Aim. The purpose of this in vitro study was to assess the antimicrobial efficacy of Biopure MTAD against E. faecalis in contaminated root canals. Materials and Methods. Forty-two single rooted extracted human teeth were inoculated with E. faecalis and incubated for four weeks. The samples were divided in two control and five experimental groups irrigated with 1.5% sodium hypochlorite solution (NaOCl); 3% NaOCl; BioPure MTAD; 1.5% NaOCl/17% EDTA; or 3% NaOCl/17% EDTA. After a one-week incubation, complete disinfection was confirmed by the absence of turbidity in the incubation media. Dentin shavings were taken from samples with no turbidity to verify whether E. faecalis was present in dentin tubules. Results were analyzed statistically using Fisher's exact test, with the level of significance set at P < 0.05. Results. Statistical analysis of the data obtained at Day 7 and after dentin shaving analysis showed that BioPure MTAD had significantly greater antibacterial activity than 1.5% NaOCl, 1.5% NaOCl/17% EDTA and 3% NaOCl/17% EDTA. No significant difference was detected between MTAD and 3% NaOCl. Conclusions. These findings suggest that BioPure MTAD possesses superior bactericidal activity compared with NaOCl and EDTA against E. faecalis.
Background:Cytotoxicity of root canal irrigants is important due to their close contact with host tissues. The aim of this study was to assess the cytotoxic effect of NaOCl 3%, Chx 2%, and MTAD on rat periodontal ligament fibroblasts, at 0.1 and 100 µl/mL, using WST-1 colorimetric method.
Material/Method:Rat ligamental fibroblasts were exposed to the irrigants and their viability was assessed after 1, 24, 48, and 72 h. The measurements were determined using WST-1 assay, using a micro ELISA reader.
Results:At 100 ml/L all 3 irrigants were strongly cytotoxic, although CHX was less so than NaOCl and MTAD. At the 0.1 ml/L concentration, NaOCl and MTAD were only moderately cytotoxic, whereas Chx was highly deleterious to cell viability at all time points. There was a significant influence of the dilution rate of the substance, because the odds ratio for cell viability being over 50% was increased 51 times between the 100 ml/L and 0.1 ml/L dilutions.
Conclusions:It seems that irrigating solutions should be used at lower concentrations to enhance cell viability.
The main goal of endodontic therapy is the three-dimen- sional obturation of the endodontic space. Tendencies for ideal root canal preparation and its disinfection are com- promised if the canal is not completely obturated. The pur- pose of this study is to determine the percentage of the vol- ume of the spaces in the root canal of the tooth. obturated with different materials by microcomputer-tomography. In this study, 40 extracted frontal teeth were included. The teeth were divided into two groups according to the mate- rial used for obturation. The teeth after root canal prepara- tion according to the standard technique were obturated with Gutta-Flow and Thermafill. The teeth were analyzed by micro-CT. Statistical analysis showed that root canals obturated with GuttaFlow showed a higher percentage of gaps inside the obturation material, while root canals ob- turated with Thermafill have a higher percentage of spaces between the obturation material and the root canal wall.
