The fungal genus Fonsecaea contains etiological agents of human chromoblastomycosis, a (sub)tropical, (sub)cutaneous implantation disease caused by plant contact. The invasive potential differs significantly between species. Infections by Fonsecaea monophora are believed to originate from the environment and the species has been reported as one of the main causative agents of the disease, but also of cases of primary brain infection. The epidemiology of the disease has not been fully elucidated and questions related to its infection route and virulence are still to be clarified. The environmental species Fonsecaea erecta was isolated from organic material and living plants in endemic areas for chromoblastomycosis in Brazil. The present paper describes Agrobacteriumtumefaciens-mediated transformation (AMT) of the environmental species F. erecta and the pathogenic species F. monophora. We propose the use of Agrobacterium transformation for future gene function studies related to Fonsecaea virulence and pathogenicity. We evaluated the co-cultivation ratios 1:1, 10:1 and 100:1 (Agrobacterium:conidia) at 28 °C during 72 h. pAD1625 and pCAMDsRed plasmids were inserted into both species. Confirmation of transformation was realized by hph gene amplification and Southern blot determined the amount of foreign DNA integrated into the genome. In order to evaluate a potential link between environmental and clinical strains, we obtained red fluorescent transformants after pCAMDsRed insertion. We observed by confocal fluorescence microscopy that both F. monophora and F. erecta were able to colonize the palm Bactris gasipaes, penetrating the epidermis. These results contribute to understanding the ability of Fonsecaea species to adapt to different environmental and host conditions.
En la presente investigación se aisló una bacteria termófila productora de enzima amilasa, de los géiseres de Calientes, Candarave (Tacna-Perú), la cual por análisis de la secuencia del gen ARNr 16S presentó un 99 % de identidad con Bacillus licheniformis. Para determinar la capacidad amilolítica se evaluó en medio sólido presentando un halo de hidrólisis de 29,5 mm de diámetro a las 72 horas. En la evaluación de la producción de amilasa se obtuvo una concentración máxima de 1410,57µg/mL de azúcares reductores a inicios de la fase estacionaria a 54,6 horas de incubación a 60 °C y pH 7. El extracto amilolítico tuvo un grado de purificación de 2,3 veces con sulfato de amonio, actuando a una temperatura óptima de 58 °C con una actividad de 10,7 U/mL ya un pH óptimo de 6,7 con una actividad de 8,3 U/mL, además la velocidad máxima (Vm) fue de 229,5µg de almidón hidrolizado por ml de enzima por minuto y el valor de la constante de Michaelis (Km) fue de 8,7 mg de almidón por mL enzima.
Para la investigación, los almidones de papa (Solanum tuberosum), camote (lpomoea batatas) y yuca (Manihot esculenta) fueron hidrolizados por la enzima amilasa obtenida por cultivo líquido de la bacteria termófila, designada como Bacillus licheniformis BA-3 y aislada de los géiseres de Calientes, Candarave, Tacna. Se obtuvo el extracto crudo, el cual fue filtrado, centrifugado y precipitado con sulfato de amonio al 60 % y dializado por 24 horas en refrigeración a - 5 °C. Asimismo, la bacteria termófila utilizada presentó el 99 % de identidad mediante la secuenciación del gen ARNr 16S. Se evaluó la concentración de azúcares reductores de los almidones y, a partir de ello, se determinó la cinética de velocidad enzimática propuesta por Michaelis-Menten, a concentraciones de 1 %, 1.5 %, 2 %, 2.5 % y 3 % de solución de cada almidón con 5 μl de enzima amilasa a temperatura de 50 °C y pH 7. Se obtuvieron las constantes de Michaelis- Menten (Km) de 0.4225 g/ml, 0.0024 g/ml y 0.1263 g/ml para Solanum tuberosum, lpomoea batatas y Manihot esculenta, respectivamente. De igual forma, se obtuvo valores de velocidad máxima (Vmax) de 0.00091 g/ml *min, 0.00011 g/ml*min y 0.00099 g/ml*min para cada almidón estudiado. Se concluyó que existe mayor afinidad de la enzima producida por Bacillus licheniformis BA-3 con el almidón de lpomoea batatas por ser el menor valor de Km obtenido.
El presente trabajo estudia las proteasas alcalinas producidas por dos bacterias termófilas designadas como BP-2 y BP-4. Estas fueron aisladas de sedimentos de los géiseres de Candarave, mostrando capacidad proteolítica en medio sólido. La cepa BP-2 mostró un diámetro de 46 mm del halo de hidrólisis a 50 °C, mientras que la cepa BP-4 mostró un diámetro de 85 mm del halo de hidrólisis a 60°C. Las bacterias seleccionadas BP-2 y BP-4 degradaron 2,9 g/L de caseína en un tiempo estimado de 51 horas de incubación y 1,5 g/L de caseína en 52 horas de incubación, respectivamente (concentración inicial de 10 g/L de caseína). La máxima actividad proteolítica se alcanzó al final de la fase logarítmica y comienzo de la fase estacionaria en las dos bacterias. Los extractos proteolíticos producidos por las bacterias BP-2 y BP-4 actuaron a una temperatura óptima estimada de 57°C y 64°C, con un pH óptimo alcalino de 7,7 y 8,0 respectivamente. Del análisis de las secuencias del gen ARNr 16S de cada bacteria se determinó que la bacteria BP-2 tiene 99% de identidad con la especie Bacillus licheniformis y la bacteria BP-4 tiene un 99% de identidad con la especie Geobacillus thermaparaffinivorans.
