The consumer risks of jamu, Indonesian traditional herbal medicines, was assessed focussing on the presence of alkenylbenzene containing botanical ingredients. Twenty-three out of 25 samples contained alkenylbenzenes at levels ranging from 3.8 to 440 μg/kg, with methyleugenol being the most frequently encountered alkenylbenzene. The estimated daily intake (EDI) resulting from jamu consumption was estimated to amount to 0.2-171 μg/kg bw/day for individual alkenylbenzenes, to 0.9-203 μg/kg bw/day when adding up all alkenylbenzenes detected, and to 0.9-551 μg/kg bw/day when expressed in methyleugenol equivalents using interim relative potency (REP) factors. The margin of exposure (MOE) values obtained were generally <10,000 indicating a priority for risk management when assuming daily consumption during a lifetime. Using Haber's rule it was estimated that two weeks consumption of these jamu only once would not raise a concern (MOE >10,000). However, when considering use for two weeks every year during a lifetime, 5 samples still raise a concern. It is concluded that the consumption of alkenylbenzene containing jamu can be of concern especially when consumed on a daily basis for longer periods of time on a regular basis.
The COD (Chemical Oxygen Demand) analysis method based on the Indonesian National Standard (SNI) is a revision of SNI 06-6989.2-2004, Water and wastewater - Part 2: Method of testing for chemical oxygen demand (COD) with closed reflux spectrophotometrically. This SNI uses references from international standard methods, namely Standard Methods for the Examination of Water and Wastewater, 21st Edition, editor of LS Clesceri, AE Greenberg, AD Eaton, APHA, AWWA and WEF, Washington DC, 2005, Methods 5220 D (Closed Reflux, Colorimetric Methods). The purpose of this study was to validate the total method for testing chemical oxygen demand (COD) in water and wastewater with the reduction of Cr2O7 2- spectrophotometrically in the range of COD values of 100 mg / L to 900 mg / L measurements were made at a wavelength of 600 nm. and COD values less than or equal to 90 mg / L measurements were made at a wavelength of 420 nm. The results showed that the method SNI 06-6898.2- 2009 has good validation results including the r results obtained from the calibration curve equation of 0.998, a precision of 1.82%, an accuracy of 98.25% and LOD and LOQ of 12.27 mg / L and 25, 61 mg / L.
Sumber daya alam yang dimiliki Indonesia akhir-akhir ini semakin banyak dieksploitasi sebagai bahan obat-obatan baik untuk farmasi maupun untuk kepentingan pertanian. Salah satu tanaman tersebut adalah tanaman kenanga yang bisa dimanfaatkan minyak atsirinya sebagai bahan antibakteri pada produk hand sanitizer gel. Minyak atsiri kenanga diperoleh melalui proses destilasi uap bunga kenanga, kemudian diuji identifikasi fitokimia lalu fisiknya dan kualitasnya meliputi bobot jenis, indeks bias, dan bilangan ester sesuai SNI 06-3949-1995 lalu diuji potensi aktivitas antibakterinya. hand sanitizer gel minyak atsiri bunga kenanga dibuat sebanyak 4 formula. dengan konsentrasi minyak atsiri bunga kenanga 5%; 2,5%; 1,25% dan 0,5%. Data yang diperoleh meliputi fisik, viskositas, pH, daya sebar, daya lekat, organoleptik dan uji aktivitas antibakteri. Hasil penelitian menunjukkan bahwa hand sanitizer gel dengan minyak atsiri bunga kenanga memiliki aktivitas antibakteri terhadap bakteri S. aureus dan E. coli. Dari uji daya hambat disimpulkan bahwa hand sanitizer gel dengan konsentrasi minyak atsiri bunga kenanga 5% paling optimal yang memiliki pH 5,75; viskositas 4120; daya sebar 5,5 cm2 dan daya lekat 18,33 detik.
Food diversification is a way to strengthen national food security, for example by reducing the dependence on wheat flour as a main raw material of various food products through the use of cassava flour. Cassava flour, a rich source of carbohydrates, can be used in the making of flakes. In this study, the Virgin coconut oil (VCO) waste flour was added in cassava flour to produce gluten-free cassava flour flakes with high dietary fiber contents. The six different formulas of flakes were used in this study with addition of 0% (control), 10%, 20%, 30%, 40% and 50% coconut waste flour, respectively. Then, the chemical and microbiological characteristics of the flakes were measured. Based on the proximate data, both flours have a good quality. The microbiological analysis shows cassava flour and coconut waste flour have good sanitation and food safety. Results showed that flakes with 80% cassava flour and 20% coconut waste flour are gluten-free and contain 2.52% water, 2.27% ash, 14.40% fat, 4.50% protein, 76.31% carbohydrate, 8.56% dietary fiber. Therefore, it can be said that cassava flour flakes can serve as gluten-free and rich dietary fiber ready-to-eat food.
Dengue is a severe health problem, especially in under-developing countries like Indonesia. The government and the community have made various efforts. However, the current programs are focusing on controlling disease vector. Medical treatment so far only treats the symptoms. On the other hand, the development of antiviral compounds has been dominated by synthetic compounds which may have serious side effects. The potential of natural compounds in Asia is still largely unexplored. Dengue virus nonstructural protein 5 (NS5) is highly conserved in all serotypes. This study aims to screen several potential natural compounds to be developed as specific antiviral compounds targeting DENV NS5. This research conducted by using in silico approaches including data mining, molecular docking, and 2D-3D visualization. About 170 compounds from 88 organisms analyzed to get the top nine compounds with the highest affinity against NS5 protein. The top nine compounds are from the halichondramide, trigocherriolide, quinadoline, tryptoquivaline, and zingiberene groups. The docking results showed that these potential compounds have an affinity of about 67-100% higher than ribavirin. The visualization results show that these compounds are grouped into three different binding sites. They are RdRP, MTase, and the junction. Compounds that bind in RdRP are thought to interfere with the process of RNA viral synthesis, compounds that bind to MTase are thought to disrupt the viral RNA capping process, while compounds that bind between the two domains are thought to interfere NS5 structural integrity. The compound with the highest affinity is halishigamide A. Halishigamide A is known to have antimicrobial properties, but the action mechanism is still unknown. Compared to the control drug, the halishigamide A tends to have more dynamic movement and interaction to the target protein’s binding site due to its residues’ chemicals motive. Together, these results showed the potency of natural products as specific antiviral compounds targeting DENV NS5.
