a Thymin (1) und 4-0-Athyl-thymin (2) werden rnit Hexamethyldisilazan in die Silyl-Verbindungen 6 und 5 ubergefiihrt, deren Struktur rnit Hilfe von IR-und UV-Spektren geklart wird. 5 und 6 reagieren leicht rnit Methyljodid zu den sonst relativ schwer zuganglichen 1 -Methyl-Derivaten. Fur weitere Alkyl-Derivate des Thymins werden neue Synthesewege beschrieben. Die IR-Spektren der Alkyl-Derivate werden diskutiert.Kurzlich wurde von Birkofer und Mitarbb. 293) uber die Synthese heterocyclischer Trialkylsilyl-Verbindungen aus verschiedenen N-haltigen Heterocyclen rnit Hexamethyldisilazan oder Trimethylchlorsilan berichtet. Diese Silyl-Heterocyclen besitzen zum Teil stark erhBhte Reaktionsfahigkeit gegeniiber Acyl-, Alkyl-und Glykosyl-halogenidenz-4).Bei unseren Arbeiten zur Synthese von Pyrimidin-Nucleosiden stellten wir eine Reihe von Silyl-Derivaten der Pyrimidine da1-5~6). Sil yl-VerbindungenBeim Erhitzen von Thymin (1) oder 4-0-Athyl-thymin (2) niit Hexamethyldisilazan werden alle reaktionsfghigen Wasserstoffatome durch den Trimethylsilyl-Rest ersetzt, und es entstehen fast quantitativ 2.4-Bis-0-trimethylsiiyl-thymin ( 6 ) 7 ) bzw. 4-O-~thyl-2-0-trimethylsilyl-thymin (5). --1) Bezuglich der Nomenklatur tautomeriefahiger Pyrimidine vgl. Auch ohne Silberperchlorat wurde 5a beim Ei-hitzen von 1 rnit Methyljodid unter RuckfluB nahezu quantitativ erhalten. 3-Methyl-thyniin oder hdher methylierte Produkte waren nicht nachueisbar. Die Silyl-Verbindung 2 (aus 4-0-Athyl-thymin) reagierte mit Methyljodid analog 7um 1 -Methql-4-O-athyl-thymin I (4a) Beim Erhitsen von 2 mit uberschussigem Methyljodid unter RuckfluB (ohne Silberperchlorat) wuide 4a jedoch nur in gerrnger Menge isoliert (10-15 %); die Hauptmefige wurde unter diesen Reaktionsbedingungen zum I -Methyl-thymin (5a) entalkyliert.Die be] der Methylrerung der Silyl-pyrimidine 1 b7w 2 erzielte hohe Selektivitat und Ausbeute an I-Methyl-Derivaten veranlaRten uns zur Untersuchung der Glykosidierung dieser Silyl-pyrimidine mit dem Ziel der Synthese von Thymin-nucleosiden 3 4). 1 ) 1 Mitteil: E.
