The complex cis-[Pt(Me2C=NOH)2Cl2], 1, isomerizes into trans-[Pt(Me2C=NOH)2Cl2], 2, upon heating in the solid phase at 140 °C. Isomerization of 1 into 2 was also performed in acetonitrile or acetone solutions, containing catalytic amounts of Me2C=NOH; in the DMF-DMSO mixture, a 2-propanone oxime is displaced by DMSO in formation of cis-, 3, and ?rans-[Pt(Me2C=NOH)(DMSO)Cl2], 4. An excess of dimethyl sulfoxide results in the formation of the well-known complex cis-[Pt(DMSO)2Cl2]. Complex 4 has also been obtained from K[Pt(DMSO)-CI3] and Me2C=NOH in water, and complex 3 results from the solid-phase thermal isomerization of 4. The oxidation of 1 and 2 by molecular chlorine in CHCI3 or in MeN02 produces cis-, 5, and ira/u-[Pt(Me2C=NOH)2-CI4], 6. Complex 6 is relatively stable in a water-acetone mixture, but complex 5 in this mixture at 20-25 °C is converted within 3 h into a product containing an unusual redox coupling of 2-propanone oximes, [Pt(N(=0)-CMe2ONCMe2)Cl2], 7. All the complexes were characterized by elemental analysis, IR spectra, and , 13C, and 195Pt NMR spectra; X-ray structure analysis was performed for 7. The compound crystallizes in the orthorhombic space group Pn2\a. The unit cell dimensions are a = 8.860 (3), b = 8.022 (2), c = 15.744 (4) Á, V -1119.0 (6) A3, Z = 4, and pca^= 2.43 g cm™1. Complex 1 is decomposed under X-ray irradiation, but crystallographic parameters have been determined (monoclinic, space group ¿2/6, a = 11.074 (3), 6 = 16.114 (4), c = 8.177 (1) A, 7 = 111.76 (1)°, V= 1355.0 (5) A3, Z = 4).

Despite the well-established fact that maternal care plays a pivotal role in the offspring development, little is known about the effects of disruption of maternal care early in life on the development of this behavior in the offspring. Using brief repeated maternal separation (45 min/day on postnatal Days 3–6), which represents a model of early life stress, we found behavioral changes in adult female mice offspring. The decrease in home cage exploratory behavior (both pup-directed and nonpup-directed) was revealed later in adulthood without changes in maternal care level. Maternal separation coupled with pain exposure caused by subcutaneous saline injection procedure had a cumulative resulting effect, which was manifested in the decreased level of nursing associated with licking-grooming in adult females. The behavioral changes found in adult female offspring could be triggered by identified changes in the behavior of their mothers, while alterations of the level of histone H3 acetylation in the neonatal brain were not detected. Histone deacetylase inhibitor sodium valproate was used in order to study the possibility of preventing the effects of early life stress through involvement of epigenetic mechanisms. Despite the increase in the level of histone H3 acetylation in the neonatal brain caused by valproate, its behavioral effects were barely detectable. These effects were reflected in prevention of the reduction of nursing associated with licking-grooming induced by maternal separation, accompanied by pain exposure. The data are discussed in terms of the possible application to the studies of mechanisms underlying long-term effects of human early life trauma.

Environmental factors acting during the neonatal period affected the formation of phenotype of adult animals via epigenetic modifications of the genome, e.g. histone acetylation. We studied the effects of blockage of histone deacetylases with sodium valproate on week 1 of life on the behavioral pattern of adult male 129Sv mice. The results of behavioral tests attest to long-term changes in emotional, but not cognitive pattern of adult animals after sodium valproate administration, which manifested in increased anxiety of animals.
