Kokristalisasi merupakan salah satu teknik untuk meningkatkan kelarutan zat aktif farmasi yang 70% kelarutannya tidak larut dalam air. Kokristal terdiri dari zat aktif dan bahan tambahan yang disebut dengan koformer yang dihubungkan oleh interaksi non kovalen. Pengaruh pembentukan kokristalisasi zat aktif dengan menggunakan koformer nikotinamid dalam upaya meningkatkan kelarutannya dikemukakan dalam kajian pustaka ini. Nikotinamid berperan sebagai koformer larut air yang paling stabil. Zat aktif yang berhasil dibuat kokristal menggunakan metode solvent evaporation yaitu katekin, artesunat, ketoprofen, diflunisal, ibuprofen, simvastatin dan prulifloxacin, menggunakan metode slurry diantaranya karbamazepin dan didanosin, sedangkan dengan metode liquid assisted grinding yaitu zaltoprofen dan furosemid. Peningkatan kelarutan zat aktif yang menggunakan solvent evaporation hingga 6 kali, menggunakan metode slurry hingga 1,86 kali, dan menggunakan metode liquid assisted grinding hingga 166.666,67 kali. Peningkatan kelarutan ini disebabkan adanya ikatan hidrogen yang terbentuk antara zat aktif dan koformer, serta karakterisasi yang menunjukan kokristal berhasil terbentuk, yaitu adanya fase kristal baru. Dalam review artikel dapat disimpulkan bahwa zat aktif yang telah dibentuk kokristal menggunakan koformer nikotinamid dengan beberapa metode yang berbeda menunjukkan peningkatan kelarutan hingga 166.666,67 kali.
