Background Laccases are multicopper enzymes that oxidize a wide range of aromatic and non-aromatic compounds in the presence of oxygen. The majority of industrially relevant laccases are derived from fungi and are produced in eukaryotic expression systems such as Pichia pastoris and Saccharomyces cerevisiae. Bacterial laccases for research purposes are mostly produced intracellularly in Escherichia coli, but secretory expression systems are needed for future applications. Bacterial laccases from Streptomyces spp. are of interest for potential industrial applications because of their lignin degrading activities. Results In this study, we expressed small laccases genes from Streptomyces coelicolor, Streptomyces viridosporus and Amycolatopsis 75iv2 with their native signal sequences in Gram-positive Bacillus subtilis and Streptomyces lividans host organisms. The extracellular activities of ScLac, SvLac and AmLac expressed in S. lividans reached 1950 ± 99 U/l, 812 ± 57 U/l and 12 ± 1 U/l in the presence of copper supplementation. The secretion of the small laccases was irrespective of the copper supplementation; however, activities upon reconstitution with copper after expression were significantly lower, indicating the importance of copper during laccase production. The production of small laccases in B. subtilis resulted in extracellular activity that was significantly lower than in S. lividans. Unexpectedly, AmLac and ScLac were secreted without their native signal sequences in B. subtilis, indicating that B. subtilis secretes some heterologous proteins via an unknown pathway. Conclusions Small laccases from S. coelicolor, S. viridosporus and Amycolatopsis 75iv2 were secreted in both Gram-positive expression hosts B. subtilis and S. lividans, but the extracellular activities were significantly higher in the latter.

Expression of the endochitinase encoding ech42 gene of the mycoparasite Trichoderma atroviride is subject to control by several environmental signals, including derepression by carbon starvation. In order to identify promoter areas involved in control by this condition, we prepared fusions of several mutant forms of the ech42 promoter to the Aspergillus niger goxA gene as a reporter. Removal of a 130-bp fragment comprising a binding site for the carbon catabolite repressor Cre1, an AGGGG element and three separate binding sites identical and highly similar, respectively, to those for the Aspergillus nidulans regulator of conidiation BrlA resulted in a three-fold increase in derepression under carbon starvation. A truncation of the promoter to 196 bp, which removed all of the observed DNA binding motifs, resulted in five-fold derepression. In vitro protein-DNA binding analyses showed that only the BrlA-like sites, but neither the AGGGG element nor the Cre1 binding site, bound proteins from cell-free extracts from carbon-starved mycelia of T. atroviride. Thus this study identifies a new regulator of chitinase gene expression in Trichoderma, a BrlA-like binding motif.

Chitosan was cross-linked to an osmium redox polymer, poly(4-vinylpyridine) osmium bipyridyl [PVP-Os-(bpy) 2 -Cl], to form PVP-Os-(bpy) 2 -Cl/chitosan composite known to make a porous and hydrophilic film with an enzyme. In this work we demonstrate such a composite is a useful trestle, for hosting various sugars oxidizing enzymes to construct biosensors. Glucose sensing ability has been proven with the following glucose oxidizing redox enzymes; Aspergillus niger glucose oxidase (AnGOX), Myriococcum thermophilum cellobiose dehydrogenase, (MtCDH), glycosylated Agaricus meleagris pyranose dehydrogenase (gAmPDH), fragmented deglycosylated Agaricus meleagris pyranose dehydrogenase (fdgAmPDH), and Aspergillus sp. glucose dehydrogenase (AspGDH), as well as recombinant Glomerella cingulata glucose dehydrogenase (rGcGDH).
