To understand the evolutionary dynamics of human parvovirus B19, we analyzed VP1 and VP2 gene sequences of B19 sampled from Belém (Amazon), the city of São Paulo, Brazil and globally. Our analysis revealed a strikingly different pattern of evolutionary change for those viral lineages introduced into Belém, which exhibited a higher rate of nonsynonymous substitutions compared to those viruses sampled from other locations. We propose that difference this is due to the high prevalence of B19 in Belém (up to 85%) compared to other locations (prevalences of approximately 50%), which imposes a more intense selection pressure. Hence, those B19 lineages introduced into Belém experienced an elevated rate of amino acid change, driven by positive selection, in order to generate serial re-infections in a small web of transmission, which can be thought of as an evolutionary "pressure pan".

Objetivo: extraer y evaluar la actividad antiviral de los compuestos de Chondracanthus chamissoi y Chlorella peruviana contra DENV-2 en células Vero-76. Materiales y métodos: se extrajeron el carragenano de Chondracanthus chamissoi, los carbohidratos solubles de Chondracanthus chamissoi y Chlorella peruviana y se realizó la prueba de toxicidad en células VERO-76 y la evaluación de la actividad antiviral. Resultados: se obtuvieron carragenanos de la fase de esporofito y gametofito de Chondracanthus chamissoi, los mismos que fueron identificados, mediante infrarrojo, como k-carragenano. Por cromatografía se identificaron nueve azúcares (ribosa, xilosa, arabinosa, fructuosa, manosa, galactosa, sucrosa, maltosa y lactosa) en la muestra de carbohidratos solubles de Chondracanthus chamissoi fase gametofito y cuatro azucares (glucosa, sucrosa, maltosa y lactosa) en la de Chlorella peruviana. Los compuestos de Chondracanthus chamissoi y la solución de carbohidratos solubles de Chlorella peruviana no presentaron efecto citotóxico; los carbohidratos del extracto crudo de Chlorella peruviana sí los tuvieron. Todas las fracciones del extracto crudo de Chondracanthus chamissoi fase gametofítica fueron positivas por la prueba de reducción del número de placas(50) a la dilución 1:5. El k-carragenano de Chondracanthus chamissoi en ambas fases y los extractos crudos de carbohidratos solubles de Chondracanthus chamissoi, Chlorella peruviana y la solución de carbohidratos solubles de Chlorella peruviana inhibieron el crecimiento del virus dengue, pero no los carbohidratos del extracto crudo de la Chlorella peruviana. Conclusiones: los compuestos obtenidos de Chondracanthus chamissoi y Chlorella peruviana presentan actividad antiviral contra DENV-2 por lo cual es necesario continuar los estudios del potencial antiviral de estos compuestos fraccionados y purificados.
Objetivos. Comparar diferentes métodos de extracción de ADN a partir de quistes y trofozoítos de Giardia spp. mediante la técnica de reacción en cadena de la polimerasa (PCR) convencional. Materiales y métodos. Se aislaron quistes de Giardia spp. a partir de 65 muestras coprológicas procedentes de hospitales de referencia nacional, obteniéndose una carga promedio de 5x10 4 parásitos. Asimismo, se cultivaron trofozoítos de Giardia intestinalis (ATCC® 30957™) obteniéndose una carga parasitaria de 5x10 6. Se compararon once métodos de extracción para quistes y seis para trofozoítos. La concentración y pureza del ADN extraído se determinó por espectrofotometría y el rendimiento de la extracción se evaluó mediante la amplificación de los genes beta giardina (bg) y glutamato deshidrogenasa (gdh) por PCR semi-anidada. Resultados. Se observó que el método I mostró la mayor concentración de ADN a partir de quistes (12,24 ng/µL), pureza (1,4) y mejor rendimiento (100% amplificación bg, 60% gdh) en comparación con los otros métodos evaluados. En el caso de los trofozoítos el método que no tuvo pretratamientos presentó la mayor concentración de ADN, pureza y rendimiento (26,56 ng/µL; 1,85; 100% amplificación bg y gdh). Conclusiones. Los pretratamientos mecánicos, de choque térmico y enzimáticos son necesarios para la ruptura de la pared quística de Giardia spp., siendo el marcador molecular bg de mayor rendimiento para detección de ADN de quistes. Los trofozoítos no requieren pretratamientos para lograr resultados satisfactorios. Se cuenta con una metodología reproducible para la extracción de ADN de Giardia spp. a partir de cualquier estadio evolutivo.
