Bras. 18(1):9-15, jan./mar. 1998 9 RESUMO.-[Espessamento intimal difuso das artérias na calcinose enzoótica dos ovinos.] Foram feitos estudos morfométrico, imunoistoquímico e ultra-estrutural do espessamento intimal difuso (DIT) das artérias de 7 ovinos com sinais clínicos de calcinose enzoótica espontânea causada pela ingestão da planta Nierembergia veitchii. As lesões caracterizavam-se por deposição de sais de cálcio na média como placas e estrias que, com frequência faziam saliência para a luz, criando rugosidades e irregularidades da íntima. Não foram observadas calcificações na artéria pulmonar e no sistema venoso. Microscopicamente, as calcificações das artéri-as restringiam-se quase que exclusivamente à média. Na imunoistoquimica, foi verificada α-actina nas células da mé-dia e nas do espessamento intimal. Receptores para 1,25(OH) 2 D 3 foram evidenciados nos núcleos das células musculares da média, da íntima e das células endoteliais. As aná-lises morfométricas em microscopia ótica revelaram, nas artérias, DIT irregularmente distribuido sem relação com a intensidade dos processos de calcificação subjacente nem com a espessura da média remanescente. A morfometria das alterações ultra-estruturais das células musculares lisas da mé-dia e da íntima espessada, mostrou que nessas últimas foi verificado aumento de até 318% na fração volumétrica das organelas de síntese em detrimento dos elementos contráteis, Morphometric, immunohistochemical and ultrastructural studies were carried out on the diffuse intimal thickening (DIT) in arteries of 7 sheep with clinical signs of naturally occurring enzootic calcinosis due to ingestion of the plant Nierembergia veitchii. Arterial lesions consisted of medial deposition of calcium salts and DIT. Calcification of the intima was rare, mild and located near the elastic lamina. By immunohistochemistry α-actin was detected in cells of the media and in cells forming the intimal thickening. Receptors for 1,25(OH) 2 vitamin D 3 were detected in nuclei of intimal, medial and endothelial cells. DIT was irregularly distributed and was neither proportionally related to the intensity of the underlying mineralization area nor to the thickening of the remaining media. Ultrastructural morphometry in smooth muscle cells (SMCs) of the media and thickened intima revealed, in the latter, an increase of 318% in the volumetric fraction of those organelles involved in synthesis and a proportional decrease in contractile elements when compared to normal values of media cells. There were histological and ultrastructural evidences of modification of SMCs and their migration to the intima, where they proliferated causing DIT. It was concluded that DIT is a consistent component of arteriosclerotic lesions in N. veitchii induced calcinosis of sheep and that the predominant cell in this process is the SMCs originated from its predecessors of the media. It is suggested that the inducing factor for the arterial changes is 1,25(OH) 2 D 3 present in N. veitchii.
RESUMOPfaffia glomerata (Spreng.) Pedersen é uma das espécies de ginseng brasileiro usada popularmente como planta medicinal. Com este trabalho objetivou-se avaliar o efeito de doses (15, 30, 45 e 60 g.L -1 ) e fontes de carboidratos (sacarose, frutose, glicose, maltose e lactose) no crescimento dessas plantas cultivadas in vitro. O delineamento experimental foi o inteiramente casualizado com 15 repetições por tratamento. A parcela experimental consistiu de um tubo de ensaio contendo 10 mL de meio MS e um segmento nodal de 1,0 cm de comprimento e sem folhas. Os parâmetros avaliados aos 41 dias após a inoculação foram o número de brotações, o número total de segmentos nodais por plântula, a altura média das brotações, a altura da maior brotação, a massa fresca e seca da parte aérea e a massa seca das raízes. A melhor fonte de carbono para o crescimento in vitro da P. glomerata foi a sacarose, e a lactose é inapropriada. O número total de segmentos nodais, altura média das brotações, altura da maior brotação, massa fresca e seca da parte aérea e a massa seca de raízes aumentam pelo acréscimo das doses de sacarose, e em 30, 45 e 60 g.L -1 foram significativamente maiores do que aquelas proporcionadas pelas demais fontes de carbono. Pedersen] IN VITRO CULTURED PLANTSABSTRACT Pfaffia glomerata (Spreng.) Pedersen is one of the brazilian ginseng species used as medicinal plant. The aim of this work was to evaluate the effect of carbohydrate concentration (15, 30, 45, and 60 g.L -1 ) and source (sucrose, fructose, glucose, maltose, and lactose) on the growth of plants grown in vitro. An entirely random statistical design with 15 replications per treatment was used. The experimental unity consisted of a test tube containing 10 mL of MS medium and 1-node segment of 1,0cm length and without leaves. After 41 days of the inoculation the growth parameters analyzed were sprout number, total number of nodal segment per seedling, average length of sprouts, length of the highest sprout, fresh and dry matter weight of aerial parts, and root dry matter weight. The best carbohydrate source for growth of P. glomerata in vitro cultured plants was sucrose, and lactose was found unsuitable. The total number of nodal segment per seedling, average length of sprouts, length of the highest sprout, fresh and dry matter weight of aerial parts, and root dry matter weight increase by sucrose concentrations, and at 30, 45 and 60 g.L -1 all parameters were significantly higher than those obtained by other carbon sources.
O ginseng brasileiro [Pfaffia glomerata (Spreng.) Pedersen] apresenta propriedades medicinais marcantes e, por isso, atualmente, é largamente explorado de forma extrativista, tanto por parte dos coletores de ervas como pela indústria farmacêutica. Este trabalho objetivou caracterizar o efeito da variação isolada da concentração de N e P do meio MS no crescimento de plantas de P. glomerata cultivadas in vitro. Segmentos nodais de 1,0cm de comprimento e sem folhas, de plantas já estabelecidas in vitro, foram cultivados em meio MS contendo cinco concentrações (0, 25, 50, 100 e 150% da concentração padrão do meio de cultura MS) de nitrogênio ou fósforo. Aos 15 dias após a inoculação (DAI), o número de raízes e o percentual de enraizamento são maiores na concentração de N e P equivalentes a 50% daquela do meio de cultura MS. Aos 40 DAI, o crescimento em altura das brotações, número de segmentos nodais, índice de área foliar, número de folhas, matéria seca de raízes, da parte aérea e total da planta é maior na concentração de N e P, em média, próxima a 80% daquela do meio de cultura MS.
A system for growing in liquid medium whole plants of Valeriana glechomifolia, endemic to southern Brazil and capable of accumulating bioactive valepotriates, is described. Murashige and Skoog (MS) and Gamborg B5 (B5) media (1.0·, 0.3· and 0.1· strength) without phytohormones were evaluated after four weeks of culture in relation to growth and valepotriate yield. Plants grown in 1.0· MS displayed greatest growth and valepotriate yields and the study of the light condition showed that plants grown under light and dark had similar weight increase and maximum valepotriate yield, 27.2 mg/g DW and 25.0 mg/g DW, respectively. Valtrate was the most abundant valepotriate, followed by acevaltrate and didrovaltrate.
