SummaryActomyosin stress fibers (SFs) enable cells to exert traction on planar extracellular matrices (ECMs) by tensing focal adhesions (FAs) at the cell-ECM interface. Although it is widely appreciated that the spatial and temporal distribution of these tensile forces play key roles in polarity, motility, fate choice, and other defining cell behaviors, virtually nothing is known about how an individual SF quantitatively contributes to tensile loads borne by specific molecules within associated FAs. We address this key open question by using femtosecond laser ablation to sever single SFs in cells while tracking tension across vinculin using a molecular optical sensor. We show that disruption of a single SF reduces tension across vinculin in FAs located throughout the cell, with enriched vinculin tension reduction in FAs oriented parallel to the targeted SF. Remarkably, however, some subpopulations of FAs exhibit enhanced vinculin tension upon SF irradiation and undergo dramatic, unexpected transitions between tension-enhanced and tension-reduced states. These changes depend strongly on the location of the severed SF, consistent with our earlier finding that different SF pools are regulated by distinct myosin activators. We critically discuss the extent to which these measurements can be interpreted in terms of whole-FA tension and traction and propose a model that relates SF tension to adhesive loads and cell shape stability. These studies represent the most direct and highresolution intracellular measurements of SF contributions to tension on specific FA proteins to date and offer a new paradigm for investigating regulation of adhesive complexes by cytoskeletal force.

While is widely acknowledged that nonmuscle myosin II (NMMII) enables stress fibers (SFs) to generate traction forces against the extracellular matrix, little is known about how specific NMMII isoforms and functional domains contribute to SF mechanics. Here we combine biophotonic and genetic approaches to address these open questions. First, we suppress the NMMII isoforms MIIA and MIIB and apply femtosecond laser nanosurgery to ablate and investigate the viscoelastic retraction of individual SFs. SF retraction dynamics associated with MIIA and MIIB suppression qualitatively phenocopy our earlier measurements in the setting of Rho kinase (ROCK) and myosin light chain kinase (MLCK) inhibition, respectively. Furthermore, fluorescence imaging and photobleaching recovery reveal that MIIA and MIIB are enriched in and more stably localize to ROCK- and MLCK-controlled central and peripheral SFs, respectively. Additional domain-mapping studies surprisingly reveal that deletion of the head domain speeds SF retraction, which we ascribe to reduced drag from actomyosin crosslinking and frictional losses. We propose a model in which ROCK/MIIA and MLCK/MIIB functionally regulate common pools of SFs, with MIIA crosslinking and motor functions jointly contributing to SF retraction dynamics and cellular traction forces.

Our recent studies have demonstrated that mechanical fractionation of tissue structure with sharply demarcated boundaries can be achieved using short (<20 μs), high intensity ultrasound pulses delivered at low duty cycles. We have called this technique histotripsy. Histotripsy has potential clinical applications where noninvasive tissue fractionation and/or tissue removal are desired. The primary mechanism of histotripsy is thought to be acoustic cavitation, which is supported by a temporally changing acoustic backscatter observed during the histotripsy process. In this paper, a fast-gated digital camera was used to image the hypothesized cavitating bubble cloud generated by histotripsy pulses. The bubble cloud was produced at a tissue-water interface and inside an optically transparent gelatin phantom which mimics bulk tissue. The imaging shows the following: 1) Initiation of a temporally changing acoustic backscatter was due to the formation of a bubble cloud; 2) The pressure threshold to generate a bubble cloud was lower at a tissue-fluid interface than inside bulk tissue; and 3) at higher pulse pressure, the bubble cloud lasted longer and grew larger. The results add further support to the hypothesis that the histotripsy process is due to a cavitating bubble cloud and may provide insight into the sharp boundaries of histotripsy lesions.

Abstract. Accurate, unambiguous detection of molecular interactions in living cells via measurements of Förster ͑or fluorescence͒ resonance energy transfer ͑FRET͒ events is experimentally challenging. We develop and apply a physiological fluorescence lifetime imaging microscopy ͑physiological FLIM͒ system to significantly improve FRET detection in living cells. Multiple positive and negative cellular controls are implemented to validate the experimental method developed. FLIM measurement techniques were found to remove fluorescence intensity-based artifacts, resulting in a seven-fold improvement in fluorescence measurement precision. The addition of cellular environmental controls, including both temperature and CO 2 stabilization, for physiological FLIM eliminates nonspecific FRET in the live-cell system studied. Overall, only physiological FLIM results in statistically significant results that clearly indicated the presence of specific molecular interactions in the live-cell system. This approach can be applied generally to improve the accuracy and precision of FRET measurements in living cells. Förster ͑or fluorescence͒ resonance energy transfer ͑FRET͒ can be used as an in vivo nanoscale ruler to detect the distance between fluorophores or interacting proteins with attached fluorophores.1 In intensity-based FRET detection, sophisticated calibration and correction procedures are usually required to cope with intensity variations, spectral bleedthrough, 2 back-bleedthrough, 3 and photobleaching. 4 In addition, nonspecific FRET, arising from random collisions of donor and acceptor and/or other unexpected associations of donor and acceptor, affects FRET accuracy, and although it can be detected, it may not be easily removed. 5Fluorescence lifetime imaging microscopy ͑FLIM͒ retrieves fluorophore lifetime data from fluorescence emission, and can be used to detect FRET with some advantages over intensity-based FRET.6 Interest in FLIM-FRET is growing, especially as commercial FLIM modules become available for confocal and multiphoton microscopy. Synaptic stimulation leads to expansion of the dendritic spine caused by rearrangement of the actin cytoskeleton triggered by phosphorylation of the calcium calmodulin dependent kinase (CaMKII) and by myosin II activity. To study CaMKII-actomyosin interactions we have visualized single GFP-tagged CaM-KII holoenzymes bound to tagRFP-tagged cortical actin in human endothelial cells by total internal reflection fluorescence microscopy. CaMKII phosphorylation mutants were used to examine whether release of CaMKII sequestered actin and myosin II induced F-actin fragmentation can explain the timing and extent of spine expansion. Thus far, we have found (i) Single GFP-CaMKII holoenzymes were resolved; in contrast to the less intense and rapidly mobile single GFP molecules. (ii) The holoenzymes have unimodal intensity distributions with < 30% standard deviation. The constitutively-active CaMKII T286D mutant, the sole exception, gave bimodal distributions with the higher mode six times as intense as the lower one. Subunit stoichiometries were determined by comparison against immobilized GFP single molecule intensities. (iii) CaMKII dwell-time distributions on stress fibers and filopodial structures were characterized. Dissociation rates were also measured by evanescent irradiation of photo-activatable GFP-CaMKII constructs bound to cortical actin (iv) The green / red fluorescence ratio of the actin structures relative to background gave an equilibrium measure of CaMKII binding. Affinity differences for different CaMKII isoforms and different phosphorylation states were estimated from the fluorescence ratios. In addition, single particle tracking was used to measure transient binding to the actin cytoskeleton. (v) We are currently measuring spatial co-operativity in the F-actin CaMKII binding sites and extending the methodology to analyse CaMKII diffusion along dendrites and spines in hippocampal neuron cultures. Our single molecule imaging data reveal spatial and macromolecular heterogeneities that may underlie aspects of the signal transduction response and cytoskeletal dynamics that are important for synaptic plasticity.
