The objective of this study was to use pyrosequencing of the 16S rRNA genes to describe the microbial diversity of bovine milk samples derived from clinically unaffected quarters across a range of somatic cell counts (SCC) values or from clinical mastitis, culture negative quarters. The obtained microbiota profiles were used to distinguish healthy, subclinically and clinically affected quarters. Two dairy farms were used for the collection of milk samples. A total of 177 samples were used. Fifty samples derived from healthy, culture negative quarters with a SCC of less than 20,000 cells/ml (group 1); 34 samples derived from healthy, culture negative quarters, with a SCC ranging from 21,000 to 50,000 cells/ml (group 2); 26 samples derived from healthy, culture negative quarters with a SCC greater than 50,000 cells/ml (group 3); 34 samples derived from healthy, culture positive quarters, with a SCC greater than 400,000 (group 4, subclinical); and 33 samples derived from clinical mastitis, culture negative quarters (group 5, clinical). Bacterial DNA was isolated from these samples and the 16S rRNA genes were individually amplified and pyrosequenced. All samples analyzed revealed great microbial diversity. Four bacterial genera were present in every sample obtained from healthy quarters (Faecalibacterium spp., unclassified Lachnospiraceae, Propionibacterium spp. and Aeribacillus spp.). Discriminant analysis models showed that samples derived from healthy quarters were easily discriminated based on their microbiota profiles from samples derived from clinical mastitis, culture negative quarters; that was also the case for samples obtained from different farms. Staphylococcus spp. and Streptococcus spp. were among the most prevalent genera in all groups while a general multivariable linear model revealed that Sphingobacterium and Streptococcus prevalences were associated with increased 10 log SCC. Conversely, Nocardiodes and Paenibacillus were negatively correlated, and a higher percentage of the genera was associated with a lower 10 log SCC.
Reproductive diseases are a great barrier to dairy production, as they diminish fertility and cause economic loss. In this context, endometritis plays an important role. Different diagnostic techniques exist for endometritis that vary in sensitivity and specificity. This work aimed to evaluate and compare ultrasonographic and histopathological examinations, and combinations thereof, for the diagnosis of endometritis by determining their sensitivity, specificity, and kappa agreement coefficients using endometrial cytology as the gold standard. We developed an adaptive reading score to perform histological examination of the uterus. In this study, 76 Holstein-Friesian cows, 21 to 47 d postpartum, without vaginal catarrh were submitted to gynecological examinations, including ultrasonographic examination, an endometrial cytology examination, and a uterine biopsy. Our results indicate that ultrasonography is a practical and effective diagnostic method, and a combination of the determination of intrauterine fluid and cervical diameter was the most efficient ultrasonographic method, with 50% sensitivity and 88% specificity. Histopathological examination was a useful diagnostic method that provided rich detail on endometrial inflammation, and allowed evaluation of different components of the tissue such as the epithelium, lamina propria, endometrial glands, and blood vessels; this method had 44% sensitivity and 92% specificity for diagnosis of endometritis.
The objective of this study was to evaluate the relationship between selected minerals' serum levels, energy metabolites, oxidative stress indicators, IL-8 and haptoglobin levels, and the potential for uterine diseases. Additionally, we investigated the effect of injectable trace mineral supplementation (ITMS) on metabolism, immune function, and animal health under field conditions involving a dairy herd with high milk production. The study was conducted in 1 dairy farm located near Ithaca, New York, with 270 multiparous cows were enrolled from October 3, 2012 until January 10, 2013. Cows were randomly allocated into 1 of 2 treatments groups: ITMS or control. Cows randomly assigned to the ITMS group received 2 injections of trace minerals at 230 and 260 d of gestation; each injection contained 300 mg of Zn, 50mg of Mn, 25mg of Se, and 75 mg of Cu. Retained placenta (RP) and metritis were diagnosed and treated by trained farm personnel. Clinical endometritis evaluation was performed by the investigators. Blood mineral levels, plasma nonesterified fatty acids and serum β-hydroxybutyrate concentrations, plasma IL-8 concentrations, serum haptoglobin concentration, and serum superoxidase dismutase and plasma glutathione peroxidase activities were measured at various time points before and after calving. Four groups of mixed general linear models were fitted to the data using MIXED procedure of SAS. Injectable trace mineral-supplemented cows had increased serum concentration of Cu, Se, and Zn. Conversely, ITMS did not affect energy metabolites or immune and oxidative stress parameters. Serum concentration of Ca, Cu, K, Mg, Mo, Ps, Pt, Se, and Zn varied according to days relative to parturition. Cows with RP had reduced serum concentrations of Ca, Mg, Mo, and Zn when compared with cows without RP. Cows affected with metritis had significantly lower serum concentrations of Ca, Mo, soluble P, total P, Se, and Zn than nonaffected cows. Serum concentration of Ca, Cu, Mo, and Zn were reduced in cows diagnosed with endometritis in comparison to nonaffected ones.
The objective of this study was to evaluate the efficacy of 3 vaccine formulations containing proteins (FimH, leukotoxin, and pyolysin), inactivated whole cells (Escherichia coli, Fusobacterium necrophorum, and Trueperella pyogenes), or both, in the prevention of postpartum uterine diseases. A randomized clinical trial was conducted at a commercial dairy farm; 800 heifers were assigned into 1 of 4 different treatment groups: control, vaccine 1 (bacterin and subunit proteins), vaccine 2 (bacterin), and vaccine 3 (recombinant subunit proteins), and each heifer received a subcutaneous injection of its respective treatment at 240 ± 3 and 270 ± 3 d of gestation. Vaccination significantly reduced the incidence of puerperal metritis when compared with control (9.1% vs. 14.9%, respectively; odds ratio 0.51). Additionally, vaccine 3 was found to reduce the incidence of puerperal metritis when compared with the control (8.0% vs. 14.9%, respectively; odds ratio 0.46). Reproduction was improved for metritic cows that were vaccinated, and the effect was stronger for cows that were treated with vaccine 3. In general, vaccination decreased the total vaginal bacterial load and decreased the vaginal load of F. necrophorum by 9 d in milk. Vaccination reduced the prevalence of puerperal metritis in the first lactation of dairy cows, leading to less metritic disease and improved reproduction.
We have shown in 2 independent studies that cows who received recombinant bovine interleukin-8 (rbIL-8) administered intrauterinely shortly after parturition have a significant and long-lasting increase in milk yield. In the present study, we hypothesized that the increased milk production associated with rbIL-8 treatment is a consequence of increased postpartum dry matter intake (DMI) and orchestrated homeorhetic changes that prioritize milk production. Cows were enrolled into 1 of 3 treatment groups: those assigned to the control group (CTR; n = 70) received an intrauterine (IU) administration of 500 mL of Dulbecco's phosphate-buffered saline (DPBS) solution and 1 mL of DPBS solution intravenously (IV; jugular vein), those assigned to the rbIL-8 IV group (rbIL8-IV, n = 70) received an IV injection of 167 μg of rbIL-8 and 500 mL of DPBS solution IU, and cows assigned to the rbIL-8 IU group (rbIL8-IU, n = 70) received an IU administration with 1,195 μg of rbIL-8 diluted in 499.5 mL of DPBS solution and 1 mL of DPBS solution IV. Animals were housed in a tiestall from calving to 30 d in milk (DIM) to measure DMI. Blood samples were collected daily from calving to 7 DIM and weekly until 28 DIM. Insulin resistance was evaluated using an intravenous glucose tolerance test and intravenous insulin challenge test (IVICT) in a subgroup of cows (n = 20/treatment) at 10 and 11 DIM, respectively. Additionally, liver biopsy samples were taken at 14 DIM from the same subgroup of cows to measure triglyceride levels and cell proliferation and apoptosis. Cows treated with rbIL8-IU produced more milk (CTR = 36.9 ± 1.5; rbIL8-IU = 38.5 ± 1.5; rbIL8-IV = 36.6 ± 1.5 kg/d), energy-corrected milk (CTR = 42.9 ± 0.9; rbIL8-IU = 46.1 ± 0.8; rbIL8-IV = 43.7 ± 0.9 kg/d), and fat-corrected milk (CTR = 44.3 ± 0.9; rbIL8-IU = 47.8 ± 0.9; rbIL8-IV = 45.2 ± 0.9 kg/d) yields when compared with CTR cows, and no differences were observed between rbIL8-IV and CTR cows. The administration of rbIL8-IU significantly increased DMI compared with CTR (CTR = 18.8 ± 0.3; rbIL8-IU = 19.9 ± 0.3; rbIL8-IV = 19.3 ± 0.3 kg/d). Recombinant bIL-8 treatment did not affect glucose, insulin, or fatty acids (i.e., IVICT only) concentrations or their area under the curve in response to an intravenous glucose tolerance test and IVICT when compared with CTR. Moreover, rbIL-8 treatment administered IU or IV increased liver triglyceride levels. Additionally, cows treated with rbIL8-IU tended to have lower odds of developing hyperketonemia (odds ratio = 0.46, 95% confidence interval: 0.19 to 1.10), lower odds of clinical ketosis and displaced abomasum combined (odds ratio = 0.17, 95% confidence interval: 0.03 to 0.89), and lower odds of diseases combined (odds ratio = 0.43, 95% confidence interval: 0.21 to 0.86) when compared with CTR. We conclude that the administration of rbIL8-IU increases DMI, milk production, fat-corrected milk, and energy-corrected milk while improving overall health during the postpartum period. The objective of this study was to evaluate the effect of prepartum intravaginal administration of an anti-Escherichia coli bacteriophage cocktail on uterine health and reproductive performance of lactating dairy cows. The study was conducted on a commercial dairy farm located near Ithaca, New York, from March to September 2012, and 107 cows were enrolled. The cows were divided randomly into 2 groups (treatment and control); treatment group cows were submitted to 3 intravaginal administrations of 20 mL of a bacteriophage cocktail including 10 different phages in a lysate with titration of approximately 10(9) plaque-forming units, on d 230, 260, and 275 of gestation. Control cows were not treated with a placebo, and physical examination and sample collection was conducted equally for both groups. Vaginal swabs were collected at d 230, 260, and 275 d of gestation, cervical swabs were collected at 3±1 d in milk, and blood was collected at 275 d of gestation for nonesterified fatty acids and interleukin (IL)-6 serum level measurements and at 10 DIM for β-hydroxybutyrate and IL-6 serum level measurements. The bacteriophage cocktail significantly increased the incidence of retained placenta and metritis. Swab samples were cultured for E. coli and no difference was found between treatment and control groups. The lysate contained 6×10(9) IU of lipopolysaccharide/mL and could have caused prepartum overstimulation of toll-like receptor-4 (TLR4)-chemokine pathway, leading to a weaker immune response. However, IL-6 was not affected by treatment, although sample size was insufficient to prove a treatment effect on the suppression of the chemokine cascade and innate immune system. Bacteriophage treatment had an excellent bactericidal effect in previous in vitro trials; however, special attention must be given to production of the cocktail to obtain a purified solution free of E. coli bacterial products and fragments, particularly lipopolysaccharide.
RESUMO.-Este estudo avaliou a função renal de fêmeas ovinas hígidas da raça Santa Inês, criadas no Estado de São Paulo, com o objetivo de estabelecer valores de referência, além de determinar possíveis variações dos valores de seus parâmetros causados por fatores etários. Foram determinados os valores de ureia, creatinina, sódio e potássio séricos e urinários, e os valores de GGT urinário de 128 animais, divididos em 4 grupos de acordo com a idade. Foi realizada, também a urinálise das 128 amostras de urina. Não foi observada diferença significativa dos valores de densidade entre todos os grupos. Entretanto, constatou-se aumento de valores de ureia, creatinina, sódio e diminuição de proteínas e GGT urinários nos grupos 2, 3 e 4 que eram mais velhos do que o grupo 1. Em relação aos parâmetros séricos, observou-se diferença significativa dos valores de ureia e creatinina entre os grupos, pois uréia e creatinina aumentaram e potássio diminuiu nos animais do grupo 2, 3 e 4 que eram mais velhos. Avaliando-se os resultados obtidos para os exames da função renal, verificou-se que os valores médios na urina da densidade foi de 1,03±0,01; pH 7,71±1,35; ureia em mg/dL foi 1342±1085; de creatinina 42,39±23,36; de proteínas totais em g/dL foi de 5,61±2,88; GGT em U/L foi de 1,44±1,31; sódio e potássio em mmol/L foi de 15,93±20 This study evaluated kidney function of healthy Santa Inês ewes, bred in São Paulo State. The aim of this paper was to establish standard reference values and stipulate value variation caused by age group factor. We determined seric and urinary levels of urea, creatinine, sodium and potassium, and urinary GGT from 128 ewes, divided into 4 groups according to age. Also urinalysis was made in 128 urine samples. No significant statistical difference was found regarding density values between all groups, although there was an increase in urea values, creatinine and sodium, and an decrease in urinary proteins and GGT in group 2, 3 and 4 older than group1. In relation to the seric parameters, there was significative statistical difference in urea and creatinine values between the groups, as an increase in urea and creatinine. Potassium decreased in the older groups 2, 3 and 4. Evaluating the results obtained for renal function, we found that the mean values of density were 1.03±0.01, pH 7.1±1.35, urea (mg/dL) 1342±1085, creatinin 42.39±23,36, total protein (g/dL) 5.61±2.88, GGT (U/L) 1.44±1.31, sodium and potassium (mmol/L) were 15.93±20.13 and 240±117 respectively. In the serum medium values were: urea (mg/dL) 32.33±19.81, creatinine 1.22±0.20, sodium and potassium (mmol/L) 145.7±8.8 and 5.5±1.30 respectively.
