The consumption of fructose has increased in children and adolescents and is partially responsible for the high incidence of metabolic diseases. The lifestyle during postnatal development can result in altered metabolic programming, thereby impairing the reproductive system and fertility during adulthood. Therefore, the aim of this study was to evaluate the effect of a high-fructose diet in the male reproductive system of pubertal and adult rats. Male Wistar rats (30 d old) were assigned to four different groups: Fr30, which received fructose (20%) in water for 30 d and were euthanized at postnatal day (PND) 60; Re-Fr30, which received fructose (20%) for 30 d and were euthanized at PND 120; and two control groups C30 and Re-C30, which received water ad libitum and were euthanized at PND 60 and 120, respectively. Fructose induced an increase in abnormal seminiferous tubules with epithelial vacuoles, degeneration, and immature cells in the lumen. Moreover, Fr30 rats showed altered spermatogenesis and daily sperm production (DSP), as well as increased serum testosterone concentrations. After discontinuing high-fructose consumption, DSP and sperm number decreased significantly. We observed tissue remodeling in the epididymis, with a reduction in stromal and epithelial compartments that might have influenced sperm motility. Therefore, we concluded that fructose intake in peripubertal rats led to changes in the reproductive system observed both during puberty and adulthood.
Nas últimas décadas, o consumo elevado de açúcar tem sido associado ao aumento da prevalência de doenças metabólicas, cardiovasculares e na fertilidade. Enquanto a ingestão exagerada de sacarose pode induzir a danos oxidativos nos tecidos, o uso de antioxidantes poderia proteger as células contra o estresse oxidativo. O pterostilbeno, um componente antioxidante dos mirtilos, demonstrou exercer benefícios terapêuticos em várias condições. OBJETIVOS: No presente estudo, investigamos os efeitos histopatológicos da ingestão de solução de sacarose (40%) e tratamento com pterostilbeno (40 mg/kg) nos testículos de ratos Wistar. Nossa hipótese é que o pterostilbeno poderia normalizar as mudanças causadas pelo consumo de açúcar. METODOLOGIA: Ratos Wistar machos foram distribuídos em quatro grupos: grupo controle, pterostilbeno, sacarose e grupo sacarose + pterostilbeno. Inicialmente, os ratos foram alimentados com solução de sacarose ou água por 140 dias. Em seguida, os ratos foram tratados com pterostilbeno por 45 dias consecutivos. RESULTADOS: O tratamento apenas com pterostilbeno não levou a alterações histopatológicas nos testículos. O percentual de túbulos seminíferos anormais foi maior no grupo tratado com sacarose que nos demais grupos, com redução de células de Leydig. As alterações histopatológicas mais observadas foram: presença de células imaturas na luz do túbulo seminífero e vacúolos no epitélio tubular (ambos grupo sacarose); células imaturas na luz e degeneração tubular (ambos no grupo sacarose em associação com pterostilbeno). Embora a presença de vacúolos seja evidente, não foi encontrado alterações no número de células de Sertoli. CONCLUSÃO: Em conclusão, o consumo de sacarose levou a alterações histopatológicas no tecido testicular, como células na luz e vacúolos. No entanto, o uso de um antioxidante concomitante levou a alterações histológicas como degeneração tubular, sendo um fator prejudicial.
INTRODUÇÃO:A leishmaniose é causada por protozoários flagelados do gênero Leishmania e é responsável por um espectro de desordens em animais e humanos. O curso da doença é determinado pela resposta imunológica e a espécie de parasito envolvida. Os macrófagos representam uma peça fundamental na infecção como células em potencial para a destruição do parasito através da fagocitose e também representam as células hospedeiras para a multiplicação do parasito. OBJETIVOS: Dessa forma, este trabalho tem como objetivo fazer uma revisão bibliográfica sobre o papel dos macrófagos na leishmaniose. METODOLOGIA: Este trabalho foi realizado através de uma pesquisa bibliográfica narrativa, realizada na plataforma Pubmed. RESULTADOS: Espécies de Leishmania interagir com receptores celulares do hospedeiro, como o CR1, CR3 e, manose-fucose. Após a ativação, os macrófagos aumentam a produção de óxido nítrico (NO) e radicais livres. A estimulação por citocinas pró-inflamatórias, em particular o Interferon-γ, estimula e intensifica a função efetora dos macrófagos contra patógenos intracelulares, alterando-se classicamente para o fenótipo M1. Macrófagos da classe M2 podem prejudicar a imunidade protetora contra protozoários e estão relacionados à cronicidade das doenças infecciosas. Existem estudos demonstrando que espécies de Leishmania, como a L. amazonensis são capazes de inibir a ativação de macrófagos ou resistir aos mecanismos leishmanicidas. A Leishmania é capaz de regular a maturação do fagossomo, a fim de tornar o fagossomo mais hospitaleiro para o crescimento e multiplicação das amastigotas. Muitos fatores de sobrevivência da Leishmania estão envolvidos na formação do fagossomo e na reprogramação do macrófago para promover a infecção. CONCLUSÃO: O macrófago possui um papel fundamental na infecção, no entanto, o parasito possui muitos recursos de escape para manter sua sobrevivência e disseminação. O estudo desses mecanismos permite uma melhor compreensão da patogenia da doença.
