The catalytic domain of clostridial neurotoxins is a substrate of tyrosine-specific protein kinases. The functional role of tyrosine phosphorylation and also the number and location of its (their) phosphorylation site(s) are yet elusive. We have used the recombinant catalytic domain of botulinum neurotoxin E (BoNT E) to examine these issues. Bacterially expressed and purified BoNT E catalytic domain was fully active, and was phosphorylated in vitro by the tyrosine-specific kinase Src. Tyrosine phosphorylation of the catalytic domain increased the protein thermal stability without affecting its proteolytic activity. Covalent modification of the endopeptidase promoted a disorder-to-order transition, as evidenced by the 35% increment of the alpha-helical content, which resulted in a 4 degrees C increase of its denaturation temperature. Site-directed replacement of tyrosine at position 67 completely abolished phosphate incorporation by Src. Constitutively unphosphorylated endopeptidase mutants exhibited functional properties virtually identical to those displayed by the nonphosphorylated wild-type catalytic domain. These findings indicate the presence of a single phosphorylation site in the catalytic domain of clostridial neurotoxins, and that its covalent modification primarily modulates the protein thermostability.
Botulinum neurotoxin A (BoNT A) is a substrate of the Src family of tyrosine kinases. Here, we report that the BoNT A light chain (LC) is phosphorylated in the tyrosine-71 located at N-terminus. Covalent modification of this residue notably increases the thermal stability of the endopeptidase activity, without affecting its catalytic efficacy. Similarly, mutation of this residue specifically affected the protein stability but not its endopeptidase function. Fusion of the Tat-translocating domain to the N-terminus of the enzyme produced a cell permeable, functional enzyme, as evidenced by immunocytochemistry and by the cleavage of cytosolic SNAP25 in intact PC12 cells. Noteworthy, truncation of cellular SNAP25 was reduced in cells when the Src kinase activity was inhibited with a specific antagonist, implying that tyrosine phosphorylation of BoNT A LC modulates the in vivo proteolytic activity of the neurotoxin. Taken together, these findings substantiate the tenet that tyrosine phosphorylation of BoNT A LC could be an important modulatory strategy of the neurotoxin stability and suggest that the phosphorylated neurotoxin may be a relevant molecule in vivo.
El estudio de la atenuación lingüística, en tanto fenómeno pragmático, necesita criterios consensuados de identificación que ayuden al analista en sus investigaciones y facilite el desarrollo de estudios contrastivos. En esta línea, este artículo se propone reflexionar acerca del fenómeno de la atenuación y, más concretamente, sobre los criterios que la bibliografía ha empleado para su reconocimiento. Dichos criterios podrían resumirse grosso modo en la vaguedad del contenido semántico, la mitigación de la fuerza ilocutiva, la desfocalización del papel de los participantes en la interacción y el reconocimiento de la función o finalidad de la atenuación. Además de estos criterios, se ofrecen tres pruebas que pueden emplearse para validar el análisis: la prueba de la ausencia, la prueba de la conmutación y la prueba de la solidaridad. Estas pruebas, junto con un estudio pormenorizado del contexto interactivo general (el género discursivo) y el contexto interactivo concreto en el que aparece el mecanismo atenuante pueden resultar claves para reforzar los criterios anteriormente señalados y facilitar el reconocimiento de la atenuación.

Estos estudios han contribuido positivamente a la descripción de la actividad atenuadora, ayudando a reforzar la hipótesis ampliamente refrendada por la comunidad científica de que atenuación y cortesía no son equivalentes (Hernández Flores, 2002, p. 57; 2004, pp. 98-99; Bravo, 2002, p. 101;Albelda y Briz, 2010 y Briz, 2012a. Sorprende, sin embargo, que puedan hallarse pocas investigaciones (como el trabajo de Martinovski, 2006) que aborden el estudio de la atenuación en uno de los géneros que se señalan como prototipo del registro formal (oral), los juicios orales. En relación con lo anterior, este trabajo quiere contribuir a la extensión el estudio de las funciones y los mecanismos de atenuación a otros géneros y para ello es indispensable partir de una metodología adecuada. De ahí que el objetivo de este trabajo sea valorar en qué medida la propuesta teórico-metodológica de Briz y Albelda (2013) para el proyecto ES.VAR. ATENUACIÓN es aplicable a otros géneros y, más concretamente, a los juicios orales o vistas.
