Die ersten den Antipyrin-Rest enthaltenden Ather wurden von L. Know und A . Pschorrl) synthetisiert. Der Methyl-und der Athylather des 4-Hydroxy-antipyrins wiesen gute antipyretische und analgetische Eigenschaften auf. Der Austausch der Alkyl-gegen Arylrestez) ergab schwer losliche Verbindungen. Um Derivate gunstigerer Loslichkeitsverhaltnisse fur den pharmakologischen Versuch bereitzustellen, haben wir nunmehr Ather des Antipyryl-(4)-methanols und des Antipyryl-(4)-atjhanols der allgemeinen Formel A . CH,. 0 . R und As CH, * CH2 . O . R gewonnen? worin A der Antipyryl-(4)-Rest CH, * C-C-I 1 C,H, CH3.N C=O '%N,' und R aliphatische und aromatische Reste sind. L41s Ausgangsstoffe geeignet erschienen Antipyrin-(4)-carbino13) und Antipyrin-(4)-athano14). Die Versuche, aus erstgenanntem Stoff die gesuchten Ather eu gewinnen, schlugen fehl. Meist fie1 das Bis-antipyryl-methan an, das auch K. Bodendorf3) bei der Behandlung von Antipyrin-(4)-carbinol mit Mineralsauren oder in der Hitze erhielt : 2 A-CHZ-OH + A-CHZ-A + H2O + CH2O Wir benutzten daher die von J . Madinavetia5) beschriebene Umsetzung von quartaren Ammoniumbasen mit Alkalialkoholaten, wobei neben Trialkylaniin der entsprechende h h e r entsteht : [R-CH,-X(CH3),]+ 9-+ Me-0-R' --f R-CH,O-R' f MeX + (CH3),N Diese Reaktion wurde spMer von T. A . Geisman und A. A m e n 6 ) auf Atherderivate des Indols angewandt. Nach Ansicht der Verfasser verhalt sich das a m der Mannich-Base herstellbare quartare Salz des Gramins wie das hypothetische 3-Chlormethyl-indol : mit Na-alkoholat entsteht der entspreohende Ather, mit Alkalicyanid das Nitril bzv. nach dessen Verseifung die 3-Indolessigsiiure und mit Alkaliacetat das (3-Acetoxymethy1)-*) Herrn Prof. Dr. E'. Barnunn zum 60. Geburtstag freundschaftlich gewidmet.
