This study investigates in-vitro activities of phytochemicals in Callistemon citrinus against multiresistant foodborne pathogens, alpha-glucosidase enzyme and MCF-7 cancer cell line. Assays were prepared with lyophilized extracts to determine antioxidant capacity, inhibition of a-glucosidase enzyme and growth of foodborne bacteria. Annexin-V detection kit was used for apoptosis detection and FT-IR spectroscopy to confirm structural and functional groups of phytochemicals. Cytotoxicity of the extracts against MCF-7 cells was monitored with xCELLigence Real-Time Cell Analyser. The result from FT-IR analysis gave a peak at 3295 cm À1 wavenumber, confirming the presence of O-H alcohol functional group. FT-IR analysis also showed the presence of different functional groups such as carboxylic acids, aromatics, alkanes, alcohols, aliphatic amines, alkenes and amine groups in the extracts. Callistemon exhibited strong antioxidant capacities with EC 50 values of 0.474 ± 0.03 and 0.787 ± 0.15 mL sample/g of 2,2-diphenyl-1-picrylhydeazyl (DPPH) from leaf and flower extracts, respectively. Growth inhibition of most gram-positive foodborne bacteria by phytochemicals from flower extract appeared more promising as an alternative antimicrobial agent for food preservation. IC 50 value of 3.69 ± 0.61 lg/mL obtained from leaf extract showed its inhibitory potential against a-glucosidase enzyme for managing diabetes type-2. A dose response obtained from real-time monitoring with xCELLigence system indicated higher cytotoxicity of the extracts against MCF-7 cell line at !200 lg/mL concentrations within 24 h of incubation. The versatility of phytochemicals in Callistemon observed in this study signifies its potential for enhancing feed or food functionality, moderating blood glucose level and inhibiting the growth of foodborne pathogens or invasive carcinoma in man.
Meme kanseri, en yaygın küresel malignitedir ve kansere bağlı ölümlerin ikinci önde gelen nedenidir. Meme kanserinin erken teşhis edilmesi, farklı tedavi yöntemleri ile tedavi başarı oranını yükseltmektedir. Ancak ilerleyen evrelerde ise hem tedavi başarı oranı düşmektedir hem de güncel tedavi yöntemlerinden kemoterapi veya radyoterapinin uygulanmasının hastaya yan etkileri de olabilmektedir. Bu nedenle kişiye özel, yan etkinin olmadığı tedavi modellerini bulmak gerekir. Eksozomlar doğal nanopartiküller olup, aynı zamanda hücreler arası iletişimde önemli roller oynayan, çok çeşitli lipidler, proteinler, metabolitler, RNA'lar ve DNA'lar içeren küçük hücre dışı veziküller olarak bilinirler. Bu çalışmanın amacı, adipoz doku kaynaklı mezenkimal kök hücrelerden presipitasyon yöntemi ile elde edilen eksozomların (ADEkso) insan meme kanseri hücre hattı, MCF-7, ve meme epitel hücre hattı, MCF10A, üzerinde farklı konsantrasyonlarda Anneksin V/PI metodu kullanılarak apoptotik etkilerinin araştırılmasıdır. Erken apoptotik, geç apoptotik, canlı ve ölü hücreler, her iki hücre hattında 24, 48 ve 72 saat boyunca farklı 40x1010, 20x1010 ve 10x1010 AD-Ekso/mL konsantrasyonları ile muamele edilerek MCF-7 gruplarında toplam yüzde total apoptotik etkileri 24 saat için 40x1010/AD-Ekso/mL’de %58.817±4.2, 48 saat için 0x1010/AD-Ekso/mL 'de %56.37±9 ve 72 saat için 20x1010 AD- Ekso/mL'de %70.9±4.1'dir. AD-Ekso verilen MCF-7 hücre grupları hem kendi aralarında hem de AD-Ekso uygulanmayan MCF-7 hücre grupları ve MCF10A hücre grupları ile karşılaştırıldığında zaman ve doza bağlı olarak aradaki fark istatistiksel olarak anlamlı bulunmuştur (p˂0.05). MCF10A ve MCF-7 kontrol grupları değerlendirilmiş ve anlamlı bir fark gözlenmemiştir (p˃0.05). Sonuç olarak, eksozomlar kanser hücrelerinde apoptozu indükleyerek kanser tedavisinde potansiyel kapasiteye sahip olabilir
Bu çalışmada, karbon tetraklorür ve tiyoasetamid (CCl 4 ve TAA) modellerinde alanin aminotransferaz (ALT), aspartat aminotransferaz (AST), albümin, trigliserit ve kolesterol gibi biyokimyasal parametreler ve histopatolojik özellikler değerlendirilerek doz ve zamana bağlı olarak daha etkin modelin ortaya çıkarılması amaçlanmıştır. Yöntem: Her bir model için ratlar 3 gruba ayrılmıştır ve intraperitoneal (i.p.) olarak CCl 4 (0.5 ml/kg, 1.0 ml/kg, 2.0 ml/kg) ve TAA (100 mg/kg, 200 mg/kg, 300 mg/kg) 4, 6 ve 8 hafta boyunca hafta da üç kez enjeksiyon yapılmıştır. Bulgular: Biyokimyasal araştırmalar sonucunda ALT ve AST değerleri, sadece 0,5 ml CCL 4 6. ve 8. hafta gruplarında kontrol gruplarına göre istatistiksel olarak anlamlı fark göstermiştir (p<0.05). Diğer biyokimyasal parametrelerin değerleri kalan gruplar arasında anlamlı farklılık göstermemiştir (p>0.05). Histopatolojik sonuçlara bakıldığında karaciğer dokusunda, kontrol gruplarının her ikisinde de karaciğer, nor-mal histolojik yapısını göstermiştir. Diğer bütün gruplarda, artan zaman ve doza bağlı olarak her iki modelde göze çarpan hepatofibrotik değişiklikler gözlemlenmiştir. Sonuç: Doz ve zamana bağlı olarak sekizinci haftaya ulaşan gruplarda siroz geliştiği gözlemlenmiştir. Bu sonuçlar hepatofibrotik çalışmalar için yarayışlı birer referans olabilecektir.
Introduction P-element induced wimpy testis-like 2 (Piwil2) is in the Piwi gene family. Piwil2 has important roles in the self-renewal mechanism of stem cell induction and progression of numerous types of human malignancies such as lung, breast, colon, prostate, and cervical cancers. Glutathione S-transferase (GST) acts as detoxification in cancer metabolism. This study aimed to investigate the effects of the stem cell protein Piwil2 on MCF10A and MCF-7 at the GST activity levels. Materials/Methods MCF-7/Piwil2 and MCF10A/Piwil2, transfected with a plasmid carrying the Piwil2 gene, and non-transfected MCF-7 and MCF10A were cultured in a complete DMEM/F12 medium. GST A1 and P1 activity was determined in these cell lines using as substrates CDNB, EA respectively. Results According to experimental results, GST P1 activity decreased in the MCF-7/Piwil2 cells as compared with the non-transfected MCF-7 cells, however, MCF-7/Piwil2 cells demonstrated increases in GST A1 (total GST) activity. The statistically significant differences were found for the comparison of non-transfected MCF-7 and MCF-7/Piwil2 (p<0,0001), for GST enzyme activities by using CDNB and EA as substrates. These results were the same for the MCF10A cell line. Discussion It is shown for the first time that transfection studies may affect GST activity at the cellular mechanism level. The study contributes to determining the effect of transfection on GST isoenzymes and also how the Piwil2 gene may affect GST activity in the stem cell line.
