Isolate UFV-101 of Bacillus cereus was selected in previous studies for promoting growth inducing resistance in plants. In a previous study, supernatant from cultures of the microorganism in a liquid medium was found to induce resistance in tomato foliage against the pathogens Pseudomonas syringae pv. tomato, Xanthomonas vesicatoria, Alternaria solani and Corynespora cassiicola. In the present work the microorganism was grown in a minimal medium for 48 h and the cells precipitated for centrifugation. The supernatant was concentrated by lyophilization, dialyzed in a 12 kDa cutoff point membrane and fractioned in column containing Sephadex G25 balanced in PBS buffer. The fractions corresponding to a protein peak were applied to tomato seedlings. After four days leaflets were collected and inoculated with the pathogen. C. cassiicola. The numbers of lesions produced by the pathogen on leaflets exposed to the bacterial supernatant were similar to those exposed to acibenzolar-S-methyl but fewer than in those treated with water. It is concluded that the supernatant contained protein which induced resistance in the tomato leaves against C. cassiicola.
Purpose
Banana is an important tropical fruit with high demand in the market. The ripe fruits are less resistant to transport making logistics difficult. Moreover, as a climacteric fruit, it has a short post-harvest shelf life. Edible coatings/films, including active substances, have been used as an alternative for preserving fruits and vegetables during post-harvest period. The purpose of this study was to evaluate the incorporating clove essential oil on the properties of cassava starch films and their effect on the post-harvest quality of different banana varieties.
Design/methodology/approach
Cassava starch films incorporating clove essential oil were developed and the films were characterized with respect to moisture, thickness, solubility, water vapor permeability (WVP), biodegradability, color and in vitro antifungal activity. Cultivars such as Prata-Anã, Grand Nine, BRS Tropical and BRS Conquista were coated with cassava starch, cassava starch film with clove essential oil (CSEO) and polyvinyl polychloride (PVC). The quality of fruits was monitored during eight storage days using mass loss, total soluble solids content (TSS) and titratable acidity (TTA).
Findings
Incorporation of clove essential oil significantly increased film thickness, reduced moisture content, solubility and WVP (p < 0.05) and did not affect the biodegradability and color of the films. The essential oil incorporated films showed antifungal activity against the fungi Colletotrichum gloeosporioides and Colletotrichum musae, but not against the yeast Saccharomyces bourladii. CSEO and PVC coating were more efficient in reducing the mass loss, SS content and TTA of the coated fruits in all varieties studied. Both CSEO and PVC coatings improve the quality attributes such as TSS and TTA and reduced mass loss, of the banana varieties such as Prata-Anã, Grand Nine, BRS Tropical and BRS Conquista during storage.
Originality/value
The active film with essential oil showed antifungal activity and essential oil can be incorporated into other food systems. This study approaches a new possibility of film coating with essential oil for a banana that showed minimum weight loss and satisfactory quality and increased shelf life. This film coating demonstrates biodegradable characteristics that could be eco-friendly and sustainable to consumers.
RESUMOO presente trabalho teve como objetivo avaliar o progresso da mancha-bacteriana do pimentão a partir de quatro quantidades iniciais de mudas infectadas (0; 3,3; 10 e 100%) associado à aplicação semanal de oxicloreto de cobre (2,4 g de i.a./L de água) ou não. Foram conduzidos cinco ensaios, em condições de campo, em Seropédica (RJ), entre junho de 1996 e setembro de 1999. Quantificou-se a intensidade de doença, regularmente, por meio de estimativas visuais da severidade da doença (área foliar lesionada e desfolha) e calculou-se a área abaixo da curva de progresso da doença normalizada (AACPD/ número de dias). Estas foram submetidas à Análise de Variância (p=0,01) e à análise de regressão linear em função do logaritmo da quantidade inicial de mudas infectadas. A redução do inóculo inicial teve efeito direto na quantidade final de doença, e de forma mais marcante sob condições mais propícias ao desenvolvimento da doença. A eficiência do oxicloreto de cobre foi variável e na maioria das vezes ineficiente, principalmente nos períodos mais propícios a epidemia, com redução de sua eficiência pela elevação da temperatura.

Dois antagonistas selecionados para o biocontrole da vassoura-de-bruxa do cacaueiro foram avaliados quanto à capacidade em ativar mecanismos de defesa de plantas contra patógenos. Para tanto, mudas seminais de cacaueiro "comum" foram cultivadas em casa-de-vegetação por 30 dias e expostas aos antagonistas aplicados a mudas de cacaueiro por atomização, individualmente e em associação. O primeiro par de folhas das mudas dos diferentes tratamentos foi coletado aos dois, quatro, 12 e 24 dias após a exposição aos antagonistas. Foi quantificada a atividade de peroxidases, polifenoloxidases, quitinases e beta-1,3-glucanases no material coletado. Observou-se um aumento na atividade de peroxidases e polifenoloxidases nos primeiros dias após a exposição das mudas, especialmente ao isolado Ac26. Não foi observado efeito aditivo ou sinergístico nas mudas expostas aos dois isolados simultaneamente.
