogon citratus é uma planta originária da Índia, pertencente à família Poaceae e facilmente adaptada em regiões tropicais como o Brasil. Este trabalho avaliou a atividade antimicrobiana do óleo essencial (OE) obtido por hidrodestilação das folhas de C. citratus. A concentração inibitória mínima (CIM) foi determinada pela técnica de microdiluição em caldo para as bactérias Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa e a levedura Candida albicans. O menor CIM foi de 7,81 µg mL-1 para S. aureus, a média de 15,62µg mL-1 para P. aeruginosa e C. albicans e a maior de 62,50µg mL-1 para E. coli. O óleo essencial de C. citratus apresentou atividade antimicrobiana para todos os micro-organismos testados. O óleo mostrou potencial aplicabilidade no controle microbiano e sugerem-se novos estudos para auxiliar na ampliação das aplicações desta planta.
The aim of this study was to evaluate the chemical composition and the larvicidal activity of Brunfelsia uniflora leaf and flower extracts against Aedes aegypti larvae.Twenty-four compounds were found in the leaf extract, and the major compounds were phytol (23.1%), 9,12,15-octadecatrienoic acid, ethyl ester (21.3%), and hexadecanoic acid, ethyl ester (12.8%). In the flower extract, twenty-four compounds were also identified and the major compounds were α-amyrin (35.7%), β-amyrin (16.4%), and (EE)-geranyl linalool (9.6%) by gas chromatography coupled to mass spectrometry. The larvicidal activity was evaluated by larval immersion test. The lethal concentrations (LC) obtained from leaf extract were LC 50 = 4.89 and LC 99.9 = 11.14 mg/mL and from flower extract were LC 50 = 3.82 and LC 99.9 = 11.03 mg/mL, and the positive control presented LC 50 = 0.40 and LC 99.9 = 1.14 mg/mL. Thus, B. uniflora extracts are promising alternatives to control A. aegypti larvae.
ABSTRACT. This paper verified possible alterations caused by a genotype II Toxoplasma gondii strain with respect to the total number and morphometry of the myenteric neurons in the terminal ileum and descending colon of rats. Eight rats were divided into two groups: control (n = 4) and experimental (n = 4). This group was inoculated orally with 20 tissue cysts of T. gondii from a genotype II strain (ME-49). Whole mounted from the terminal ileum and the descending colon were stained with Giemsa. There was not any neuronal loss on both organs. The neurons became hypertrophied in the terminal ileum, whereas morphometric alterations were not observed for the neurons in the descending colon.
ABSTRACT. Brunfelsia genus is traditionally utilized in popular medicine due to its antibacterial and antifungal properties to name but a few. However, studies on the antimicrobial activity of Brunfelsia uniflora flower oleoresin have not been found yet. This study aimed to evaluate the chemical composition and antimicrobial activity of B. uniflora flower oleoresin obtained by supercritical carbon dioxide. Oleoresin from the plant dried flowers was obtained by carbon dioxide, and the chemical composition was analyzed by gas chromatographicmass spectrometry. The minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and minimum fungicidal concentration (MFC) of this oleoresin for seven bacteria and eight fungi were determined using 96-well microtiter plates. The oleoresin MBC for Bacillus cereus, Enterobacter cloacae, Escherichia coli, Listeria monocytogenes, Pseudomonas aeruginosa, Salmonella enterica, and Staphylococcus aureus ranged from 0.01 to 0.08 mg/mL, whereas the controls streptomycin and ampicillin varied from 0.1 and 0.5 mg/ mL. The oleoresin MFC for Aspergillus fumigatus, Aspergillus niger, Aspergillus ochraceus, Aspergillus versicolor, Penicillium funiculosum, Penicillium ochrochloron, Penicillium verrucosum var. cyclopium, and Trichoderma viride varied from 0.01 to 0.08 mg/mL, whereas the controls bifonazole and ketoconazole ranged from 0.2 to 3.5 mg/ mL. The oleoresin obtained by supercritical carbon dioxide presented bacteriostatic, bactericidal, fungistatic, and fungicidal activities that were higher than the positive controls streptomycin, ampicillin, bifonazole, and ketoconazole. The high antimicrobial activity was related to the high content of (E, E)-geranyllinalool that composes 21.0% of the oleoresin and a possible synergic action with fatty acid esters that made up 50.5% of the oleoresin. The oleoresin antimicrobial activity against common multiresistant bacteria in severe infectious processes as P. aeruginosa or against toxin-producing fungi such as P. ochrochloron or fungi that are difficult to control such as T. viride suggests the development of promising applications of this product in the food, farming, livestock, and pharmaceutical industry.
The aim of this paper was to conduct a bibliometry search about Brazilian scientific publications that describe experimental orthopedic surgeries in dogs. It was analyzed 14 publications, and there was wide variation in the number of experimental animals, showing that there is no consensus on the appropriate number for science experiment with dogs. Most articles had been reviewed by an ethics committee (13/14, 92.85%) and only two articles (2/14; 14.28%) did not report the use of analgesic protocol. Among analgesics, morphine was the most widely used, noted in seven studies (7/14; 42.85%). In six of the articles (6/14; 42.85%), the final destination of animals was not informed, but in four experiments (4/14; 28.57%) the dogs were euthanized. In four studies (4/14; 28.57%) the animals were sent for adoption. In conclusion, the ethics in animal research in Brazil has advanced, but is necessary proper planning in sample size.
Bovine tick has caused losses in livestock production profitability in Brazil. However, tick control has caused resistance of these ectoparasites against utilized acaricides. Alternative tick controls have been utilizing plants as sources of effective botanical acaricides. Brunfelsia uniflora is a Brazilian plant with antimicrobial and antioxidant activity; however, there are no reports on its acaricidal activity. Therefore, this study aimed to evaluate the chemical composition of B. uniflora leaf ethanolic extract and its efficiency to control bovine tick in vitro and free-living stage ex situ. The crude leaf extract was analyzed by gas chromatographer coupled to mass spectrometer (GC-MS) with identification of 17 compounds. The major compounds were phytol (22.96%), 9,12,15octadecatrienoic acid, ethyl ester (Z,Z,Z) (21.18%), hexadecanoic acid, ethyl ester (12.74%) and vitamin E (8.77%). The crude extract presented acaricidal activity in vitro against ingurgitated adult females, larvae and eggs of bovine tick. The LC 99.9 for larvae was 103.21 mg mL -1 in in vitro tests and was 100% efficient for ex situ larva test (free-living stage). B. uniflora leaf extract is an alternative for the control of the bovine tick cycle, mainly in the free-living stage (non-parasitic stage) under field conditions.
