The size of the polypeptide chains of the hemocyanin from the Dungeness crab, Cancer magister, has been determined in a number of ways: (1) Sedimentation equilibrium in concentrated guanidine hydrochloride (Gdn-HCl), with or without reducing agents, indicates a homogeneous or nearly homogeneous material with M-x ~77,000. ( 2) Gel filtration in 6 m Gdn • HC1 yields a single peak, corresponding to a molecular weight of about 70,000. (3) Quantitative aminoterminal analysis shows 1 aspartic end group/80,000 molecular weight. (4) High-resolution sodium dodecyl sulfate polyacrylamide gel electrophoresis indicates that there are two chains, of molecular weights about 76,000 and 83,000 daltons, present in roughly equal quantities.
