Dermatological disorders related to pigmentation result in tenuous hyper or hypopigmentation Cosmetic and pharmaceutical products containing depigmenting substances are used in the treatment of patients who have high pigmentation disorders, such as melasma or chloasma, post-inflammatory hyperpigmentation, senile lentigo and ephelides. Skin lightening agents are not yet totally effective or safe and therefore intensive research for the discovery of new agents is continuous. Enzyme inhibitors involved in melanogenesis, such as tyrosinase, have been discovered in Asian countries, including those isolated from plant extracts. The Brazilian flora has the highest species diversity in the world, and the chemical, pharmacological and cosmetic potential for the discovery of new skin whitening agents is in proportion with this biodiversity. For these reasons, 25 aqueous and 24 organic extracts obtained from 19 plants native to the Amazon rain forest and to the Atlantic forest, belonging to 11 different families, were evaluated as tyrosinase inhibitors. Nine out of 49 extracts showed inhibitory activity in the screening process. The 50% inhibitory activity (IA 50 ) was calculated, revealing that the most active extracts were the organic extracts from the leaves and stem of Ruprechtia sp. (IA 50 33.76 mg.mL -1 ) and the organic extract from the aerial organs of Rapanea parviflora (IA 50 64.19 mg.mL -1 ).Uniterms: Tyrosinase. Melanin. Plant extract/in vitro evaluation. Produtos cosméticos e farmacêuticos com atividade despigmentante são utilizados para o tratamento de pacientes que apresentam distúrbios de hiperpigmentação, tais como melasma ou cloasma, hiperpigmentação pós-inflamatória, lentigem senil e efélides. Os despigmentantes atualmente utilizados não são totalmente eficazes ou seguros, razão pela qual há intensa pesquisa, principalmente em países asiáticos, com a finalidade de se obter novos agentes com esta ação, em especial inibidores de enzimas envolvidas na melanogênese, como a tirosinase. Considerando-se que algumas substâncias obtidas de plantas apresentam essa atividade, a flora brasileira constitui-se uma fonte potencial de obtenção de novos despigmentantes. Por essa razão, 25 extratos aquosos e 24 orgânicos obtidos de 19 plantas da Floresta Amazônica e Mata Atlântica, provenientes de 11 diferentes famílias, foram avaliados quanto à atividade de inibição da tirosinase. Do total de 49 extratos testados, 9 mostraram atividade. Os valores de concentração da atividade inibitória 50% (AI 50%), foram calculados e o mais ativo foi o extrato orgânico das folhas e caule de Ruprechtia sp. RESUMO -Lesões desmielinizantes induzidas pelo gliotóxico brometo de etídio (BE) têm sido estudadas com o objetivo de permitir a compreensão do limitado processo de re p a ro mielínico no sistema nerv o s o central, bem como avaliar estratégias terapêuticas no sentido de acelerar a re c o n s t rução das bainhas de mielina perdidas. Muito embora estudos eletrofisiológicos correlacionando situações de desmielinização e remielinização experimental sejam bem estabelecidos, os efeitos comportamentais não têm sido adequadamente investigados. Neste estudo, foram analisadas ultra-estruturalmente as lesões desmielinizantes e a atividade locomotora de ratos submetidos à indução focal de desmielinização pelo modelo do BE na s u p e rfície ventral do tronco encefálico, mediante observação de sua movimentação e controle motor durante a travessia de uma trave elevada de madeira (beam walking test). Foi observada a ocorrência de deficiências locomotoras até 31 dias pós-injeção de BE, constatando-se ainda que a subseqüente remielinização estava relacionada com o retorno da função perdida.PALAVRAS-CHAVE: brometo de etídio, desmielinização, ratos, remielinização, teste da passarela elevada. Evaluation of locomotor activity after a local induction of toxic demyelination in the brainstem of Wistar ratsABSTRACT -Ethidium-bromide (EB) -induced lesions have been used to investigate the incomplete re m y elination in the central nervous system, as well as to evaluate therapeutic strategies to accelerate the re c o n s t ruction of the lost myelin sheaths. Although many electrophysiologic studies were perf o rmed in situations of experimental demyelination and remyelination, their behavioural effects have not been pro p e r l y analyzed. In this study, we investigated ultrastructurally the EB -demyelinating lesions as well as the locomotor activity of rats during the beam walking test after a focal induction of demyelination using the EB model in the ventral surface of the brainstem. The main objective of the present study was to investigate whether male rats that receive a hypercaloric diet only during the juvenile period (Postnatal Day [PND] 23 to PND65) and a normocaloric diet thereafter exhibit changes in reproductive aspects in adulthood. Body weight gain during the juvenile period and adulthood and the morphology of adipose tissue in adulthood were recorded to determine the effects of the hypercaloric diet. We also evaluated the time of balanopreputial separation, serum testosterone levels, and sexual behavior. Obesity and being overweight are related to an inflammatory process. Therefore, we also examined tumor necrosis factor α (TNF-α) levels in adulthood. Compared with the control group that received only the normocaloric diet, the hypercaloric diet group exhibited an increase in body weight on PND65 and PND90, impairment of sexual behavior, an increase in the number of large adipocytes relative to small cells, a decrease in testosterone levels, and an increase in plasma TNF-α levels. The hypercaloric diet during the juvenile period caused the rats to become overweight, although they were subsequently fed a normocaloric diet until adulthood. The hypercaloric diet resulted in reproductive impairment, reflected by a reduction of sexual behavior, low serum testosterone levels, a reduction of sexual organ weight, and an increase in TNF-α levels.
Prenatal undernutrition impairs copulatory behavior and increases the tendency to become obese/overweight, which also reduces sexual behavior. Re-feeding rats prenatally undernourished with a normocaloric diet can restore their physiological conditions and copulatory behavior. Thus, the present study investigated whether a hypercaloric diet that is administered in rats during the juvenile period prevents sexual impairments that are caused by maternal food restriction and the tendency to become overweight/obese. Female rats were prenatally fed a 40% restricted diet from gestational day 2 to 18. The pups received a hypercaloric diet from postnatal day (PND) 23 to PND65 (food restricted hypercaloric [FRH] group) or laboratory chow (food restricted control [FRC] group). Pups from non-food-restricted dams received laboratory chow during the entire experiment (non-food-restricted [NFR] group). During the juvenile period and adulthood, body weight gain was evaluated weekly. The day of balanopreputial separation, sexual behavior, sexual organ weight, hypodermal adiposity, striatal dopamine and serotonin, serum testosterone, and tumor necrosis factor α (TNF-α) were evaluated. The FRH group exhibited an increase in body weight on PND58 and PND65. The FRC group exhibited an increase in the latency to the first mount and intromission and an increase in serum TNF-α levels but a reduction of dopaminergic activity. The hypercaloric diet reversed all of these effects but increased adiposity. We concluded that the hypercaloric diet administered during the juvenile period attenuated reproductive impairments that were induced by maternal food restriction through increases in the energy expenditure but not the tendency to become overweight/obese.
Pyrethroid insecticides are extensively used for pest control around the house, flea prevention for pets, and plant sprays for the home and in agriculture. Deltamethrin (DTM) is a Type II pyrethroid insecticide used to control a variety of insects in agriculture and domestic environments. The present study investigated the possible anxiogenic effects of DTM (1, 3, and 10 mg/kg) in rats using behavioral and neurochemical methods. We assessed general locomotor activity and behavior in the elevated plus maze and open field test. Striatal and hippocampal neurotransmitter and metabolite levels were also measured. DTM (i) reduced locomotion and rearing frequency, (ii) slightly increased the duration of immobility, (iii) reduced the time engaged in social interaction, (iv) reduced the percentage of entries into and time spent on the open arms of the elevated plus maze, (v) reduced the number of center crossings in the elevated plus maze, (vi) Striatal and hippocampal neurotransmitter and metabolite levels were also measured. DTM (i) reduced locomotion and rearing frequency, (ii) slightly increased the duration of immobility, (iii) reduced the time engaged in social interaction, (iv) reduced the percentage of entries into and time spent on the open arms of the elevated plus maze, (v) reduced the number of center crossings in the elevated plus maze, (vi) increased striatal serotonin neurotransmitter and its metabolite, and (vii) did not alter motor coordination on the rotarod, grooming duration in the open field test, rectal temperature, or hippocampal neurotransmitter levels. These data suggest that DTM at the present doses and under these experimental conditions presented a similar profile to that of anxiogenic drugs, unrelated with the increased serotonin neurotransmission.Keywords: Deltamethrin. Deltametrina (DTM) é um inseticida piretróide tipo II usado para controlar uma variedade de insetos na agricultura e ambientes domésticos. O presente estudo investigou os possíveis efeitos ansiogênicos de DTM (1, 3 e 10 mg/kg) em ratos, utilizando métodos comportamentais e neuroquímicos. Foi avaliada a atividade locomotora geral e comportamento no labirinto em cruz elevado e teste de campo aberto. Os níveis de neurotransmissores e metabólitos no estriado e hipocampo também foram mensurados.
