(Eiiigeg. 25. hlirz 1930.)In1 Jahre 1897 ist Paul E h r 1 i c h I ) , dessen Geburtsfag wir heute begehen, zuerst mit seiner bekannten ,,Sei t en ket t en theorie" her vo rget r e t en, der A nschauung, daD hochmolekulare, physiologisch aktive Stoffe wie die Antigene oder auch die Fermente ihre besondere Aktivitat der Ausbildung spezifischer Seitenketten verdanken, inittels welcher ihre Verankerung an den Substraten ihrer Reaktiori erfolge. Mit dieser einfachen Vorstelllung gewann man erstmals eine klare Einsicht in die chemischen Grun'dlagen vieler physiologisch wichtiger Vorgange, vor allem auf den Gebieten der Tnimunchemie und der Chemothterapie; ihre Giiltigkeit fur serologiwhe Reaktionen ist heute durch zahllose experimentelle Belege und Befunde gesichert. Auf dem Gebiete der enzymatischen Reaktionen hat man die Bestltigung der E h r 1 i c h schen Anschauung bisher vornehmlich durch Untersuchungen iiber das Schicksal von Enzymen nach ihrer Einfiihrung in den lebenden Organismus gesucht, welche ebenfalls zur Bildung spezifischer Abwehrstoffe, von ,,8ntienzymen", zu fiihren scheint. Wie fruchtbar die E h r 1 i c h sche Seitenkettentheorie nber auch f i r das Verstandnis der normalen enzymatischen Reaktionen, der Reaktionen zwisch.en Enzym und Substrat, sich erwiesen hat, das zu zeigen, sol1 der Inhalt nieiner heutigen Ausfiihrungen sein. Hier hat ja schon Emil F i s c h e r zur Erklarung der enzymatischen Spezifitat den anschaulichen und cheniisch durchdachten Vergleich von SchloD und Schliisael gebraucht.Die Enzymforschung ist diesen einfachen und klareii Vorstellungen nicht immer gefolgt, sie hat manchen Irrweg beschritten. Unter dem EinfluD der sich entwickelnden Kolloidchemie hat W. M. B a y 1 i s s 2) versucht, das Wesen der Enzymwirkung in erster Linie auf eine durch Adsorption bedingte Konzentrationserhilhung des Substrates an der Grenzflache der Enzymteilchen, also auf eine Oberflachenwirkung des Enzyms, nicht auf bestimmte chemische Affinitaten zuriickzufuhren: der Adsorptionsvorgang sollte die Voraussetzung bilden fur die nachfolgende chemische Reaktion, fiir deren Ausmai3 nur der jeweils adsorbierte Anteil der Reaktionspartner als maeebend angesehen wurde. Fur die Erklarung der fein auswahlenden enzymatischen Spezifitat beliei3 diese Anschauung nur geringen Spielraum; sie ist experimentell nie bewiesen worden, und man hat sie heute verlassen.Eine der wichtigsten Grundlagen fur die moderne, mehr chemische Analyse enzy ma t isch er Reakt ionen boten die Untersuchungen von L.
