The presence of carbohydrates, especially glucomannan, causes porang tuber to be important in some industries including food, cosmetics, medicines, and other industries. Porang has four growing periods (GP) in its life cycle. The GP1 until GP3 is the vegetative phase and GP4 is the generative phase. The tubers used as industrial materials generally were derived from GP2 and GP3 plants. The study aimed to analyse carbohydrate components in porang tubers from GP2 and GP3 plants using Liquid Chromatography Mass Spectrometry (LC-MS) method. The tubers produced from plants at GP2 and GP3 were extracted by maceration method using methanol solvent (pa) for 1 h. Liquid extracts obtained was further identified for the levels of carbohydrate components using LC-MS technique. The carbohydrate components identified with LC-MS were selected for quantifications. LC-MS analysis showed that both GP2 and GP3 tubers contained 67 types of chemical compounds. In the GP2 tubers 8 types of carbohydrate components were detected. Whereas in the GP3 tuber in addition to the 8 types of carbohydrate components that are the same as ini the GP2 tuber there is also 1 other type of component, namely trehalose. Glucomannan is the highest levels of the carbohydrate component in GP2 and GP3. However, glucomannan levels in GP2 tuber (58, 845.54µg/g) were lower than those in GP3 tuber (99, 436.85µg/g). Therefore, GP3 tubers are better than GP2 to be used as industrial materials.
AbstrakTujuan dari penelitian ini adalah untuk untuk menentukan perlakuan terbaik dari kombinasi proporsi buah : sukrosa dan lama ekstraksi osmosis pada sari buah tapus (Curculigo Latifolia Dryand) terhadap parameter fisikokimiawi. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 2 faktor yaitu faktor 1 adalah proporsi buah:sukrosa sebesar 1:0.5, 1:1, 1:1,5 dan faktor 2 adalah lama ekstraksi osmosis yaitu 12, 24, 35 jam. Parameter pengamatan yang digunakan adalah pH, total padatan terlarut, vitamin C, dan total gula. Hasil menunjukkan bahwa semakin tinggi proporsi penambahan buah dan semakin lama ekstraksi osmosis maka terjadi peningkatan terhadap parameter derajat keasaman (pH), peningkatan terlarut, dan total gula. Ekosistem padang lamun merupakan habitat bagi ikan dan biota perairan lainnya. Berbagai jenis penyu juga menjadikan daerah padang lamun sebagai daerah mencari makan, khususnya di perairan Pulau Lemukutan, Kalimantan Barat. Tutupan lamun, khususnya jenis Thalassia hempricii di perairan Dusun Karang Utara, Pulau Lemukutan belum diketahui. Data mengenai tutupan lamun ini akan dijadikan sebagai acuan dalam pengelolaan wilayah perairan tersebut. Penelitian dilakukan pada 4 stasiun di perairan Dusun Karang Utara, Pulau Lemukutan, Kalimantan Barat. Stasiun pengamatan memiliki panjang 50 m tegak lurus terhadap garis pantai. Setiap stasiun dibagi menjadi 5 plot transek. Pemilihan plot transek dilakukan dengan metode purposive random sampling. Transek kuadrat yang digunakan adalah berukuran 1 x 1 m dengan setiap pixelnya berukuran 20 cm x 20 cm. Hasil nilai rata-rata persentase tutupan lamun pada stasiun 1, 2, 3 dan 4 adalah 2,68%; 2,34%, 11,4% dan 9,8% secara berurutan. Kondisi persentase tutupan lamun Thalassia hempricii di Perairan dusun Karang Utara, Pulau Lemukutan ini termasuk dalam kategori sedikit. Parameter fisika-kimia air laut di Perairan Dusun Karang Utara, Pulau Lemukutan adalah arus 0,2 m/s, suhu 30,5 o C, salinitas 32 o /oo , kedalaman 0,5-1 m, dan pH 8,5.
Triggerfish (Abalistes stellatus) is a non-commercial fish. This species is a small triggerfish genus found in West Kalimantan waters. The aim of this research was to determine the levels of macronutrient such as protein, lipid, ash and water content of the fish. The levels of macronutrients were determined using the methods recommended by Association of Official Analytical Chemists (AOAC). The fish were fairly high in macronutrient protein (19,75%) while the lipid content was 5,94%. Water and ash content were 78.87% and 1,57%, respectively. This result can be concluded that triggerfish is rich in food value, especially protein source for human dietary.
