The green algae of the genus Codium have recently been demonstrated to be an important source of sulfated galactans from the marine environment. Here, a sulfated galactan was isolated from the species Codium isthmocladum and its structure was studied by a combination of chemical analyses and NMR spectroscopy. Two fractions (SG 1, approximately 14 kDa, and SG 2, approximately 20 kDa) were derived from this highly polydisperse and heterogeneous polysaccharide. Both exhibited similar structures in (1)H 1D NMR spectra. The structural features of SG 2 and its desulfated derivative were analyzed by COSY, TOCSY, DEPT-HSQC, HSQC, and HMBC. This sulfated galactan is composed preponderantly of 4-sulfated, 3-linked beta-D-galactopyranosyl units. In minor amounts, it is sulfated and glycosylated at C-6. Pyruvate groups are also found, forming five-membered cyclic ketals as 3,4-O-(1'carboxy)-ethylidene-beta-D-galactose residues. A comparison of sulfated galactans from different marine taxonomic groups revealed similar backbones of 3-beta-D-Galp-1.
The year 2007 was marked by widespread adverse clinical responses to heparin use, leading to a global recall of potentially affected heparin batches in 2008. Several analytical methods have since been developed to detect impurities in heparin preparations; however, many are costly and dependent on instrumentation with only limited accessibility. A method based on a simple UV-scanning assay, combined with principal component analysis (PCA), was developed to detect impurities, such as glycosaminoglycans, other complex polysaccharides and aromatic compounds, in heparin preparations. Results were confirmed by NMR spectroscopy. This approach provides an additional, sensitive tool to determine heparin purity and safety, even when NMR spectroscopy failed, requiring only standard laboratory equipment and computing facilities.

Rev Bras Cineantropom Desempenho Hum 2010, 12(2):98-105Resumo -O estudo teve como objetivo verificar o efeito da atividade física programada sobre os testes de aptidão física em escolares adolescentes durante um ano de período letivo. Amostra foi composta de 383 alunos, divididos em dois grupos: caso, com 186 (96 meninos e 90 meninas) e controle, com 197 (108 meninos e 89 meninas), com idade entre 10 e 15 anos. Trata-se de estudo de intervenção com pré e pós-teste, com um ano de duração, no qual o grupo caso foi submetido à atividade física programada e o grupo controle com aulas convencionais de educação física escolar. A aptidão física foi avaliada por testes de sentar-e-alcançar (flexibilidade), resistência muscular (flexão e extensão do cotovelo) e aptidão cardiorrespiratória (corrida e caminhada de nove minutos). O desempenho motor verificado nos três testes analisados (flexibilidade, força e resistência) não apresentou melhora significativa quando envolveu a variável tempo do pré para o pós-teste, em ambos os grupos, mas quando comparado (tipo) o grupo caso (intervenção) com o controle, mostrou diferença significativa com melhora nos testes de força e resistência em ambos os sexos para o caso. Os meninos de ambos os grupos também mostraram predomínio nos testes de força e resistência. De uma forma geral, os valores foram maiores nos meninos em relação às meninas para resistência muscular e aptidão cardiorrespiratória e menor para flexibilidade, e todos aumentaram no pós-teste em relação ao pré-teste e foram maiores no grupo caso em relação ao controle. Em relação à flexibilidade, foi observada diferença estatisticamente significativa apenas entre os gêneros. Em relação à resistência muscular, após ajuste para idade e nível socioeconômico, foi observada diferença estatisticamente significativa entre os gêneros e entre os tipos. Em relação à aptidão cardiorrespiratória, foi observada diferença estatisticamente significativa entre os gêneros e entre os tipos. Abstract-Previous reports have shown that heparin may promote human hypotension and vascular relaxation by elevation of NO levels through unclear mechanisms. We hypothesized that endothelial muscarinic M 3 receptor activation mediates the heparin-induced vasodilation of rat aortic rings. The experiments were carried out using unfractionated heparin extracted from bovine intestinal mucosa, which elicited an endothelium and NO-dependent relaxation of aortic segments with maximal potency and efficacy (EC 50 : 100Ϯ10 mol/L; E max : 41Ϯ3%). Atropine and 1,1-dimethyl-4-diphenylacetoxypiperidinium iodide inhibitors reduced the heparin-dependent relaxation, indicating that M 3 muscarinic receptor is involved in this phenomenon. However, no direct binding of heparin to muscarinic receptors was observed.More importantly, studies performed using the arginine-glycine-aspartic acid peptide and 1-(1,1-dimethylethyl)-3-(1-naphthalenyl)-1H-pyrazolo[3,4-day]pyrimidin-4-amine, an Src family inhibitor, reduced by 51% and 73% the heparin-dependent relaxation, respectively, suggesting the coupling of heparin and M 3 receptor through extracellular matrix molecules and integrin. Furthermore, unfractionated heparin induced activation of focal adhesion protein kinase, Src, and paxillin. Finally, fluorescence resonance energy transfer approach confirmed the interaction of the M 3 receptor to integrin. Taken together, these data demonstrate the participation of M 3 receptor and integrin in heparin-dependent relaxation of vascular smooth muscle. These results provide new insights into the molecular mechanism and potential pharmacological action of heparin in vascular physiology. Este estudo objetivou analisar a associação da força de preensão manual com morbidades referidas e multimorbidade em adultos de Rio Branco, Acre, Brasil, mediante inquérito de base populacional com 1.395 adultos de ambos os sexos. As associações, por sexo, foram estimadas com a técnica de regressão logística. A média de força de preensão manual nos homens (44,8kg) é maior que entre as mulheres (29kg) e reduz com a idade. A diferença da força de preensão manual média entre aqueles classificados como fortes e fracos foi 21kg e 15,5kg, para homens e mulheres, respectivamente. Controlando para a faixa etária, índice de massa corporal e atividade física quando relevante, homens com baixa força de preensão manual tiveram maiores chances de ocorrência de hipertensão [OR = 2,21 (1,35; 3,61)], diabetes [OR = 4,18 (1,35; 12,95)], distúrbio musculoesquelético [OR = 1,67 (1,07; 2,61)] e multimorbidade [OR = 1,99 (1,27; 3,12)]. Nas mulheres, associações entre força de preensão manual e evento cardiovascular, dislipidemia, distúrbio muscolesquelético e multimorbidade não se mantiveram nos modelos multivariados. Este estudo endossa o uso da força de preensão manual como biomarcador de saúde.
ObjetivoAvaliar o estado nutricional em escolares da rede municipal de ensino da cidade de Porto Velho (RO) em relação à idade, ao sexo e às características sócio-demográficas em uma população de baixo nível socioeconômico. MétodosA amostra constituiu-se de 1.057 escolares com baixo nível socioeconômico, de ambos os sexos, entre 7 e 10 anos, selecionada por meio de amostragem aleatória proporcional por setor e, divisão intencional por conglomerado de turma. Foi utilizado um questionário para obter informações sobre os dados sócio-demográficos. Os escolares foram classificados segundo o peso para a estatura em desnutridos, eutróficos, sobrepesos e obesos; e segundo o peso para a estatura e estatura para idade em eutróficos, desnutridos agudos, pregressos ou crônicos. ResultadosNão foi observada diferença significativa entre os sexos para peso ou estatura. Em relação apenas ao peso para a estatura, 86,0% foram classificados com eutróficos, 4,0% desnutridos, 7,0% sobrepesos e 3,0% obesos. Em relação aos índices de peso para estatura e estatura para idade, 89,0% foram classificados em eutróficos, 2,5% desnutridos agudos, 7,0% desnutridos pregressos e 1,5% desnutridos crônicos.
